Daily urine samples were obtained from 38 women of proven fertility, who were attempting to conceive, to assess the frequency of clinically unsuspected ‘biochemical pregnancies’ on the basis of late luteal phase rises of hCG. In 25 clinically confirmed conception cycles, the earliest pregnancy-induced hCG rises from baseline occurred 8 days after the endocrinologically defined date of ovulation (mean 11·1 days, SD 1·3). Urine samples obtained throughout 50 non-conception ovulatory cycles provided no evidence of any ‘biochemical pregnancies’ on the basis of late luteal phase rises of hCG occurring 8 or more days after the endocrinologically defined date of ovulation. If, as in previous studies, the date of ovulation had been estimated on the basis of menstrual cycle length and only the relatively non-specific NIH antiserum had been used in RIAs, 7 of the 50 (14%) cycles would have been classified as resulting in biochemical pregnancies on the basis of hCG 〉50 m-i.u./ml on one occasion or 〉20 m-i.u./ml on two or more occasions after day 20 of the cycle. These levels of ‘hCG’ occurred at or soon after ovulation and could be explained by LH cross-reacting in the RIA. The rises of ‘hCG’ were not confirmed when the urine samples were retested with an antiserum (Wellcome 895) of very high specificity for hCG. These results raise the possibility that the frequency of clinically unsuspected ‘biochemical pregnancy’ may have been substantially overestimated.

    Notizen: SYNOPSIS. Theophylline inhibited the phagocytosis of latex beads by Acanthamoeba castellanii (Neff). The cells recovered the ability to engulf beads after 1–2 hr of exposure to theophylline. Cells which have been exposed to 25 mM theophylline for a period of inhibition and recovery were not inhibited further by incubation with a fresh medium containing the same concentration of theophylline. However, the medium in which the cells recovered was as effective as a fresh medium in inhibiting phagocytosis in a fresh batch of cells, suggesting that the development of insensitivity to theophylline inhibition resides with the cells themselves. Dibutyryl cyclic AMP also inhibited bead uptake.

    Notizen: Abstract— With sucrose density gradient analysis, bovine retinal cytosol demonstrates 2S and 7S retinol-binding species; binding in pigment epithelial cytosol is predominantly to a 2S species. Binding of retinol to the 2S component in retina is unaffected by retinoic acid or retinyl palmitate whereas the ester effectively competes for 2S binding in pigment epithelium. Specific retinol binding can also be demonstrated by gel filtration on Sepharose 4B; no high molecular weight (〉 100–200,000) retinol binding species are observed by this technique. Both the 2S and 7S binding species in retinal cytosol are protein in nature and differentially susceptible to proteolysis. The 2S and 7S species appear to be separate and distinct since chaotropic agents such as sodium thiocyanate or KCl and CaCl2 do not seem to convert the 7S species into 2S subunits. Scatchard plot analysis indicates high affinity retinol binding to the 2S receptor. Computer analysis of the binding data yields Ka=3 × 108, n = 1, a molecular size of 16,200 and ΔG0=−9.5 kcal/mol.

    Notizen: The interaction between immune system and growing tumor can be expressed differently at different stages of the disease. This presentation covers three facets of these reactions in melanoma patients. A. The Primary Tumor. Time-lapse cinematography, with an analysis of lymphocyte movement demonstrated positive and negative chemotaxis against tumor tissues which correlated with their histological presence or absence within the primary tumor. B. The Regional Lymph Nodes. Histological examination of regional lymph nodes showed an increase in germinal center activity and B cell number, with a decrease in sinus histiocytosis and monocyte count as the tumor progressively invaded the node. This correlated with the elution studies, wherein the anti-membrane antibody decreased and the anti-cytoplasmic antibody increased during the same period of progression. C. Humoral Immunity and Metastasis. Clinical metastasis heralded the decrease of anti-membrane antibodies with a rise in anti-immunoglobulins, especially anti-idotypic antibodies and immune complexes containing tumor-directed antibody and either antigen or anti-immunoglobulin. This triad of anti-immunoglobulin, immune complexes and anergy as seen in other diseases with persistent antigenic stimulation, results in abnormal regulation and derangement.

    Notizen: SUMMARY— When a strain of brewer's yeast grown under suitable conditions was suspended in a solution of fermentable sugar, nucleotides and other U.V.–absorbing materials were rapidly released from the cells. The extent of release was dependent on the pH of the medium, the temperature, the concentration of fermentable sugar and on the presence of membrane–protecting (Ca++ or Mg++) or membrane–damaging (butanol, detergent) reagents. The released material was of low molecular weight and appeared to originate in a free intracellular pool. It was concluded that the mechanism of release of nucleotidic material was the result of a change in permeability of the cytoplasmic membrane of yeast contingent upon the transport and metabolism of fermentable sugar. Leakage of nucleotidic material from yeast was considered to be a normal physiological process of consequence in the brewing and wine–making industries.

    Notizen: To summarize the train of events in cholera as we visualize them: Severe dehydration results in hypovolemia, and cardiac output and all pressures fall. Acidosis causes peripheral venous constriction, thereby reducing the capacity of the venous reservoirs and maintaining, to some extent, venous return to the heart. Fluid administration without correction of acidosis favors a disproportionate venous return to the heart and pulmonary circulation. Alleviation of acidosis during fluid replacement results in a more even distribution of the circulating blood volume and reduces the possibility of engorgement of the pulmonary bed.This mechanism may also explain the occasional appearance of pulmonary congestion in patients with severe burns and metabolic acidosis. Fluid therapy without correction of acidosis in such patients may also favor a disproportionate venous return to the heart and pulmonary circulation. Treatment of the acidosis may be expected to reduce, in these patients as well, the possibility of pulmonary engorgement.In conclusion, these data indicate that the blood pH affects the distribution of the circulating blood volume and thereby influences the volume of blood returning to the heart, and through it, the central blood volume, ventricular filling pressures and cardiac output. We suggest that the site of action is in the peripheral venous bed.

An average of 11 naevi was present in each adult.The association between the distribution of naevi and that of malignant melanoma is discussed and evidence is presented that pigmented skin is able to prevent the malignant transformation of naevi and the spread of already established malignant melanoma.
