    Notes: SYNOPSIS. Entamoeba histolytica grown with H3-thymidine in CLG medium took up tritium into DNase-sensitive material in the nucleus and cytoplasm. The distribution of nuclear activity indicated that the entire nucleus, including the peripheral chromatin, may possess DNA; previous investigators reported DNA only in the endosome.The penicillin-inhibited bacterial associate (Bacteroides sp.) used in the CLG medium incorporated tritium from H3-thymidine into autoradiographically detectable DNase-sensitive material. Autoradiographs of amebae fed bacteria prelabeled with H3-thymidine also revealed some nuclear and cytoplasmic label. Thus, the amount of cytoplasmic label due to ingested, prelabeled bacterial DNA and/or actual biosynthesis of cytoplasmic DNA by the amebae themselves, is not known. Also, at least some of the nuclear DNA of amebae is synthesized from ingested bacteria, or, more likely, from bacterial degradation products.

    Notes: Estrogen is known to play an important role in regulating reproductive function in female rats through actions exerted at the preoptic area, a part of the brain that is markedly sexually dimorphic and which contains abundant estrogen receptors. A critical question to our understanding of estrogen's action on the brain is to identify the types of neurons that contain estrogen receptors (ER). Previous studies have shown that atrial natriuretic peptide (ANP) is in abundance in the preoptic area, and that ANP and other natriuretic peptides are capable of regulating gonadotropin secretion. In an effort to determine whether ERs are present in natriuretic peptide-immunoreactive (NP-ir) neurons in the preoptic area of the rat, double label immunocytochemistry was performed. Since ER-ir, as demonstrated with antibody H222 is known to be localized predominantly in cell nuclei, while NP-ir is present in the cytoplasm, single cells can be double labeled. Diaminobenzidine tetrahydrochloride was used for localization of NP-ir neurons, while nickel-enhanced diaminobenzidine tetrahydrochloride was used for localization of ER-ir. The results revealed that many nuclei throughout the preoptic area contained neurons that were ER-ir or NP-ir and that a substantial number were double labeled. Cell counts in selected preoptic nuclei and components, including the anteroventral periventricular nucleus, periventricular preoptic nucleus, medial part of the medial preoptic nucleus, and central part of the medial preoptic nucleus revealed that 13.6%, 11.1%, 13.5%, and 24.4%, respectively, of the NP-ir neurons in these nuclei also contained ER-ir. Collectively, a total of 14.9% of the NP-ir neurons in these nuclei also contained ER-ir. These results support the hypothesis that estrogen may regulate the activity of NP-ir cells in the preoptic area and thereby may be capable of altering functions associated with NPs in this region of the brain, including not only regulation of gonadotropin secretion, but also control of blood pressure and fluid balance.

    Notes: Gonadotropin secretion from the pituitary is regulated in large part by steroid action on the brain. An important question concerns whether luteinizing hormone-releasing hormone (LHRH) neurons themselves transduce steroid signals, or whether, alternatively, steroid-sensitive interneuronal populations regulate their activity. A previous study in the rat employing steroid autoradiography combined with LHRH immunocytochemistry revealed that only an exceedingly small percentage of LHRH-immunoreactive (ir) neurons was estrogen concentrating. This study has examined the relationship of estrogen receptive and LHRH-ir cells in the male and female guinea-pig brain with double label immunocytochemistry. Since estrogen receptor-ir, as demonstrated with antibody H222, is known to be confined predominantly to the cell nucleus, whereas LHRH-ir is localized mainly in the cytoplasm, single cells can be double-labeled. Diaminobenzidine tetrahydrochloride was used for localization of LHRH-ir while nickel-enhanced diaminobenzidine tetrahydrochloride was used for localization of estrogen receptor-ir. The results revealed that there were many brain nuclei that contained both LHRH and estrogen receptor-positive cells, including the preventricular periventricular nucleus, the anterior subcompact nucleus of the medial preoptic nucleus (MPNa), the remainder of the medial preoptic nucleus, the retrochiasmatic area, and the anterior, dorsomedial, ventrolateral and arcuate nuclei. However, of a total of 2,604 LHRH-ir cells that were examined, we observed only 5 double-labeled cells (〈0.2%). The double-labeled cells were not restricted to a single nucleus; they were present in the MPNa, the retrochiasmatic area and the arcuate nucleus. Moreover, at the light microscopic level, LHRH cells quite frequently appeared to be apposed to estrogen receptor-positive cells (8.8% in the female), especially in the MPNa. These results lend further support to the notion that estrogen-mediated regulation of the LHRH system is achieved primarily through estrogen receptive interneurons. However, due to the existence of LHRH-LHRH synaptic interactions, the possibility also exists that a small population of estrogen-sensitive LHRH neurons could contribute to generalized activation of the LHRH system.

    Notes: The effect of Mueller–Hinton (MH), MH+blood or brain heart infusion medium (agar or broth) on 13 Enterococcus isolates was determined, when testing their antibiotic susceptibility. Disk diffusion and Vitek methods were used to determine vancomycin resistance, while broth dilution and E-test methods were used to measure the minimum inhibitory concentration. The data were correlated with the presence of vancomycin resistance genes. A definite correlation pattern could not be established between the presence of van genes and vancomycin resistance in any plating medium, when tested by the disk diffusion assay. The broth dilution, irrespective of the plating medium, and Vitek methods were more reliable than the E-test method in testing isolates with vanA or vanB genes. However, for vanC2/C3 genotypes, the E-test method, irrespective of the plating medium, tested better than the broth dilution assay.

    Notes: The reproductive biology of the bat Macrotus waterhousii was investigated anatomically and physiologically several times monthly throughout the year.The male spermatogenic cycle is initiated in June; sperm are available in August. Testicular hypertrophy is accompanied by increased levels of testicular ascorbic acid and circulating plasma testosterone. Sperm are present in the seminiferous tubules and later the epididymides from August to early December. Regression in testicular size begins in late September and testes are involuted by early December. Levels of testicular ascorbic acid and plasma testosterone decline at this time, although detectable amounts of both are present year-round. Mature sperm are absent from the male tract after late November or early December.The accessory sex gland complex and Cowper's glands undergo an annual cycle in synchrony with the testicular cycle. The accessory complex contains fructose, the concentration of which varies seasonally in direct proportion to glandular hypertrophy. Only trace levels of fructose are elaborated by Cowper's glands.In contrast to vespertilionid bats, where the accessory glands remain enlarged and active when testes involute, those in Macrotus regress.The penis differs structurally from that of other temperate North American bats, primarily by lacking a bony baculum and specialized erectile accessory corpus cavernosus bodies.Males are not reproductively active in their first autumn.

    Notes: We have measured the specific viscosity of closed circular PM2 DNA in the presence of concentrations of ethidium bromide ranging up to 5 mg/ml. Both native viral PM2 DNA I and enzymatically prepared relaxed, closed circular PM2 DNA I0 exhibit a complex dependence of the specific viscosity upon the extent of supercoiling. As the number of superhelical turns is increased in the positive sense from zero, the viscosity first decreases to a minimum, then passes through a secondary maximum, and eventually again increases as the dye-induced duplex unwinding proceeds. In the case of DNA I, a corresponding behavior is mirrored in the negative sense as dye is removed from the principal viscometric maximum (complete relaxation of the DNA by dye). The shape of the curve relating specific viscosity to extent to supercoiling is similar for superhelical DNAs of either handedness, a result which we interpret to mean that the influence of any regions of special secondary structure (such as denatured loops) upon the viscosity is minimal. At very high dye concentrations the specific viscosity decreases dramatically. This effect might arise either from intermolecular aggregation or from a dye-induced collapse in the DNA secondary structure.

    Notes: To understand how comparatively simple macromolecular components become biological systems, studies are made of the morphogenesis of bacteriophages. Pulsed field agarose gel electrophoresis (PFGE) has contributed to these studies by: (i) improving the length resolution of both mature, linear, double-stranded bacteriophage DNAs and the concatemers formed both in vivo and in vitro by the end-to-end joining of these mature bacteriophage DNAs, (ii) improving the resolution of circular conformers of bacteriophage DNAs, (iii) improving the resolution of linear single-stranded bacteriophage DNAs, (iv) providing a comparatively simple technique for analyzing protein-DNA complexes, and (v) providing a solid-phase quantitative assay for all forms of bacteriophage DNA; solid-phase assays are both less complex and more efficient than liquid-phase assays such as rate zonal centrifugation. Conversely, studies of bacteriophages have contributed to PFGE the DNA standards used for determining the length of nonbacteriophage DNAs. Among the solid-phase assays based on PFGE is an assay for excluded volume effects.

    Description: Phosphorus (P) availability to crops in organic systems can be a major issue, with the use of readily available forms often restricted. One product that can be used in organically managed systems, that is also relatively easily accessible to growers, is phosphate rock, although its solubility and therefore crop availability is often poor. One possible approach to improve this situation is co-composting phosphate rock with selected organic waste materials. Various ratios of phosphate rock and cabbage ( Brassica oleracea L.) residues were co-composted and the products tested at different rates of application. The effects were assessed over 12 weeks using oilseed rape ( Brassica napus L.) and perennial ryegrass ( Lolium perenne L.) as bioassay crops in a pot experiment. At harvest, estimates of P derived from cabbage and phosphate rock for the lowest of two rates of compost were ≈ 2 and 10 mg P pot –1 for oilseed rape, compared to 5 and 2 mg P pot –1 for perennial ryegrass, respectively. Roots tended to have higher P concentrations than shoots. The crops showed differences in their abilities to access various P sources, with oilseed rape effectively taking P from phosphate rock, whereas perennial ryegrass was more effective at accessing cabbage-derived P (the main substrate in the compost). Oilseed rape was able to take up 20% of the total P applied as phosphate rock, whereas perennial ryegrass took up less than 5% of the total P applied from this material. Both pre- and post-application solubilisation/transformation mechanisms were involved in supplying plant-available P. Quantifying the relative contribution from individual P sources remains problematic even within this relatively simple system.
