    Notes: Morphology of the bovine mammary gland during the nonlactating periodLightmicroscopical and histochemical investigations were performed on biopsies, which were obtained from the mammary glands of healthy pregnant cows during different stages of the nonlactating period. The results demonstrate characteristic morphological changes in the different phases of the nonlactating period. They can be interpretated as involution and regeneration processes, which occur in the alveoli and interalveolar tissue of the bovine mammary gland during the nonlactating period.

    Notes: Morphology of the bovine mammary gland during the nonlacting periodAccording to the electron microscopic investigations the most evident sign of the phase of the active involution of the mammary gland in cattle is the occurrence of different great vacuoles with a diffuse content in the cytoplasma of the secretory cells till the 30th day after the beginning of the dry period. Secretory cells which go over in the rest phase show a great reduction of the cell organelles and many filaments and secundary lysosomes or residual bodies. One can observe many macrophages and lymphocytes in the partly enlarged intercellular rooms between the secretory cells and the myoepithelial cells. The myoepithelial cells are adapting to the changing forms of alveoles. The phase of the colostrogenesis is characterised by the enlargement of the cell organelles and the reappearence of vacuoles.

    Notes: Hypothalamo-neurohypophysial magnocellular neurons display specific electrical activities in relation to the mode of release of their hormonal content (vasopressin or oxytocin). These activities are under strong glutamatergic excitatory control. The implication of NMDA receptors in the control of vasopressinergic and oxytocinergic neurons is still a matter of debate. We here report the first detailed characterization of functional properties of NMDA receptors in voltage-clamped magnocellular neurons acutely dissociated from the supraoptic nucleus. All cells responded to NMDA with currents that reversed polarity around 0 mV and were inhibited by D-2-amino-5-phosphonovalerate (d-APV) and by 100 μM extracellular Mg2+ (at -80 mV). Sensitivity to the co-agonist glycine (EC50, 2 μM) was low compared with most other neuronal preparations. The receptors displayed low sensitivity to ifenprodil, were insensitive to glycine-independent potentiation by spermine, and had a unitary conductance of 50 pS. No evidence was found for two distinct cell populations, suggesting that oxytocinergic and vasopressinergic neurons express similar NMDA receptors. Characterization of NMDA receptors at different postnatal ages revealed a transient increase in density of NMDA currents during the second postnatal week. This was accompanied by a specific decrease in sensitivity to d-APV, with no change in NMDA sensitivity or any other properties studied. Supraoptic NMDA receptors thus present characteristics that strikingly resemble those of reconstituted receptors composed of NR1 and NR2A subunits. Understanding the functional significance of the development of NMDA receptors in the supraoptic nucleus will require further knowledge about the maturation of neuronal excitability, synaptic connections and neurohormone release mechanisms.

    Notes: We computed by simulation the null distributions and power function of 10 statistics for comparing the mean directions of two Fisher distributions for various sample sizes and concentrations. 10000 simulations were used for each null distribution and for each degree of angular separation out to 40°. The statistics have been suggested by, or are variations of suggestions of, McFadden & Lowes (1981) (which we show are not conditional as they insisted) and Watson (1956, 1983).

    Notes: Abstract Cytokines are produced by a variety of cells and have numerous of overlapping activities. There is increasing evidence that cytokines play a crucial role in the pathogenesis of psoriasis and of other dermatologic diseases. This review summarizes current knowledge as to how the altered cytokine network is involved in the accumulation of inflammatory cells in lesional skin, and how the cytokines are involved in epidermal hyper-proliferation. The actions of the most important therapeutic compounds, such as corticosteroids, dithranol, cyclosporine, retinoids, vitamin D3 analogues and ultraviolet radiation, on the cytokine system are also discussed. Consideration is given as to how the effects on the production of cytokines and/or cytokine receptors contribute to their therapeutic action.

    Notes: The activity of various antibacterial agents (amoxicillin, josamycin, doxycycline and metronidazole) was established in vitro using a rapid micromethod. The activity of these agents, which are widely used in oral medicine, was evaluated against microorganisms responsible for periodontitis and bucco-dental infections. Their action against α-hemolytic streptococci (including pneumococci) which make up the majority of the indigenous oral flora was also tested. Amoxicillin was found to be effective against all the strains tested. Doxycycline was active against periodontal bacteria, but not against 50% of the streptococcal flora. Josamycin was found to be effective against streptococci, but appeared without effect on Eikenella corrodens and Actinobacillus actinomycetemcomitans. Metronidazole, inactive against streptococci, displayed greater activity towards the strict anaerobes. The use of these antibiotics for the treatment of bucco-dental infections, especially periodontitis, is discussed. For periodontitis and periodontal suppurations, antimicrobial agents present a valuable adjunct to local treatments such as scaling or rootplaning. This may prevent more serious infections such as endocarditis that can develop after tooth extraction.

    Notes: Age-related changes in motor endplates were studied in the fibularis longus, semitendinosus and longissimus muscles in pigs. They develop from small, compact AChE-active figures in fetuses to typical myoneuronal synapses with a branched subsynaptical structure of AChE-active units. High correlations were found between size of endplates and both myofibre diameter and body mass. Growth analyses with respect to age, as well as allometrical approaches, showed that endplate growth slightly precedes that of myofibres, with both having similar growth patterns. The morphological changes, however, imply that the increment of synaptical area could lag behind that of myofibres.

    Notes: Antigen 60 (A60). the main thermostable immunogen of tuberculin and PPD. has been purified from Mycobacterium bovis BCG cytoplasm, and identified by crossed Immunoelectrophoresis with anti-BCG polyclonal antiserum. Two A60 fractions, free lipids and lipid-conjugated compounds, have been recognized. The free lipids represented about 30% (dry weight), and consisted essentially of C16-C18, fatly acids, and of phosphatidyl-inositol-mannosides. Lipocon-jugates, upon DEAE-cellulose chromatography and gel filtration, yielded two main fractions of neutral and polar components. Chromatography of delipidated and deproieinizd A60 on Sephadex G-100yielded: a high molecular weight fraction (Al, 18%, a lipoglucan of ± 106) and a low molecular weight fraction (B, 10%, a lipopeptidoglycan of ± 104) containing mannose, glucose, and small amount of arabinose. The polysaccharide moieties of fractions Al and B were submitted to aeetylalion, methylation, and acid hydrolysis, and ihe structure of the hydrolysed polymer was deduced by combined gas chromalography/mass spectrometry analysis. The results indicated a branched structure involving 1. 4-, 1, 6-, and 1, 4, 6-linked D-gluco- or n-manno-pyranosyl residues. Glucan- and peptidoglycan-bound fatty acids were identified as saturated (C16-C18) and monounsaturated linear acids (C12, C18). Immunodiffusion on agarose gel indicated that delipidalion and proteolysis did not suppress the ability of A60 to yield immunoprrecipitates with anti -A60 antiserum. The high polymer fractions obtained by chromatography on DEAF cellulose and Sephadex G-100 were also reactive. It is concluded that A60 is made of tree lipids and of hpopcptidoglycans of high molecular weights (106-107) endowed with immunogenic properties.
