    Description: Photosynthetic production of organic matter by microscopic oceanic phytoplankton fuels ocean ecosystems and contributes roughly half of the Earth's net primary production. For 13 years, the Sea-viewing Wide Field-of-view Sensor (SeaWiFS) mission provided the first consistent, synoptic observations of global ocean ecosystems. Changes in the surface chlorophyll concentration, the primary biological property retrieved from SeaWiFS, have traditionally been used as a metric for phytoplankton abundance and its distribution largely reflects patterns in vertical nutrient transport. On regional to global scales, chlorophyll concentrations covary with sea surface temperature (SST) because SST changes reflect light and nutrient conditions. However, the ocean may be too complex to be well characterized using a single index such as the chlorophyll concentration. A semi-analytical bio-optical algorithm is used to help interpret regional to global SeaWiFS chlorophyll observations from using three independent, well-validated ocean color data products; the chlorophyll a concentration, absorption by CDM and particulate backscattering. First, we show that observed long-term, global-scale trends in standard chlorophyll retrievals are likely compromised by coincident changes in CDM. Second, we partition the chlorophyll signal into a component due to phytoplankton biomass changes and a component caused by physiological adjustments in intracellular chlorophyll concentrations to changes in mixed layer light levels. We show that biomass changes dominate chlorophyll signals for the high latitude seas and where persistent vertical upwelling is known to occur, while physiological processes dominate chlorophyll variability over much of the tropical and subtropical oceans. The SeaWiFS data set demonstrates complexity in the interpretation of changes in regional to global phytoplankton distributions and illustrates limitations for the assessment of phytoplankton dynamics using chlorophyll retrievals alone.

    Notes: The aim of this randomised controlled study was to determine whether an esmolol infusion affected the incidence of ST segment changes during weaning from intermittent positive pressure ventilation and tracheal extubation after coronary artery surgery. Thirty-one patients received an infusion of esmolol 0–300 µg.kg−1.min−1 and 37 patients comprised the control group. ST segment changes were monitored using a continuous ambulatory surveillance system. The electrocardiogram, direct arterial pressure and pulse oximetry were monitored continuously. The period of analysis was from 120 min before until 180 min after tracheal extubation. Three patients in the esmolol group developed myocardial ischaemia during the study period compared with 12 in the control group (p = 0.05). Heart rate increased with time during the study period (p = 0.002) in the control group but was unchanged in the esmolol group. Mean heart rate was significantly higher in the control group than in the esmolol group from 40 min before until 180 min after tracheal extubation. Seven patients in the esmolol group suffered adverse events related to the esmolol infusion. Although the use of esmolol reduced the incidence of myocardial ischaemia, the incidence of adverse effects makes it unsuitable prophylaxis for patients after coronary artery surgery.

    Notes: The occurrence of nitrogen isotope discrimination with absorption and assimilation of nitrate (NO3–) and ammonium (NH4+) was investigated using two genotypes of barley, Hordeum vulgare L. cv. Steptoe and Az12 : Az70, the latter of which lacks the characterized nitrate reductase isozymes. Plants were grown under two situations: a closed system with limited nitrogen or an open system with unlimited nitrogen, to elucidate the conditions and processes that influence discrimination. There was no discrimination observed for Az12 : Az70 when supplied with limited nitrogen. Discrimination was observed for Steptoe seedlings at high external NO3– concentrations, but not with low NO3– when assimilation is probably rapid and complete. The same pattern was observed for Steptoe when NH4+ was supplied; indicating that for both nitrogen forms discrimination is dependent upon the presence of the assimilatory enzyme and the external concentration. The implications of this study are that both internal (assimilatory enzyme distribution) and external (source concentration) factors may have a larger impact on tissue δ 15N than the form of nitrogen utilized. This suggests that tissue δ 15N may not always be a reliable indicator of a plant's integrated nitrogen nutrition.

    Notes: Immunocytochemical and morphometric techniques were used to quantify the distribution of cyclooxygenase (cox)-containing neurons in rat L5 dorsal root ganglia (DRG). Cox-1 immunolabelling was almost exclusively restricted to small diameter DRG neurons (〈 1000 μm2), and was extensively colocalized with calcitonin gene-related peptide (CGRP) and isolectin B4 (IB4). Cox-1 was present in 65% and 70% of CGRP- and IB4-labelled neurons, respectively. Cox-1 labelling was also found in neurons expressing the sensory neuron-specific (SNS) Na+ channel. Cox-2 labelling was absent in DRG from normal rats. In the Freund's adjuvant model of monoarthritis, the proportion of cox-1-positive DRG neurons was unchanged and no neurons were found to be labelled for cox-2. In primary tissue culture, cox-1 immunolabelling persisted in vitro for up to 9 days and was present in morphologically identical neurons. The selective expression of cox-1 in peripheral ganglia was confirmed by the small number of nodose ganglion neurons and superior cervical ganglion (SCG) neurons labelled for cox-1. These data suggest that cox-1 is a marker for a subpopulation of putative nociceptive neurons in vitro and in vivo, and suggests that the prostaglandins synthesized by these neurons may be important for nociceptor function. These data may have important implications for the mode and mechanism of action of non-steroidal anti-inflammatory drugs (NSAIDs).

    Notes: P2X receptors for adenosine 5′-triphosphate (ATP) comprise a family of ligand-gated cation channels with distinct characteristics which are dependent on the receptor subunits (P2X1–7) expressed, and the homomeric or heteromeric assembly of protein subunits in individual cells. We describe the properties of P2X receptors expressed by cultured adult rat dorsal root ganglion cells on the basis of the time course of responses to ATP, α,β-methylene adenosine 5′-triphosphate (α,β-meATP) and 2-methyl-thioadenosine 5′-triphosphate (2-meSATP), and using the antagonists 2′,3′-O-(2,4,6-trinitrophenyl) ATP (TNP-ATP), a novel and highly selective purinoceptor antagonist, suramin and iso-pyridocalphosphate-6-azophenyl-2′,5′ disulphonic acid (PPADS). ATP (10 μm) evoked inward currents in ≈ 95% of neurons tested and 〉 80% responded with a fast transient inward current that rapidly inactivated during the continued presence of ATP. Of the remaining neurons, ≈ 4% showed a sustained response and ≈ 10% showed a combination of transient and sustained components. Rapid application of ATP, α,β-meATP and 2meSATP demonstrated these to be full agonists of the rapidly inactivating P2X receptor (pA50 values = 5.83, 5.86 and 5.55, respectively), whilst uridine triphosphate (UTP) and 1-β,γ-methyleneadenosine 5′-triphosphate (1-β,γ-meATP) were ineffective as agonists. These rapidly inactivating responses could be inhibited by TNP-ATP, suramin and PPADS (pIC50 = 9.5, 6.5, 6.4, respectively). Using inactivation protocols, we demonstrate the presence of homomeric P2X3-like receptors and non-inactivating P2X receptors, which indicates that individual subsets of adult dorsal root ganglion neurons have distinct P2X receptor phenotypes, and that individual DRG neurons may express multiple P2X receptor subtypes.

    Notes: The effects of elevated carbon dioxide (CO2) on plant litter are critical determinants of ecosystem feedback to changing atmospheric CO2 concentrations. We measured concentrations of nitrogen (N) and carbon (C) and calculated C : N ratios of green leaves of two desert perennial shrubs, and the same quality parameters plus lignin and cellulose content of leaf litter from four shrub species exposed to elevated CO2 (FACE technology; Hendrey & Kimball, 1994) for 3 years in an intact Mojave Desert ecosystem. Shrubs tested were Larrea tridentata, Lycium pallidum, Lycium andersonii and Ambrosia dumosa. We calculated resorption efficiency from green tissue and leaf litter N data and measured lignin and cellulose content in litter in the last year study. Green leaves of L. tridentata grown under elevated CO2 had significantly lower N concentrations and higher C : N ratios than shrubs grown in ambient conditions in 1999 (P 〈 0.05). Lycium pallidum green leaves grown under elevated CO2 had significantly lower N concentrations and higher C : N ratios than shrubs grown under ambient conditions in 2000 (P 〈 0.05). There was no CO2 effect on C content of either species. We found no effect of CO2 on N or C content, C : N ratios, or lignin or cellulose concentrations in leaf litter of L. tridentata, L. pallidum, L. andersonii, or A. dumosa. There was no significant effect of CO2 on estimates of shrub resorption efficiency. There was a seasonal effect on green tissue and litter tissue quality for L. tridentata, with lower tissue N content in summer than in spring or winter months. These data suggest that any productivity increases with elevated CO2 in desert ecosystems may not be limited by lower leaf litter quality and that resorption efficiency calculations are best performed on an individual leaf basis.
