    Notizen: Perinuclear type of antineutrophil cytoplasmic antibodies (p-ANCA) have been found in patients with periarteritis nodosa. Churg-Strauss arteritis. or pauci-immune idiopathic crescentic glomerulonephritis. Recently, the association of p-ANCA and normotensive renal failure, in patients with systemic sclerosis (SSc), was reported. We have studied the incidence of p-ANCA in patients with SSc and have investigated its relationship to clinical and laboratory findings. Sera from 77 patients with SSc were examined by the indirect immunofluorescence (IIF) test, employing an ethanol-fixed human neutrophil and enzyme-linked immunosorbent assay, using purified myeloperoxidase (MPO) as an antigen (MPO-ELISA). The sera from seven patients (9-1%) were p-ANCA positive, by both IIF and MPO-ELISA. One patient died from systemic necrotizing angiitis but the remaining six patients have shown no symptoms of systemic vasculitis or of renal involvement. There was a tendency for patients positive for p-ANCA (anti-MPO antibody) to also be positive for other autoantibodies. such as anti-Sc1-70 antibody, anti-centromere antibody, anti-microsome antibody, anti-thyroglobulin antibody and rheumatoid factor. Although the incidence of p-ANCA (anti-MPO antibody) is low in patients with SSc, and its clinical significance in SSc needs further investigations, this could be a serological marker for certain symptoms in SSc.

    Notizen: : Folate contents of a total of 210 dairy products, including milk, yogurt, ice cream, sherbet, frozen yogurt, pudding, sour cream and cheese, were measured by Lactobacillus casei microbiological assay after the extraction using trienzyme treatment. These values are compared with those using traditional folate conjugase treatment alone and those in previously published food tables. Folate contents by the method using trienzyme treatment were uniformly higher than those using the traditional method and the published contents. To our knowledge, the values published here are the largest collection of folate content of dairy products and can be used to accurately assess dietary folate intake in the general population.

    Notizen: We report a 57-year-old woman with systemic sclerosis (SSc) who died suddenly, following a haemoptysis. At post-mortem, systemic necrotizing angiitis of small vessels was observed in several organs. Necrotizing angiitis has been reported as a rare complication of systemic sclerosis, and is usually lethal.

    Notizen: Psoriatic hyperproliferative epidermis is characterized by a decreased β2-adrenergic adenylate cyclase response as well us by altered differentiation markers that include decreased loricrin and increased involucrin, Using a quantitative reverse transcription-polymerase chain reaction, we analysed the expression of β2-adrenergic receptor-mRNA. loricrin-mRNA, and involucrin-mRNA in the epidermis of five patients with psoriasis vulgaris. The mRNAs of the β2-adrenergic receptor and loricrin in the involved epidermis were significantly decreased, by 0.35-fold (P 〈 0.01) and 0.55-fold (P 〈 0.05) respectively, compared with uninvolved epidermis. In contrast, the involucrin mRNA expression of the involved epidermis was significantly increased, by 3.77-fold (P 〈 0.01). No significant difference in β-actin mRNA transcripts was detected between the involved and the uninvolved epidermis. These results indicate that the altered expression of the β2-adrenergic receptor, loricrin. and involucrin. in the psoriatic involved epidermis, is associated with different amounts of each mRNA transcripts.

    Essential role of the IRF8-KLF4 transcription factor cascade in murine monocyte differentiation (2013)

    Beschreibung: Monocytes regulate host defenses, inflammation, and tissue homeostasis. The transcription factor interferon regulatory factor-8 (IRF8) stimulates monocyte/macrophage differentiation, yet genome-wide understanding of the differentiation program initiated by IRF8 is lacking. By combining chromatin immunoprecipitation sequencing with gene expression profiling, we show that during IRF8-dependent monocyte differentiation, IRF8 binding occurs at both promoter-proximal and promotor-distal regions together with the transcription factor PU.1 and is associated with gene induction. Many of the promoter-distal IRF8 binding sites show an increase in histone H3 lysine 4 monomethylation, a signature for enhancers. However, about half the IRF8-induced genes were not bound by IRF8, suggesting the involvement of downstream transcription factors. Analysis of DNA motifs in cis -regulatory elements of these indirect IRF8 target genes predicted that Krüppel-like factor-4 (KLF4)—essential for Ly6C + monocyte development—is one such factor. Indeed, monocyte development in Irf8 –/– mice is as defective as that in Klf4 –/– chimeric mice. Moreover, Irf8 –/– monocyte-dendritic cell progenitors do not express Klf4 messenger RNA. Introduction of KLF4 into an Irf8 –/– myeloid progenitor cell line induced a subset of IRF8 target genes and caused partial monocyte differentiation. Taken together, our present results uncover genome-wide behavior of IRF8 and identify an IRF8-KLF4 axis that operates during monocyte differentiation.

    Transcription factor IRF8 plays a critical role in the development of murine basophils and mast cells (2015)

    Beschreibung: Basophils and mast cells play critical roles in host defense against pathogens and allergic disorders. However, the molecular mechanism by which these cells are generated is not completely understood. Here we demonstrate that interferon regulatory factor-8 (IRF8), a transcription factor essential for the development of several myeloid lineages, also regulates basophil and mast cell development. Irf8 –/– mice displayed a severe reduction in basophil counts, which was accounted for by the absence of pre-basophil and mast cell progenitors (pre-BMPs). Although Irf8 –/– mice retained peripheral tissue mast cells, remaining progenitors from Irf8 –/– mice including granulocyte progenitors (GPs) were unable to efficiently generate either basophils or mast cells, indicating that IRF8 also contributes to the development of mast cells. IRF8 appeared to function at the GP stage, because IRF8 was expressed in GPs, but not in basophils, mast cells, and basophil/mast cell-restricted progenitor cells. Furthermore, we demonstrate that GATA2, a transcription factor known to promote basophil and mast cell differentiation, acts downstream of IRF8. These results shed light on the pathways and mechanism underlying the development of basophils and mast cells.
