    Notizen: The Hartree-Fock method (standard Roothaan closed-shell HF-LCAO theory) and the Hartree-Fock-Slater method (restricted HFS-LCAO-DV method developed by Baerends and Ros) have been compared with emphasis on the respective one-electron equations and on the matrix elements of the respective Fock operators. Using the same STO basis in the two cases, the matrix elements of the Fock operators and of their separate one-electron, Coulomb, and exchange contributions have been calculated for the same orbitals and density of the ground state of the diatomic molecule ZnO. The effects of methodical (exchange potential) and numerical (DV method, density fit) differences between the HF and HFS methods on the various matrix elements have been analyzed. As expected the methodical effect prevails and is responsible for the higher (less negative) values of the matrix elements of the HFS Fock operator compared to those of the HF Fock operator. Numerical effects are observable also and are caused by the difference in integration procedures (DV method), not by the density fit.

    Notizen: The production of prourokinase (PUK) by a human kidney tumor cell line is studied in long term cultures. Cells are grown on microcarriers which are retained inside the reactor by sedimentation or with a spin filter. Two modes of operation are compared: feed harvest at an average medium exchange rate of 0.3 d-1 and continuous perfusion at a higher dilution rate of 1.5 d-1. In the two systems a stable production of PUK has been maintained for more than 400 h. Kinetics of cellular growth, nutrient consumption, and metabolite and PUK excretion are similar. After an initial rapid growth period, one observes a 10-fold reduction in all the cellular metabolic activities during the stationary phase. Continuous perfusion yields a higher cell density (7 × 106 cells·mL-1) than feed harvest (3 × 106 cells·mL-1), which results in a twofold increase in the reactor productivity. But higher final enzyme activities, about 250 ru·mL-1, are obtained in the feed harvest recovered medium than in the perfusion medium, 100-150 ru·mL-1. The cumulative medium consumption per mass of product is the same in the repeated batch and in the continuous operation mode.

    Notizen: A general methodology is proposed to estimate the actual specific growth and death rate of mammalian cells in continuous perfusion reactors from the monitoring of the release of the cytoplasmic enzyme lactate dehydrogenase (LDH) in the culture medium. The procedure is illustrated on a perfusion culture of human tumor kidney cells growing on microcarriers and producing prourokinase (PUK). The intracellular LDH content of living attached cells is checked to be constant during the culture. However, cells detached from the microcarriers, and counted dead because of the uptake of trypan blue, have only released part of their intracellular LDH. In the culture medium, LDH is relatively stable as the loss of activity does not exceed 5% per day. The time variation of the LDH concentration in the medium is used to calculate the total amount of lysed and actually produced cells in the reactors, hence, the actual specific rates of cell growth and death. It is thus found that the stationary phase observed after 400 h of perfusion culture is the result of equal growth and death rates, with a daily renewal of living cells on the microcarriers near 10%. Moreover, for the cell line tested, the production of PUK is associated with cellular growth.

    Notizen: Mutants of the luminescent bacterial strain NRRL B-11177 were isolated with pleiotropic hypersensitivity towards hydrophobic antimicrobial agents. SDS-PAGE analyses of outer membrane proteins and lipopolysaccharides revealed that the outer membrane structure of the ahs-mutants was altered. QSAR analysis showed that the inhibitory effect of chloro-substituted phenols on bioluminescence of the ahs-mutants depended on their hydrophobicity. The effect of chlorinated phenols and detergents on bioluminescence was increased in the ahs-mutants. The potential use of these mutants in bioluminescent toxicity tests was discussed.

    Notizen: Acetylcholine neurons in the striatum are believed to be primarily intrinsic. In cats a cylinder of caudate tissue was isolated unilaterally from all afferent and efferent connections while preserving the blood supply through the base of the cylinder. Cats were decapitated two and four weeks after operation. Dorsal, intermediate and ventral portions of the viable isolated cylinder, and corresponding tissue from the contralateral hemisphere serving as control, were removed, weighed and frozen. Deuterated internal standards at concentrations approximating those of acetylcholine and choline in the tissue were added and acetylcholine and choline extracted, reacted with propionyl chloride and demethylated with sodium benzenethiolate. The derivatives were separated by gas chromatography, and the acetylcholine and choline concentrations in the tissue evaluated using selected ion monitoring of m/z 58, 60 and 64. The observed decrease in the acetylcholine concentration in the dorsal portion could be due either to the interruption of the cholinergic thalamic input or to the more severe ischemia in this portion. The increase in the acetylcholine concentration in the ventral portion, where blood circulation is normal, may reflect accumulation of acetylcholine in inactive intrinsic neurons after severance of their input, or indicate sprouting of intrinsic acetylcholine neurons.

    Beschreibung: The mesoscale structure of a mature polar low was studied on the basis of high-resolution airborne measurements and numerical modelling. A polar low was measured by light detection and ranging (lidar) and dropsonde observations over the Norwegian Sea on 3 and 4 March 2008. Lidar observations provided cross-sections of water-vapour mixing ratio, backscatter ratio and horizontal wind speed around the polar low and through its centre. Mesoscale structures, such as shallow convection in a cold-air outbreak, a dry intrusion in the eye-like centre of the cyclone and deep convection surrounding it could be identified. Numerical simulations were performed with the European Centre for Medium-Range Weather Forecasts (ECMWF) Integrated Forecast System (IFS) and a high-resolution, polar version of the Weather Research and Forecasting (WRF) model. WRF simulations reproduced these structures and showed that the polar low had a warm, upper-level core with descending motions. The eye-like centre had a diameter of about 100–150 km and was characterized by rather stable stratification, horizontally constant potential temperatures and calm winds. Beyond the centre, wind speeds increased rapidly. The observed radial wind and temperature profiles support previous idealized simulations. Several WRF sensitivity tests showed the influence of the initialization time and sensible and latent heat fluxes from the surface on the simulated polar-low development. The polar-low simulations were more accurate in runs starting at the mature stage. Heat fluxes from the surface were important for the polar-low energetics, especially at the final stages.
