    Notes: Abstract. The rBAT protein, when expressed in Xenopus oocytes, was previously shown to reproduce the selectivity of the Na+-independent neutral and basic amino acid transport system called bo,+. More recently, the capacity of rBAT to generate a transmembrane current was demonstrated when addition of neutral amino acids stimulated the efflux of cations (presumably basic amino acids) in rBAT-injected oocytes. In the present paper, aminoisobutyric acid (AIB), a neutral amino acid analogue, was shown to induce outward currents (efflux of basic amino acids) through rBAT similar to those caused by alanine in terms of affinity, maximal currents and I-V curves. Despite generating similar currents, the AIB transport rate was more than 30 times lower than that of alanine, thus challenging the assumption that rBAT functions as a classical exchanger. Experiments using a cut-open oocyte voltage clamp demonstrated that AIB was capable of stimulating rBAT-mediated currents from either side of the membrane. AIB, like alanine, was able to stimulate the efflux of radiolabeled alanine and arginine while no rBAT-mediated efflux was measurable in the absence of external rBAT substrates. These results demonstrate that (i) the presence of amino acids is required on both sides of the membrane for rBAT to mediate amino acid flux and thus rBAT must be some type of exchanger but (ii) rBAT-mediated amino acid influx is not stoichiometrically related to the efflux. A model of a ``double gated pore'' is proposed to account for these properties of rBAT, which contravene standard models of exchangers and other transporters.

    Notes: Abstract  Genes cloned from diverse plants for resistance to different pathogens have sequence similarities in domains presumably involved in pathogen recognition and signal transduction in triggering the defense response. Primers based on the conserved regions of resistance genes often amplify multiple fragments that may not be separable in an agarose gel. We used denaturing polyacrylamide-gel electrophoresis to detect PCR products of plant genomic DNA amplified with primers based on conserved regions of resistance genes. Depending upon the primer pairs used, 30–130 bands were detected in wheat, rice, and barley. As high as 47%, 40%, and 27% of the polymorphic bands were detected in rice, barley, and wheat, respectively, and as high as 12.5% of the polymorphic bands were detected by certain primers in progeny from a cross of the wheat cultivars ‘Stephens’ and ‘Michigan Amber’. Using F6 recombinant inbred lines from the ‘Stephens’בMichigan Amber’ cross, we demonstrated that polymorphic bands amplified with primers based on leucine-rich repeats, nucleotide-binding sites and protein kinase genes, were inherited as single loci. Linkages between molecular markers and stripe rust resistance genes were detected. This technique provides a new way to develop molecular markers for assessing the genetic diversity of germplasm based upon potential candidate resistance genes in diverse species.

    Notes: Abstract  Lycopersicon peruvianum LA2157 originates from 1650 m above sea level and harbours several beneficial traits for cultivated tomatoes such as cold tolerance, nematode resistance and resistance to bacterial canker (Clavibacter michiganensis ssp. michiganensis). In order to identify quantitative trait loci (QTLs) for bacterial canker resistance, a QTL mapping approach was carried out in an F2 population derived from the interspecific F1 between Lycopersicon esculentum cv Solentos and L. peruvianum LA2157. Three QTLs for resistance mapped to chromosomes 5, 7 and 9 respectively. The resistance loci were additive and co-dominant with the QTL on chromosome 7 explaining the largest part of the variation for resistance in the F2 population. The combination of this QTL with either of the other two QTLs conferred a resistance similar to the level in the resistant parent L. peruvianum. Some RFLP markers flanking this QTL on chromosome 7 were converted into SCAR markers allowing efficient marker-assisted selection of plants with high resistance to bacterial canker.

    Notes: Summary.  The genome organization of the Helicoverpa armigera single-nucleo-capsid nucleopolyhedrovirus (HearNPV) was investigated and compared to that of other baculoviruses. A detailed physical map was constructed for the restriction enzymes BamHI, BglII, EcoRI, HindIII, KpnI, PstI, SacI and XhoI. The 130.1-kilobase pairs (kb) long viral genome was cloned as restriction fragments into a plasmid library, which covered 97.5% of the viral genome. From this plasmid library about 45 kb of dispersed sequence information was generated. Fifty-three putative open reading frames (ORFs) with homology to ORFs of other baculoviruses were identified and their location on the genome of HearNPV was determined. The arrangement of the ORFs along the HearNPV genome is significantly different from the multiple-nucleocapsid NPVs, AcMNPV, BmNPV and OpMNPV, which have a highly collinear genome, and the granulovirus XcGV. In contrast the genomes of HearNPV and SeMNPV are highly collinear. This close relatedness between an MNPV and an SNPV is supported by the phylogeny of specific genes of these two viruses and suggests that the morphotype (S or M) is only a taxonomic and not a phylogenetic denominator. The data presented here also give further support to the view that the gene distribution along baculovirus genomes can be used as a character to describe baculovirus relatedness independent of gene phylogeny. Five homologous regions (hrs) were located on the genome of HearNPV. This is also the first report of the existence of hrs in SNPVs and this observation suggests that hrs are a common feature of baculovirus genomes.

    Notes: Summary We present a (semilocal) Kantorovich-type analysis for Newton-like methods for singular operator equations using outer inverses. We establish sharp generalizations of the Kantorovich theory and the Mysovskii theory for operator equations when the derivative is not necessarily invertible. The results reduce in the case of an invertible derivative to well-known theorems of Kantorovich and Mysovskii with no additional assumptions, unlike earlier theorems which impose strong conditions. The strategy of the analysis is based on Banach-type lemmas and perturbation bounds for outer inverses which show that the set of outer inverses (to a given bounded linear operator) admits selections that behave like bounded linear inverses, in contrast to inner inverses or generalized inverses which do not depend continuously on perturbations of the operator. We give two examples to illustrate our results and compare them with earlier results, and another numerical example to relate our results to computational issues.

    Notes: Summary Surface melting is strongly influenced by the interaction between the solid-liquid interface and the liquid-vapour interface. Unretarded electromagnetic dispersion forces yield a potential of the formH/l 2, wherel is the distance between the interfaces (thickness of the quasi-liquid film) andH is the appropriate Hamaker constant. We have adopted the well-known treatment of dispersion forces of Lifshitzet al., where the frequency-dependent conductivities of solid and liquid are the primary quantities for the calculation of this Hamaker constant. Using the dielectric functions obtained from experimental data, numerical values forH are obtained for Al, Pb, Au, Bi and Ge. The Hamaker constant of the metals is found to be positive and is discussed in connection with recent experimental data on surface melting, particularly of Pb(110) and Al(110). The Hamaker constants of Bi (semimetal) and Ge (semiconductor) are negative, allowing at most blocked surface melting.

    Notes: Abstract  Fat facets is a Drosophila deubiquitinating enzyme required for eye development and early embryogenesis. Genetic evidence suggests that Fat facets deubiquitinates and thereby prevents the proteasomal degradation of specific substrates. The Drosophila Liquid facets protein is implicated as the critical substrate of Fat facets in the eye. A mouse homolog of Fat facets, called Fam, has been identified. The results of biochemical experiments implicate two different proteins, Af-6 and β-catenin, as substrates for Fam. Here, the functional relationship between Fat facets and Fam is explored. We show that Fam can substitute for Fat facets in all of its essential functions in Drosophila. In addition, we tested the hypothesis that Canoe and Armadillo, the Drosophila homologs of Af-6 and β-catenin, respectively, are important substrates for Fat facets in the Drosophila eye. We found no genetic evidence to support a role for either Canoe or Armadillo in the essential Fat facets pathways in Drosophila eye development. The significance of these results is discussed in light of the biochemical experiments that suggest that Af-6 and β-catenin are substrates of Fam.

    Notes: Summary A surgical method for bilateral retrocollis (BR) is presented: Selective resection of specific posterior cervical muscles for the treatment of bilateral retrocollis is described. The operative procedure includes partial resection of the upper part of the trapezius, part of the splenius muscles of both sides, part of the semispinalis capitis and semispinalis cervicis muscles of both sides. 15 cases were treated surgically during 1980–1989. Postoperative follow up (3–10 years) showed that the abnormal movements were abolished completely or for the greater part in 13 patients (87%) while preserving the normal posture and mobility.

    Notes: Abstract A cytofluorometric method was used to measure the number of IgE receptors available for binding and the native IgE content on microscopically identified peritoneal mast cells of Sprague-Dawley rats aged 4 to 25 weeks. During this period of growth, the mast cells increased progressively in size, granule mass and histamine content, but the number of IgE receptors did not increase in relation to body weight or age, although it varied greatly within the mast cell populations of individual rats as well as between the different rats. The peritoneal mast cells of these normal, naive, specific pathogen-free rats raised under controlled conditions and without signs of infections, were found to carry a significant number of IgE molecules of unknown specificity. The IgE content varied greatly among the individual mast cells within a given population, but, on average, as much as 74% of the receptors available for binding were occupied.

    Notes: Abstract Bradykinin, enhances prostanoid synthesis in aorta smooth muscle cells. Free arachidonic, acid also enhances prostanoid synthesis and bradykinin, unlike fatty acid releasing agents, has a synergistic effect with free arachidonic acid. Bradykinin promotes metabolite release from cells prelabeled with [14C]-arachidonic acid and this effect is blocked completely by indomethacin. High performance liquid chromatography shows increase amounts of labeled 6-keto-prostaglandin F1α, prostaglandin E2 and three additional cyclooxygenase-dependent metabolites but no increase in free arachidonic acid or other metabolites either in the absence or presence of indomethacin. Fatty acid releasing agents such as A23187 and cyclosporine A have very different effects on cells. These agents enhance levels of prostanoids, a number of other cyclooxygenase-independent metabolites, and free arachidonic acid which is even more elevated with added indomethacin. Bradykinin behaves in all respects like another agent, bacterial lipopolysaccharide, and the action of both agents is consistent with a mechanism involving cyclooxygenase rather than fatty release in the arachidonic acid cascade.
