Notes: Abstract. Three proteins have been identified in the milk of the common brush tail possum, Trichosurus vulpecula that from sequence analysis are members of the lipocalin family. They include β-lactoglobulin, which appears to have two forms; a homologue to the late-lactation protein found in tammar, Macropus eugenii; milk; and a novel protein termed trichosurin. Whereas β-lactoglobulin and trichosurin are both expressed throughout lactation, the late-lactation protein is not detected in samples taken before days 100–110 of lactation. The cDNAs encoding each of these proteins have been isolated from cDNA libraries prepared using possum mammary mRNA and sequenced. Phylogenetic analysis showed that the T. vulpeculaβ-lactoglobulin, along with two other macropod β-lactoglobulins, forms a subclass of β-lactoglobulins distinct from those for eutherian mammals; both marsupial late-lactation proteins appear to have similarities to a family of odorant-binding proteins, whereas trichosurin has similarities to the major urinary proteins of rodents.

Notes: Summary Genomic DNA from 18 phylogenetically diverse species of bony fish was hybridized with probes specific for the channel catfish immunoglobulin heavy chain constant (CH) gene, as well as with immunoglobulin heavy chain variable (VH) probes specific for five channel catfish VH gene families. The results showed that CH probes strongly hybridized only to genomic fragments from other catfish species. In contrast, restricted DNA from most other species hybridized with at least two channel catfish VH probes. In those species whose DNA hybridized with multiple VH probes, the restriction pattern of hybridizing fragments was probe-dependent. These studies suggest that (1) the CH gene defined in channel catfish appears to share similarity only with CH genes in other catfish species, (2) families of VH genes appear to have diverged in early phylogenetic lineages of teleosts, and (3) VH genes similar to those defined in catfish appear to be widely represented in phylogenetically diverse species of teleosts.

Notes: Abstract Aspartate aminotransferase isoenzymes are located in both the cytosol and organelles of eukaryotes, but all are encoded in the nuclear genome. In the work described here, a phylogenetic analysis was made of aspartate aminotransferases from plants, animals, yeast, and a number of bacteria. This analysis suggested that five distinct branches are present in the aspartate aminotransferase tree. Mitochondrial forms of the enzyme form one distinct group, bacterial aspartate aminotransferase formed another, and the plant and vertebrate cytosolic isoenzymes each formed a distinct group. Plant cytosolic isozymes formed a further group of which the plastid sequences were a member. The yeast mitochondrial and cytosolic aspartate aminotransferases formed groups separate from other members of the family.

Notes: Abstract A THE-1 sequence in intron 7 of the human dystrophin gene has been found to represent a new subfamily of THE-1 elements. The sequence is closely related to the MstII family of repetitive sequences and is more like single-copy sequences found in the galago genome than any other THE-1 sequence previously reported. This new THE-1 sequence has been compared with two other complete THE-1 sequences and three related long-terminal repeat elements that we have previously found in intron 7 of the dystrophin gene, and with members of the same family from elsewhere in the primate genome. Parsimony and deletion analysis show that the cluster of THE-1 sequences in intron 7 of the dystrophin gene has arisen from at least three individual insertion events, rather than from the insertion and duplication of a single progenitor sequence.

Description / Table of Contents: Zusammenfassung Eisenhaltiger Gahnite (Zn,Fe)Al2O4 ist in den metamorphen vulkanogenen massiven Sulfiderzkörpern Virginias als akzessorisches Mineral weit verbreitet.
Notes: Summary Ferroan-gahnite, (Zn,Fe)Al2O4, is a common accessory mineral found in association with the metamorphosed volcanogenic massive sulfide ore bodies of the Mineral District, Virginia. Gahnite is present throughout the mineralized zones that contain pyrite, sphalerite, chalcopyrite, and sulfosalts. Although constituting only 1–2 modal percent of most samples and displaying not well-defined distribution pattern, gahnite is especially abundant at the margins of the mineralized zones and locally constitutes as much as 35 modal percent. The compositions vary slightly from one deposit to another but all lie within the range of 68 to 88 mol.% gahnite (ZnAl2O4), 8 to 25 mol.% hercynite (FeAl2O4), and 1–17 mol.% spinel (MgAl2O4). Gahnite occurs within a wide variety of mineral assemblages, with or without coexisting sphalerite. The reflectance and unit cell dimension are sensitive to composition and decrease with increasing gahnite component. The approximate 8% reflectance at 540 nm is intermediate between most silicates and ore minerals such as sphalerite and magnetite and thus provides a ready method for recognition in reflected light. The gahnite is believed to have formed during amphibolite grade regional metamorphism as a result of either sphalerite desulfidation or the reaction of original zinc oxide phases.

Notes: Abstract  Objective. To demonstrate the MR depiction of the intertrochanteric or femoral neck extension of fractures of the greater trochanter, when standard radiographs show only a fracture of the greater trochanter. Design and patients. A retrospective review was performed of the MR and radiographic findings in 13 consecutive patients (10 men, 3 women; ages 24–86 years) with radiographic evidence of fracture of the greater trochanter who were examined with MR imaging. Results. The MR study displayed the fracture of the greater trochanter in all cases. In all but three patients, MR examinations displayed an extension of the fracture into the intertrochanteric region, and in one, also an extension into the femoral neck, although the cortex at this level was not interrupted. Conclusion. When there is radiographic evidence of an isolated fracture of the greater trochanter, MR often shows an intertrochanteric or femoral neck extension of the fracture in both young and older adults. This finding may be a factor in determining the need for surgical intervention.

Notes: Summary A case of superior fracture-dislocation of the hip following a low-energy injury is reported. The direction of the dislocation is uncommon. Radiological investigation was delayed for 5 months and then showed the appearances of a radiologically perfect false acetabulum. The fracture-dislocation was a type IV with an associated fracture of the femoral neck, an injury which is expected to produce a high rate of avascular necrosis of the femoral head. However, bone scans suggested that the femoral head was viable.

Notes: Abstract The productivity of ethanol fermentation processes, predominantly based on batch operation in the U.S. fuel ethanol industry, could be improved by adoption of continuous processing technology. In this study, a conventional yeast fermentation was coupled to a flat-plate membrane pervaporation unit to recover continuously an enriched ethanol stream from the fermentation broth. The process employed a concentrated dextrose feed stream controlled by the flow rate of permeate from the pervaporation unit via liquid-level control in the fermentor. The pervaporation module contained 0.1 m2 commercially available polydimethylsiloxane membrane and consistently produced a permeate of 20%–23% (w/w) ethanol while maintaining a level of 4%–6% ethanol in a stirred-tank fermentor. The system exhibited excellent operational stability. During continuous operation with cell densities of 15–23 g/l, ethanol productivities of 4.9–7.8 gl−1 h−1 were achieved utilizing feed streams of 269–619 g/l glucose. Pervaporation flux and ethanol selectivities were 0.31–0.79 lm−2 h−1 and 1.8–6.5 respectively.

Notes: Summary The influence of insulin on the enzyme glycogen synthase was studied in cultured skin fibroblasts from 5 non-diabetic subjects and 3 diabetic patients. Confluent fibroblast cultures were maintained in serum-free medium for 24 hours before exposure to graduated concentrations of insulin (0.1 to 2000 mU/ml) for one hour. The independent form of glycogen synthase activity (determined in the absence of glucose-6-phosphate) was increased in some experiments by exposure of cells to 0.1 mU of insulin/ml; 1.0mU/ml produced a consistent increase; but at least 1000 mU/ml was required for maximum effect. In the non-diabetic subjects, the mean baseline total activity of glycogen synthase was 0.182 μmol/mg protein/h with 16.8% in the independent form; with 1000 m U of insulin/ml the total activity was 0.167 μmol/mg/h with 45.4% in the independent form. In the diabetic fibroblasts, the mean percentage of glycogen synthase in the independent form was lower than in the control cells at baseline and with all concentrations of insulin utilised; at baseline, total activity was 0.117 μmol/mg protein/h with 11.7% in the independent form while with 1000 mU of insulin/ml the values were 0.146 μmol/mg protein/h and 30.7% respectively. Demonstration of an effect of insulin on glycogen synthase in human fibroblasts provides another phenomenon for comparison in cells from control subjects and diabetic patients.

Notes: Summary Both DFMO and methyl-GAG inhibit sequential enzymatic reactions in the pathway of polyamine biosynthesis. Since polyamines may be important factors in proliferation of cancer cells, we initiated a phase-I study of these agents in patients with advanced cancer. DFMO was given by mouth at a constant daily dose of 4 g/m2 starting on day 1 of the treatment protocol. The dose of methyl-GAG ranged from 200 to 700 mg/m2 administered IV every 2 weeks beginning on day 4. Twenty-two patients were entered into the protocol. Toxic reactions to this therapy were dose-related and included nausea, fatigue, diarrhea, and myelosuppression. One patient with colon cancer experienced a〉50% decrease in measurable disease but developed severe myelotoxicity. While DFMO was well tolerated, the combination of drugs appeared to cause substantially more hematologic and gastrointestinal toxicity than encountered during our recent experience with methyl-GAG used alone. We suggest that future studies of this drug combination carefully evaluate levels of polyamines and inhibition of enzymatic activity to minimize toxicity.
