    Notes: Asparagus is especially suitable for polyploidy breeding. The aim is to develop clone-based hybrid varieties, in particular male 4 × varieties. For this purpose, genetically divergent diploid idiotypes*) are subjected to mitotic or meiotic polyploidization. In developing hybrid partners, special attention is paid to resistance in addition to yield and quality characters. Early diagnoses are applied in selection so as to shorten breeding times. Somaclonal variations may occur relatively often on in vitro propagation; these need to be eliminated. Triplex and quadruplex type males are suitable for paternal hybrid partners. Strong inbreeding depressions need to be avoided in their production.

    Notes: Carrot breeding in the past 150 years has resulted in varieties with high yield, a short growing period, and excellent root colour. Recently, hybrid varieties have demonstrated good uniformity of roots, a quality accepted by most consumers. By contrast, only a few resistant varieties (mainly open-pollinated varieties) are offered by seed companies, most being resistant to Alternaria. Hybrid breeding offers a chance of combining good uniformity and different sources of resistance. Efforts in future breeding should concentrate on the improvement of health and the development of genotypes suitable for cultivation in suboptimal climates and regions, as well as for special applications.

    Notes: Studies were made on the components of lysosomes. Analyses were based upon the distribution of constituents between the insoluble fraction containing the membrane and bound proteins and tbe soluble fraction containing released enzymes. Results were compared with those obtained for mitochondria and microsomes. The composition of the lysosomal membrane was characteristic of a unit phospholipid-protein membrane. The sialic acid content of lysosomes was not much greater than that found in other subcellular fractions, suggesting that sialopolymers are not involved in a special role in lysosomes. The released enzyme fraction, which contained about myc of the total protein, also contained free flavins and free amino acids in amounts very much larger than those observed for mitochondria. Fe, Zn, Cu, Mn, and MO were the principal metals concentrated in lysosomes. The relationship between the chemical composition, structure, and function of lysosomes is discussed.

    Description: Rationale: Both β-adrenergic receptor (β-AR) and G q -coupled receptor (G q R) agonist–driven signaling play key roles in the events, leading up to and during cardiac dysfunction. How these stimuli interact at the level of protein kinase D (PKD), a nodal point in cardiac hypertrophic signaling, remains unclear. Objective: To assess the spatiotemporal dynamics of PKD activation in response to β-AR signaling alone and on coactivation with G q R-agonists. This will test our hypothesis that compartmentalized PKD signaling reconciles disparate findings of PKA facilitation and inhibition of PKD activation. Methods and Results: We report on the spatial and temporal profiles of PKD activation using green fluorescent protein-tagged PKD (wildtype or mutant S427E) and targeted fluorescence resonance energy transfer–based biosensors (D-kinase activity reporters) in adult cardiomyocytes. We find that β-AR/PKA signaling drives local nuclear activation of PKD, without preceding sarcolemmal translocation. We also discover pronounced interference of β-AR/cAMP/PKA signaling on G q R-induced translocation and activation of PKD throughout the cardiomyocyte. We attribute these effects to direct, PKA-dependent phosphorylation of PKD-S427. We also show that phosphomimetic substitution of S427 likewise impedes G q R-induced PKD translocation and activation. In neonatal myocytes, S427E inhibits G q R-evoked cell growth and expression of hypertrophic markers. Finally, we show altered S427 phosphorylation in transverse aortic constriction–induced hypertrophy. Conclusions: β-AR signaling triggers local nuclear signaling and inhibits G q R-mediated PKD activation by preventing its intracellular translocation. PKA-dependent phosphorylation of PKD-S427 fine-tunes the PKD responsiveness to G q R-agonists, serving as a key integration point for β-adrenergic and G q -coupled stimuli.

    Description: Background: The Deepwater Horizon disaster was the largest marine oil spill in history, and total vertical exposure of oil to the water column suggests it could impact an enormous diversity of ecosystems. The most vulnerable organisms are those encountering these pollutants during their early life stages. Water-soluble components of crude oil and specific polycyclic aromatic hydrocarbons have been shown to cause defects in cardiovascular and craniofacial development in a variety of teleost species, but the developmental origins of these defects have yet to be determined. We have adopted zebrafish, Danio rerio, as a model to test whether water accumulated fractions (WAF) of the Deepwater Horizon oil could impact specific embryonic developmental processes. While not a native species to the Gulf waters, the developmental biology of zebrafish has been well characterized and makes it a powerful model system to reveal the cellular and molecular mechanisms behind Macondo crude toxicity. Results: WAF of Macondo crude oil sampled during the oil spill was used to treat zebrafish throughout embryonic and larval development. Our results indicate that the Macondo crude oil causes a variety of significant defects in zebrafish embryogenesis, but these defects have specific developmental origins. WAF treatments caused defects in craniofacial development and circulatory function similar to previous reports, but we extend these results to show they are likely derived from an earlier defect in neural crest cell development. Moreover, we demonstrate that exposure to WAFs causes a variety of novel deformations in specific developmental processes, including programmed cell death, locomotor behavior, sensory and motor axon pathfinding, somitogenesis and muscle patterning. Interestingly, the severity of cell death and muscle phenotypes decreased over several months of repeated analysis, which was correlated with a rapid drop-off in the aromatic and alkane hydrocarbon components of the oil. Conclusions: Whether these teratogenic effects are unique to the oil from the Deepwater Horizon oil spill or generalizable for most crude oil types remains to be determined. This work establishes a model for further investigation into the molecular mechanisms behind crude oil mediated deformations. In addition, due to the high conservation of genetic and cellular processes between zebrafish and other vertebrates, our work also provides a platform for more focused assessment of the impact that the Deepwater Horizon oil spill has had on the early life stages of native fish species in the Gulf of Mexico and the Atlantic Ocean.
