    Abstract: Photoelectrochemical immunoassays incorporating specific antigen-antibody recognition reactions with the photon-electron conversion capabilities of photocatalysts have been developed for biomarker detection, but most involve bulky and expensive equipment and are unsuitable for point-of-care testing. Herein, a portable smartphone-based photoelectrochemical immunoassay was innovatively designed for the on-site detection of breast cancer biomarkers (human epidermal growth factor receptor 2; HER2). The system consists of a split-type immunoassay mode, disposable screen-printed electrode covered with hierarchical Co 9 S 8 @ZnIn 2 S 4 heterostructures, an integrated circuit board, and a Bluetooth smartphone equipped with a specially designed app. Using alkaline phosphatase (ALP) catalytic strategy to in situ generate ascorbic acid (AA) for electron-donating toward Co 9 S 8 @ZnIn 2 S 4 heterostructures, an immunoreaction was successfully constructed for the HER2 detection in the real sample due to the positive correlation of the photocurrent signal to electron donor concentration. Differential charge density indicates that the formation of Co 9 S 8 @ZnIn 2 S 4 heterojunction can facilitate the flow of charges in the interface and enhance the photocurrent of the composite. More importantly, the measured photocurrent signal can be wirelessly transmitted to the software and displayed on the smartphone screen to obtain the corresponding HER2 concentration value. The photocurrent values linearly with the logarithm of HER2 concentrations range spanned from 0.01 ng/mL to 10 ng/mL with a detection limit of 3.5 pg/mL. Impressively, the clinical serum specimen results obtained by the proposed method and the wireless sensing device are in good agreement with the enzyme-linked immunosorbent assay (ELISA).

    Abstract: Carbon’s unique ability to have both sp 2 and sp 3 bonding states gives rise to a range of physical attributes, including excellent mechanical and electrical properties. We show that a series of lightweight, ultrastrong, hard, elastic, and conductive carbons are recovered after compressing sp 2 -hybridized glassy carbon at various temperatures. Compression induces the local buckling of graphene sheets through sp 3 nodes to form interpenetrating graphene networks with long-range disorder and short-range order on the nanometer scale. The compressed glassy carbons have extraordinary specific compressive strengths—more than two times that of commonly used ceramics—and simultaneously exhibit robust elastic recovery in response to local deformations. This type of carbon is an optimal ultralight, ultrastrong material for a wide range of multifunctional applications, and the synthesis methodology demonstrates potential to access entirely new metastable materials with exceptional properties.

    Abstract: Although the molecular mechanism is not clear, the clinically tested drug ketamine has rapid antidepressant action that does not require the multiple weeks of treatment needed for other antidepressant drugs to have an effect. We showed that ketamine potentiated Schaffer collateral–CA1 cell excitatory synaptic transmission in hippocampal slice preparations from rodents and enhanced the phosphorylation of the GluA1 subunit on Ser 845 of the AMPA-type glutamate receptor in the hippocampal area CA1. These effects persisted when γ-aminobutyric acid (GABA) receptors were pharmacologically blocked. Ketamine reduced behavioral despair in wild-type mice but had no effect in GluA1 S845A knock-in mutant mice. Presynaptic (CA3 pyramidal cell), but not postsynaptic (CA1 pyramidal cell), deletion of N -methyl- d -aspartate (NMDA)–type glutamate receptors eliminated the ketamine-induced enhancement of excitatory synaptic transmission in hippocampal slices and the antidepressant actions of ketamine in mice. The synaptic and behavioral actions of ketamine were completely occluded by inhibition or deletion of the hyperpolarization-activated cyclic nucleotide–gated channel 1 (HCN1). Our results implicate presynaptic NMDA receptor inhibition followed by reduced activity of presynaptic HCN1 channels, which would result in an increase in glutamate release and postsynaptic glutamate receptor activity, as a mechanism of ketamine action. These data provide a mechanism for changes in synaptic activity that could explain the fast-acting antidepressant effects of this drug.
