    Notes: A model for hemoglobin is proposed and its application to allosteric enzymes is discussed with particular reference to asparate transcarbamylase. The main assumptions made are that the molecule is composed of subunits and that occupation of a sub-unit produces a conformational change which affects the occupational probability of neighboring subunits. The results compare favorably with experiment and a number of specific predictions are made for aspartate transcarbamylase.

    Notes: Utilization of n-heptane by a Pseudomonad was studied in pilot-size butch cultures. Optimal pH and temperature were determined by a factorial design and a medium based upon mineral uptake rates was formulated. High cell yields were obtained by volatilizing heptane in the incoming air and thereby achieving good hydrocarbon dispersion. Hydrocarbon carried by effluent gases was recovered and recycled. In cultures where pH is not controlled, decrease in the electrolytic conductivity of the medium was found to be indicative of viable cells and was used in monitoring bacterial propagation. If not checked, increase in salinity in pH controlled cultures was found to affect cell production negatively. Viscosity changes were not very significant. Heptane to aqueous medium ratio was found to affect oxygen supply to the system due to higher dissolved oxygen concentrations associated with hydrocarbons.

    Notes: A new vapor-liquid equilibrium experimental facility has been built for and operated on light gas-hydrocarbon systems. The range of operating conditions are 3 to 1,000 atm. and 25° to 400°C. A dynamic flow type of equilibrium cell is used. This and most of the other apparatus are from designs by Michels. Solubilities of hydrogen in benzene and in cyclohexane were obtained at 150° and 250°F. and at pressures up to 10,000 lb./sq.in.abs.Compositions of the equilibrium vapor phases were determined so the vapor-liquid K ratios could be derived. Thermodynamic consistency tests were applied to the data with the conclusion that the new data are consistent with volumetric properties, predicted by other methods.

    Notes: The horizontal rotating cylinder blood oxygenator has been developed to minimize shear, eliminate bubbles, reduce holdup, and provide for rapid change of the controlled temperature of the blood.To aid in the development of this apparatus a detailed mathematical model was formulated. A simplified model was also developed which yielded an analytical expression relating the amount of material transferred between the gas and liquid in the apparatus to operating variables. In developing these models an attempt was made to include a realistic mechanism for oxygen absorption in blood.Preliminary experiments were carried out by absorbing carbon dioxide into distilled water in the horizontal rotating cylinder. These preliminary results were found to be in good agreement with previous theoretical predictions and calculations that indicated that the equipment should be adequate to act as a lung substitute for a dog. When it was employed in this manner during open-heart surgery, insufficient oxygen was transferred to support the dog.It is concluded that the most probable reason for failure of the horizontal cylinder to act satisfactorily as a blood oxygenator is poor mixing in the blood pool.

    Notes: We have observed that the viscosity of a solution of E. coli chromatin increases as the shear to which the solution is subjected is suddenly increased. This dilatant effect is opposite to the usual thixotropic non-Newtonian effects that are observed in DNA solutions. We suggest that the observed dilatancy may be explained in terms of shear-induced aggregation of large DNA molecules. Conditions for aggregation are discussed in the context of the “cluster theory” of condensation.
