    Abstract: A system of auxotyping described in 1973 is based on the differing nutritional requirement patterns of Neisseria gonorrhoeae strains. Our ongoing evaluation of the reliability of auxotyping has involved a study of the constancy of characteristics of gonococci isolated at one time from two or more sites of a given subject. The auxotypes and minimal inhibitory concentration (MIC) of penicillin G were determined for 181 isolates obtained from 84 patients with uncomplicated gonorrhea, for 16 isolates from 8 couples with uncomplicated gonorrhea, and for 21 isolates from 12 other patients, 9 with disseminated gonococcal infection and three consorts. The penicillin MIC served to distinguish between many members of auxotypes 1, 2, and 3, which are commonly involved in uncomplicated gonorrhea. Thus, for proline-requiring gonococci (auxotype 2) the MIC ranged from 0.01 to 1.2 IU of penicillin per ml. The profile of gonococcal responses to seven other antibacterial drugs provided useful additional information where the extent of phenotypic similarity was in doubt. In all but seven instances, the gonococci isolated from different sites of the same patient, or from a consort, had the same nutritional requirements and penicillin MIC. The gonococci isolated from one patient with disseminated gonococcal infection and from one of her two sexual contacts had nutritional requirements for arginine, hypoxanthine, uracil, and thiamine pyrophosphate, whereas the strain isolated from her second contact differed in having no requirement for thiamine pyrophosphate. The paired cervical and rectal isolates from one patient with uncomplicated gonorrhea differed only with respect to a requirement for hypoxanthine. Pairs of isolates from three patients differed slightly in degree of susceptibility to penicillin. In the remaining two instances, however, numerous differences between the isolates from the endocervix and the anal canal of a given patient indicated the presence of concomitant infections with different strains of N. gonorrhoeae .

    Abstract: The lifetimes of the 7 2 P 3/2 and 7 2 P 1/2 states of atomic cesium have been determined using delayed coincidence techniques. A nitrogen laser pumped tunable dye laser was used to excite the cesium vapor and direct measurements of the fluorescence decay yielded the following lifetimes: τ 3/2 (7 2 P 3/2 ) = 136 ± 4 ns and τ 1/2 (7 2 P 1/2 ) = 158 ± 5 ns. These values are in good agreement with recent measurements and calculations.

    Abstract: Surveillance of seven types of delicatessen salads (chicken, egg, ham, macaroni, American potato, shrimp, and tuna) was conducted from March through July, 1974. Liver spread and sandwich spread were included in the sampling. Products were obtained from approximately 150 retail establishments. A separate surveillance of one central production unit and approximately 18 retail establishments, which it supplied, was also conducted. Twenty six percent to 85% of samples of products obtained from general retail outlets had aerobic plate count (APC) values of less than 100,000 per gram. Thirty six percent to 79% had coliform Most Probable Number (MPN) levels of less than 100 per gram, while 83 to 100% contained less than 10 Escherichia coli per gram. Some samples of each of the products contained low numbers of coagulase-positive staphylococci. Seventy one per cent to 96% of samples of various products, obtained from a central production unit, yielded APC values of less than 100,000 per gram. Forty five percent to 94% had coliform MPN values of less than 100 per gram. Two of six products contained low levels of coagulase-positive staphylococci in 9% and 2% of the samples examined.

    Abstract: Electrophysiological studies of rat islet cells in monolayer culture were undertaken to determine the role of transmembranous ionic fluxes in the inhibitory action of somatostatin on insulin release. In the presence of somatotropin release inhibiting factor (SRIF) (2.5 nM), hyperpolarization occured with or without glucose (16.6 mM) in the medium. SRIF also inhibited the incidence of glucose-induced spike activity. The inhibitory action of SRIF occurred within 5 min and was readily reversible. An increase in extracellular K + (5–13 mM) or Ca2+ (2.3–4.6 mM) prevented SRIF inhibition of glucose-induced electrical activity. The secretory response of cultured islets to glucose (16.6 mM) was completely inhibited by SRIF (2.5 nM). The presence of high [Ca 2+ ]0 or [K + ]0, enhanced insulin release in the presence of SRIF and glucose. Although phentolamine (5.0 μg/ml) did not block the inhibition of glucose-induced electrical responses by SRIF, it prevented the inhibitory action of epinephrine (0.2 μg/ml). It is concluded that the primary action of SRIF is to alter transmembranous cationic fluxes, as manifested by hyperpolarization and a decrease in the incidence of spike activity, which may prevent glucose from eliciting a normal secretory response. insulin secretion; epinephrine; alpha-adrenergic blockade; stimulus-secretion coupling Submitted on February 14, 1977
