    Notizen: In this paper we analyse a simple but non-trivial steady-state creeping elasto visco-plastic (Maxwell fluid) radial flow problem, paying particular attention to the effects of the boundary conditions. Solutions are obtained by integration of a governing equation on stress, using the Runge-Kutta method for initial value problems and finite differences for boundary value problems. A more general approach by the finite element method, which solves for the velocity field rather than the stress field, and which is applicable to a wide range of problems, is presented and tested using the radial flow example.

    Notizen: A frontal-skyline method is presented which allows for compact skyline storage while using the frontal method. It is shown that the method requires the same minimum core storage as the frontal method and fewer transfers to and from disc than the blocked-skyline method. Subroutines necessary for its implementation with finite element codes are given.

    Notizen: The evolution of the latex particle diameter distribution during batch emulsion polymerization is investigated, with emphasis on changes in the breadth of the size distribution. A model utilizing a surface area-dependent volumetric growth rate of a single particle results in a time-invariant standard deviation of the size distribution during periods of particle growth only. This behavior is reconciled with some experimental observations by considering the occurrence of particle nucleation during some part of the growth interval. Conclusions based on the results of the model suggest that higher inhibitor and low emulsifier concentrations favor narrow particle size distributions.

    Notizen: Two monoclonal antibodies reactive for α-tubulin but not for β-tubulin have been prepared, characterized in terms of their relative binding to tubulins from differnt sources by a solid-phase binding assay, immunoautoradiography, and indirect immunofluorescence, and utilized to study flagellar motility. Our results demonstrate that α-tubulins from different species, and even from different tissues of the same species, are nonidentical. Especially interesting was the observation that one of the antibodies, Ab2, immunofluorescently stained microtubules of chick embryo fibroblast cells, but was completely unreactive for microtubules of rat kangaroo (PtK2) fibroblasts; a different antibody, Ab1, stained both cell types. Results of these and additional experiments clearly show that Ab1 and Ab2 recognize discrete and different epitopes on α-tubulin.Monoclonal antitubulins Ab1 and Ab2 each inhibited the bend amplitude of reactivated sea urchin spermatozoa without affecting beat frequencies or the ability of the outer doublet microtubules to slide past each other in elastase-digested models. These results, together with those obtained previously using rabbit polyclonal antitubulin antibodies [Asai and Brokaw, 1980], demonstrate that inhibition of bend amplitude is a common property of antitubulin antibodies and is not due to the binding of antibodies to one specific site on the axoneme. Our results suggest that tubulin subunit conformational changes may occur on the outer doublet lattice and may be integrally involved in the mechanism and control of flagellar bending.

    Notizen: Dissociated bovine brain microtubule protein has been shown to reassemble at 0°C in the presence of the drug taxol. Tubulin polymerization was monitored both by electron microscopy of the polymeric structures and by incorporation of tritiated GTP into filterable polymeric structures. Most of the labeled guanine nucleotide uptake into tubulin polymeric structures occurred in the first 30 minutes of incubation with the drug. The initial polymerization event results in the formation of protofilamentous tubulin ribbons. The first microtubules were noted after 1 hour of incubation with the drug. After 20 hours of incubation at 0°C with taxol, the bulk of the polymerized tubulin appeared to be in the form of microtubules. Cold-stable tubulin rings with a mean diameter of 34 nm were present in the reaction mixture before the addition of taxol and throughout the 20-hour incubation. Most of the rings were apparantly not involved in the taxol-induced microtubule assembly. The results are consistant with a model whereby taxol induces an initial formation of protofilamentous ribbon structures, mostly from free tubulin dimers, and a slower subsequent folding of the ribbon structures into microtubules.

    Notizen: Enzymatic corn starch liquefaction with α-amylase was carried out. Molecular weight distributions of the resulting hydrolysates are presented using aqueous size exclusion chromatographic techniques. It is demonstrated that despite the fact that the enzyme employed reacts in a random endoacting manner, the product distributions are nonrandom. The results are explained in part by a multimerization process whereby the polymeric substrate molecules preferentially associate, forming intermolecular aggregates. These aggregates are either a consequence of the manner in which the material is deposited into the native granular structure of starch or due to intrinsic physical chemical properties of the polysaccharide. In the latter case, the results are shown to correspond to known multimerized amylose, although complete characterization of the polysaccharide is currently not available. The results presented are used to develop a simplified kinetic model of starch liquefaction and shown to simulate the product distributions accurately.

    Notizen: A well-known nucleation-growth model, used extensively to describe the crystallization of synthetic polymers, has been applied to collagen. The model gives (1) a general representation of the effect of temperature on the rate of collagen fibril formation, (2) a semi-quantitative description of the effect of collagen concentration on growth rate, and (3) an estimate of fibril diameter that is in agreement with observations made by electron microscopy. The mechanism described is apparently that of the early stages of fibril growth, in which microfibrils are formed. An important parameter required in this approach is the free energy of fibril formation. The free energy was approximated by use of an expression derived by Flory for rodlike polymers. Flory's equation plus experimental data on collagen solubility permitted calculation of the free energy over a range of temperature. The resultant expression was of the same form as one commonly used in describing crystallization of synthetic polymers.
