    Notes: Two studies are diescribed in which synthetic peptides have been designed and examined to address biochemical problems inherent in hydorphobic environments: (1) The cyclic hexapeptide cyclo-(D-Tyr(Bzl)-Gly-Ile-Leu-Gln-Pro) was synthesized as a model of an interior β-turn from the protein lysozyme. Conformational analysis by proton nmr methods, including two-dimensional nulcear Overhauser effect spectroscopy, revealed that the model peptide adopts one conformation in chloroform/dimethyl sulfoxide (98.2) and tetramethylene sulfone solutions. The conformation consists of two linked β-turns, one with the same sequence (Gly-Ile-Leu-Gln) and geometry (Type I) as the protein turn. (2) Major portions of the λ-receptor protein (LamB) signal sequences from E. coli wildtype and mutant strains have been synthesized. The conformational properties and membrane interactions of these synthetic signal peptides correlate with the in vivo export function of the wild type and mutant strains. Functional signal sequences are significantly richer in α-helix in aaqueous trifluoroethanol, lysolecithin, or sodium do-decyl sulfate solution than is a nonfunctional mutant signal sequence.

    Notes: The complex series of reactions that takes place between WCl6 and cyclopentene was studied spectroscopically. The products of reaction remain soluble when chlorobenzene is used as solvent. A number of intermediates was observed and the kinetics of formation and removal of these was studied in detail. A complex series of reactions was postulated to account for these changes and the significance of each reaction to the overall scheme was determined by computer simulation of the changes that occur. One of the products has particular significance in the scheme of reactions since it is believed that this species is eventually responsible for the monometallic initiation of the polymerization of cyclopentene. The reaction was also studied gas chromatographically when it was shown that the reaction is accompanied by the formation of a complex mixture of products. One of these products was shown to have the mass spectrum that would be expected of 3-chlorocyclopentene and a reaction scheme that would account for its formation is postulated. The reactions involved in the generation of centres active towards the formation of polypentenamer were considered for both monometallic and bimetallic (WCl6/AlR3) catalyst.

    Notes: A novel method for crosslinking polyetheretherketone via imine formation at carbonyl groups is presented. In polyetheretherketone these Schiff bases form short, stiff crosslink junctions which are stable up to 630°C. Dynamic mechanical and thermal properties of polyetheretherketone and imine crosslinked polyetheretherketone are reviewed. A large reduction in the temperature sensitivity of the material stiffness has been achieved. Increases in the glass transition temperature from 143°C to greater than 280°C have been obtained.

    Notes: Increasingly widespread usage of stable isotope tracers to aid clinical diagnosis and support basic research has stemmed from both advances in mass spectrometry and the availability of competitively priced labelled compounds. Stable isotopes have been used generally to investigate normal and abnormal metabolic pathways, to estimate energy expenditure and body composition and to quantitate substrate flux and oxidation rates. Despite the fact that the underlying principles relating to the use of stable isotopes for in vivo studies are straightforward, careful consideration must be given to all aspects of human studies. This review highlights some of these, including choice of label and tracer molecule, mode of tracer administration and sampling site, analytical instrumentation, interpretation of data and ethical constraints.

    Notes: A control system has been devised for the maintenance of stable ammonium concentrations throughout a fedbatch fermentation. The control system is based on an ammonia gas-sensing electrode that monitors a pH-adjusted effluent stream from the fermentor. To overcome the time lag between the fermentor and the electrode, feedback control included metered flows of ammonium to both the fermentor and the electrode vessel. The system was used to study the growth of Escherichia coli B (ATCC 11303) at controlled ammonium concentrations of 5 to 200mM. Apparent specific growth rates, biomass and protein production, and glucose yields were essentially constant from 5 to 170mM. Above 170mM ammonium growth was inhibited. As ammonium concentration decreased from 170 to 5mM, ammonium yields increased from 1 to 24 g cell dry wt/g ammonium utilized. The results demonstrate that control of ammonium concentrations at levels so low that ammonium would be exhausted in batch fermentations can significantly increase overall ammonium yields.

    Notes: A nitrate control system has been devised for the maintenance of stable nitrate concentrations throughout fed-batch fermentations of Corynebacterium glutamicum. The feedback control system was based on the use of a nitrate-ion-selective electrode to directly monitor the nitrate levels in the fermentor and an automatic controller to activate a nitrate feed pump. The electrode which was used for controlling the nitrate level was stable through-out the fermentation period. The apparent maximum specific growth rate, biomass production, protein production, biomass yields on glucose and nitrate, and amino acid production were all optimal at approximately 50mM nitrate.
