    Abstract: Sequence analysis has shown significant homology between the catalytic regions of the mammalian ras GTPase-activating protein (GAP), yeast Ira1p and Ira2p (inhibitory regulators of the RAS-cyclic AMP pathway), and neurofibromin, the protein encoded by the NF1 gene. Yeast expression experiments have confirmed that a 381-amino-acid segment of neurofibromin, dubbed the GAP-related domain (GRD), can function as a GAP. Using the RNA polymerase chain reaction with primers flanking the NF1-GRD, we have identified evidence for alternative splicing in this region of the NF1 gene. In addition to the already published sequence (type I), an alternative RNA carrying a 63-nucleotide insertion (type II) is present in all tissues examined, although the relative amounts of types I and II vary. The insertion is conserved across species but is not present in GAP, IRA1, or IRA2. GenBank searches have failed to identify significant similarity between the inserted sequence and known DNA or protein sequences, although the basic amino acid composition of the insertion shares features with nuclear targeting sequences. Expression studies in yeasts show that despite the partial disruption of the neurofibromin-IRA-GAP homology by this insertion, both forms of the NF1-GRD can complement loss of IRA function. In vivo assays designed to compare the GAP activity of the two alternatively spliced forms of the NF1-GRD show that both can increase the conversion of GTP-bound ras to its GDP-bound form, although the insertion of the 21 amino acids weakens this effect. The strong conservation of this alternative splicing suggests that both type I and II isoforms mediate important biological functions of neurofibromin.

    Abstract: The gene for neurofibromatosis type 1 (NF1) was recently identified by positional cloning. The complete cDNA encodes a polypeptide of 2818 amino acids. To study the NF1 gene product, antibodies were raised against both fusion proteins and synthetic peptides. Initial characterization of two anti-peptide antibodies and one fusion-protein antibody demonstrated a specific protein of approximately 250 kDa by both immunoprecipitation and immunoblotting. This protein was found in all tissues and cell lines examined and is detected in human, rat, and mouse tissues. To demonstrate that these antibodies specifically recognize the NF1 protein, additional fusion proteins containing the sequence specific to the synthetic peptide were generated. Both peptide antisera recognize the proper specific fusion proteins so generated. Immunoprecipitates using the peptide antisera were shown to recognize the same protein detected by immunoblotting with either the other peptide antiserum or the fusion-protein antiserum. Immunoblotting using antiserum specific to spatially distinct epitopes conducted on tissue homogenates demonstrated the NF1 protein in all adult tissues. Based on the homology between the NF1 gene product and members of the GTPase-activating protein (GAP) superfamily, the name NF1-GAP-related protein (NF1GRP) is suggested.

    Abstract: Carbapenem resistance was studied in a clinical isolate of Enterobacter cloacae, strain 201 (MIC of imipenem and meropenem, 16 micrograms/ml). This strain was analyzed comparatively with the carbapenem-susceptible parent strain 200, an equally susceptible revertant, 201-Rev, and in vitro-selected mutants with different levels of carbapenem resistance. All strains produced similarly high amounts of the same cephalosporinase (pIapp = 8.8). Strain 201 apparently lacked two major outer membrane proteins of ca. 37 and 38 kDa, while 201-Rev produced only the 37-kDa protein. The permeability coefficient, determined with cephaloridine, was reduced up to ninefold in the resistant strains which also showed a substantial reduction in the uptake of [14C] meropenem. The introduction of the plasmid-borne ampD gene (whose product decreases the expression of ampC) resulted in almost complete cessation of cephalosporinase production in all strains and a substantial decrease in the MICs of the carbapenems which remained, however, 8- to 16-fold higher than those determined for the susceptible strains containing the ampD gene. This "residual" resistance was attributed to reduced outer membrane permeability. The contribution of cephalosporinase production was verified in a reverse experiment, in which the introduction of ampC into a low-level cephalosporinase producer resulted in a fourfold increase in the carbapenem MICs. From these results, we infer that reduced outer membrane permeability and high-level cephalosporinase production can operate in conjunction in clinical isolates of E. cloacae to confer imipenem resistance.

    Abstract: In this paper (III) we report real-time studies of the picosecond dynamics of iodine in Ne clusters I*2Nen(n = 2–4) → I*2 + nNe. The results are discussed in relation to vibrational predissociation (VP), basic to the I2X systems, and to the onset of intramolecular vibrational-energy redistribution (IVR). The latter process, which is a precursor for the evaporation of the host atoms or for further fragmentation, is found to be increasingly effective as the cluster size increases; low-energy van der Waals modes act as the accepting (bath) modes. The reaction dynamics for I2Ne2 are examined and quantitatively compared to a simple model which describes the dynamics as consecutive bond breaking. On this basis, it is concluded that the onset of energy redistribution is observed in I2Ne2. Comparison of I2Ne and I2Ne2 to larger clusters (n=3,4) is accomplished by introducing an overall effective reaction rate. From measurements of the rates and their dependence on v′i, the initial quantum number of the I2 stretch, we are able to examine the dynamics of direct fragmentation and evaporation, and compare with theory.

    Abstract: In this second paper (II) of a series, we report our picosecond time-resolved studies of the state-to-state rates of vibrational predissociation in iodine–rare gas (van der Waals) clusters. Particular focus is on the simplest system, I2He, which serves as a benchmark for theoretical modeling. Comparisons with I2Ne and I2H2 are also presented. The results from measurements made in real time are compared with those deduced from linewidth measurements, representing a rare example of a system studied by both methods under identical conditions and excited to the same quantum (v′i) states. The discrepancies are discussed in relation to the origin of the broadening and preparation of the state. The rates as a function of v′i display a nonlinear behavior which is examined in relation to the energy-gap law. The measured absolute rates and their dependence on v′i are compared with numerous calculations invoking classical, quantum, and semiclassical theories. In the following paper (III in this series), the cluster size of the same system, I2Xn, is increased (n=2–4) and the dynamics are studied.

The state-to-state rates, k(νi′;νf′), are directly measured and related to the homogeneous broadening of the initial state, and to product state distributions in the exit channel.
