    Notes: The production of the neuroinhibitory and neuroprotective metabolite kynurenic acid (KYNA) was investigated in rat brain by examining its biosynthetic enzyme, kynurenine aminotransferase (KAT). By using physiological (low micromolar) concentrations of the substrate L-kynurenine (KYN) and by determining the irreversible conversion of [3H] KYN to [3H] KYNA as a measure of KAT activity, a novel, simple, and sensitive assay was developed which permitted the detailed characterization of the enzyme. Only a single protein, which under routine assay conditions showed approximately equal activity with 2-oxoglutarate and pyruvate as the aminoacceptor, was found in rat brain. The enzyme was distributed heterogeneously between the nine brain regions studied, with the KAT-rich olfactory bulb displaying approximately five times higher activity than the cerebellum, the area with lowest KAT activity. In subcellular fractionation studies, the majority of KAT was recovered in mitochondria. In contrast to many known aminotransferases, partially purified KAT was shown to be highly substrate-specific. Thus, of the amino acids tested, only α-aminoadipate and tryptophan displayed moderate competition with KYN. Notably, 3-hydroxykynurenine, reportedly a very good substrate of KAT, competed rather poorly with KYN as well. Aminooxyacetic acid, a nonspecific transaminase inhibitor, blocked KAT activity with an apparent Ki of 5 μM. Kinetic analyses with partially purified rat brain KAT revealed a Km of 17 μMfor KYN with 1 mM 2-oxoglutarate, but a much higher Km(910 μM) with 1 mM pyruvate. Km values for 2-oxoglutarate and pyruvate were 150 and 160 μM, respectively. The cellular localization of KAT was examined in striatal homogenates obtained from rats 7 days after an intrastriatal injection of quinolinate. At that time of almost complete neuronal destruction and pronounced astrocytic proliferation, enzyme activity in the lesioned striatum was almost twice as high as in controls, suggesting a preferential astroglial localization of brain KAT. The characteristics of KAT described here are compatible with a central role of the enzyme in brain KYNA function in vivo. Abnormal KAT activity, therefore, should be considered as an etiological factor in pathological phenomena related to dysfunction of excitatory amino acid receptors in the brain.

    Notes: Abstract The paraflagellar bodies (PFBs) of isolated flagella ofEuglena gracilis were investigated microspectrophotometrically using a visible- and infrared-light microscope with image analyzer and microspectrophotometer. Flagella with attached PFBs were separated from the cell bodies by a short exposure to near-UV light. Fluorescence-emission spectra (excitation at 365 nm) of single PFBs had maxima near 470 and 520 nm, indicating the presence of pterins and flavins. No fluorescence was associated with the flagella themselves. Pterin- and flavin-like fluorescence emission was also found in blue-fluorescing vesicles distributed throughout the entire cell body ofEuglena. Their characterization by microfluorimetry was greatly aided by the use of chlorophyll-free mutants in which the signal-to-noise ratio was distinctly enhanced because of the lack of chlorophyll fluorescence. Our finding of flavin-like fluorescence associated with PFB strengthens similar earlier reports in the literature. The occurrence of pterin-like fluorescence in the PFB lends further support to our earlier proposal that pterins as well as flavins may function as photoreceptor pigments for near-UV and blue light.

    Notes: Abstract The AMPA antagonists NBQX (2.5, 5, 10 mg/kg) and GYKI 52466 (4.8, 8 mg/kg) were investigated in haloperidol (0.5 mg/kg)-induced catalepsy in the rat. The effects of AMPA antagonists administered either alone or in combination with the noncompetitive NMDA antagonist dizocilpine (0.02 mg/kg), with the dopamine D-2 agonist quinpirole (1 mg/kg) or with L-DOPA (50, 100 mg/kg plus benserazide) were tested. NBQX or GYKI 52466 did not exert anticataleptic effects, neither alone nor in combination with dizocilpine, quinpirole or l-DOPA. Thus, in the rat inhibition of AMPA receptors with NBQX or GYKI 52466 does not have effects predictive for an antiparkinsonian potential.

    Notes: Abstract The high-affinity uptake of transmitter glutamate and aspartate in hippocampal slices incubated in sera from patients with hepatic encephalopathy was dramatically reduced in neuropil areas by more than 50 % compared with the control level. Adenosine compensates for these reduction in reuptake capacity in a concentration-dependent fashion reaching normal values at 500 μM adenosine. The renormalization of glutamate and aspartate uptake caused by adenosine might reasonably be expected to have potential therapeutic implications for the treatment of hepatic encephalopathy.

    Notes: Abstract The present work was carried out to study the influence of ammonia and factors from sera and cerebrospinal fluid (CSF) from patients with different degrees of chronic liver diseases on [3H]D-aspartate (Asp) and [3H]L-glutamate (Glu) high-affinity uptake into the rat hippocampal formation. For comparison, high-affinity uptake of Glu and Asp was determined in human hippocampal brain tissue obtained at autopsy from cirrhotic patients dying in hepatic coma and from control brains free from neurological, psychiatric, or hepatic diseases. Sera and CSF from patients with chronic liver failure and hepatic encephalopathy (HE) were seen to reduce dramatically Glu and Asp uptake into rat hippocampal dendritic layers. A close inverse relationship was found to exist between the level of ammonia in the sera and the inhibition of uptake, both phenomena correlating highly with the extent of liver failure. The present findings, obtained after dilution of sera from patients with HE while maintaining initial ammonium levels, elucidate, however, that ammonia alone cannot account for the reduction in Glu/Asp uptake capacity. The inhibition of Asp uptake into human hippocampal formation of patients dying in hepatic coma was even more pronounced when compared to that found in rat hippocampus incubated in sera and CSF from patients. Glu/Asp uptake into brain tissue is supposed to be an important factor in the pathogenesis of HE accompanying liver dysfunctions.

    Description / Table of Contents: Die Einwirkung von oxidierenden oder reduzierenden Gasen oder Dämpfen auf Polyacetylen, Polypyrrol, Poly(phenylacetylen) und Poly-p-phenylen führt zu Änderungen der elektrischen Leitfähigkeit. Dieser Effekt wird an Hand eines Drei-Stufen-Modells erklärt. Als erste Stufe wird die Adsorption an der Polymeroberfläche angenommen. Die zweite Stufe besteht in der Diffusion der Adsorbatmoleküle in das Innere des Polymeren, während die dritte Stufe durch eine irreversible Reaktion zwischen Adsorbatmolekülen und Polymerem gegeben ist. Diese drei Reaktionstufen sind manchmal zeitlich voneinander abgegrenzt. Während der Adsorptionsstufe ist die elektrische Leitfähigkeitsänderung reversibel, wenn durch Abpumpen das Gas oder der Dampf wieder entfernt werden.
    Notes: Exposure of polyacetylene, polypyrrole, poly(phenylacetylene), and poly-p-phenylene to oxidizing or reducing gases and vapors leads to changes of their electrical conductivity which can be explained by a three step model involving as a first step adsorption at the surface, as a second step diffusion into the bulk of the material and as a third step irreversible chemical changes due to reaction between the dopant and the polymer chain. These three regimes are sometimes well separated in time. During the adsorption regime, the conductivity change is reversible with respect to removal of the vapor atmosphere by evacuation.
