    Notes: Abstract: The in situ freezing technique has been widely used to fix labile metabolites and cellular second messengers in cerebral cortex. In this study, we isolated specific brain regions at 0°C from coronal sections of frozen heads following in situ brain freezing and measured regional concentrations of labile metabolites and cellular messengers. These levels in the cortex were compared with those in cortical punches obtained at freezing temperature (less than −40°C) from the same in situ frozen brains and those of cortex dissected from decapitated animals. In both isoflurane- and pentobarbital-anesthetized animals, we observed that the levels of lactate, free fatty acids, inositol 1,4,5-trisphosphate, and diacylglycerol, as well as the proportion of protein kinase C associated with the membrane fraction, were similar in cortical punches taken at freezing temperature and those dissected at 0°C. However, with animals decapitated at room temperature, cortical and hippocampal levels of lactate, free fatty acids, and inositol 1,4,5-trisphosphate and the proportion of membrane protein kinase C were significantly higher than those of corresponding brain regions isolated at 0°C from in situ frozen brains (p 〈 0.05). These results indicate that dissection of cortex and hippocampus at 0°C following in situ freezing will eliminate decapitation-induced production of artifacts and changes in the levels of cellular second messengers such as inositol 1,4,5-trisphosphate, diacylglycerol, and protein kinase C. The present technique, used in conjunction with in situ freezing, will fix cellular second messengers and labile metabolites in several regions of brain and may facilitate accurate characterization of molecular and cellular mechanisms underlying CNS function.

    Notes: We have studied 50 nm Si0.8Ge0.2 buried layers and 800 nm Si0.9Ge0.1 layers, grown by molecular beam epitaxy, which were implanted with 200 keV oxygen O+ ions to a dose of 1.8×1018/cm2. This implantation procedure is similar to that used in the formation of SIMOX (separation by implantation of oxygen), although it was not followed by the usual high temperature anneal. Because the initial implantation profile extends as far as 600 nm below the Si surface, the 50 nm Si0.8Ge0.2 layers were embedded in Si at depths of 600, 500, 400, and 300 nm from the Si surface. The effects of Ge incorporation could then be studied as a function of distance from the surface and position within the implantation profile. Using a scanning transmission electron microscope equipped with energy dispersive x-ray spectroscopy, we examined cross-sectional specimens for the distribution of the Ge in and around the implanted oxide layer. We found that the Ge tended to be excluded from the oxide in all the samples with the 50 nm buried layer. For the case where the oxide was implanted into the midst of an 800 nm layer, the Ge remained in the oxide.

    Notes: The Water Authorities Association guide ‘Sewers for Adoption’ has achieved considerable success in unifying practice and establishing well-understood and recognized standards for the design and construction of sewers by developers under Section 18 of the Public Health Act 1936. However, the supervision provided by the adopting authorities — particularly in relation to supervision fees — has been an area of dispute for some time.The Construction Industry Research and Information Association (CIRIA) has recently completed a study on the optimum levels of supervision for sewer construction. The study included a national survey on the nature and extent of problems occurring during the course of Section 18 work and the supervision practices presently being undertaken. The study puts forward recommendations on the nature and timing of periodic inspections undertaken during construction. Several specific problem areas are discussed, ranging from measures required to ensure that design submissions are of an adequate standard, to the timing of any pre-adoption closed-circuit television inspection. This paper draws upon the contents of the report and discusses some of the principal findings and recommendations of the study.
