    Notes: Summary Hypertension has a high prevalence among subjects with decreased insulin sensitivity and/or hyperinsulinemia. Furthermore, angiotensin II plays a pivotal role in the regulation of vascular tone and is known to induce hypertrophy and/or hyperplasia in vascular smooth muscle cells. In the present study, the effect of insulin on angiotensin II induced smooth muscle cell growth (Wistar-Kyoto rat) was investigated. Cell growth was assessed by the measurement of [3H]thymidine incorporation into cell DNA. Insulin in a concentration range of 1.7 × 10−10–1.7 × 10−6 M lacked any effect on cell DNA synthesis. However, insulin enhanced the angiotensin 11 induced DNA synthesis in a concentration-dependent manner. This effect was similar in cells with a weak and in cells with a marked response in DNA synthesis to stimulation with 100 nM angiotensin 11. In conclusion, insulin is able to enhance angiotensin 11 induced DNA synthesis and may therefore function as a growth cofactor in vascular smooth muscle cells.

    Notes: Summary Structural changes within the blood vessel wall such as hyperplasia and hypertrophy of vascular smooth muscle cells are important factors in the pathogenesis of hypertension. Humoral growth factors such as angiotensin II (AII) and platelet-derived growth factor BB (PDGF-BB) may participate in the remodelling of the blood vessel wall. Whether and by which mechanisms antihypertensive treatment is capable of influencing the structural blood vessel alterations to date remains unclear. In the present study, the effect of nifedipine and diltiazem on AII- and PDGF-BB-induced vascular smooth muscle cell proliferation was examined. Nifedipine and diltiazem at a concentration of 10 μM did not affect baseline DNA synthesis in isolated vascular smooth muscle cells in culture. AII (final concentration 100 nM) and PDGF-BB (50 ng/ml) stimulated DNA synthesis by approximately 9.0- and 4.6-fold, respectively. Both AII- and PDGF-BB-induced DNA synthesis was significantly blunted by diltiazem and nifedipine in a concentration of 10 μM, while no significant influence was seen with concentrations from 10 nM up to 1 μM. In contrast, no significant influence of these drugs could be observed on fetal calf serum 5%-induced DNA synthesis. The findings indicate that calcium antagonists possess antimitogenic potential and that they may thus contribute to the regression of structural changes of the blood vessels associated with hypertension.

    Notes: Thick-walled components subjected to pulsating internal pressure are widely applied in high-pressure technology and in manufacturing processes such as fluid-jet cutting and high-pressure cleaning, mainly in conjunction with reciprocating pumps. Corrosive fluids require high-strength and tough chrome-nickel steels with soft martensitic or semi-austenitic structure. This contribution reports on the fatigue of thick-walled plain and cross-bored pipes made from high alloy chrome-nickel steels such as X5 CrNiMoCu 21 8 and X5 CrNiMo 16 5. The speciments, uniaxial standard form and thick-walled pipes, were cut from forged blocks in the three axial directions. For loading with pulsating pressure, a suitable, high-frequency piston pulsation machine has been developed. The fatigue tests on pipe specimens show typical Woehler characteristics with only slight scatter and relatively good isotropy. The surprisingly large admissible pulsating pressure can be explained for the applied steels by dynamic generation of residual stresses as a result of shake-down effects. Presentation in a Smith digram explains the occuring dynamic shake-down and its favourable results in comparison to the more brittle highly tensile steels. It also reveals that heat treatment to higher tensile strength does not always yield an increase in the admissible pulsating pressure. It will be shown that static autofretting and shake-down affect the fatigue strength of thick-walled pipe specimens in the same way. Tests with internal liners in the tube specimens provide indications on the sensitivity of material failures towards fluids. The investigation aids the understanding of the fatigue behaviour and the design of components made of modern high-strength corrosion resistant steels.

Size histograms are taken from transmission electron micrographs. Temporal changes of activity and selectivity for the hydroxylation of phenol and anisole are monitored. It is postulated that the p/o product ratios are due to the greater chemisorption of the ortho molecule.

    Notes: Eighty one polychlorinated biphenyls (PCBs) and nineteen chlorinated pesticide standards have been analyzed on a newly developed apolar high resolution gas chromatography (HRGC) column. Emphasis was placed on the determination of PCB indicator congeners which are part of national regulations, and of toxic coplanar PCB congeners.The new column enables almost unambiguous quantification of the PCB indicator congeners (PCB 138 can be separated from both PCB 163 and PCB 164; PCB 28 and PCB 31 were also separated and no coelution was observed for PCB 52, PCB 118, and PCB 180). The new column furnished better results than CP-Sil 8 for the analysis of PCB indicator congeners in a sample of seal blubber.
