    Notes: Abstract Stone samples from corroding historical buildings in Germany released NO in significant amounts during oxidation of ammonium to nitrate. Nitrite intermediately accumulated to low concentrations. Nitrification and NO release were detectable only in stones with neutral pH. Addition of nitrapyrin inhibited both ammonium oxidation and NO production. NO was produced in stone samples containing Nitrosomonas or Nitrosovibrio, but was not produced in samples containing only Nitrobacter. Addition of nitrite stimulated NO production in autoclaved stone samples significantly only if dried to 〈 2% water content, or in acidic stone samples. In moist neutral stones, NO production was only slightly stimulated by nitrite giving less than about 8% of the NO production observed under nitrifying conditions. Most of the NO production by corroding building stones was apparently due to ammonium-oxidizing nitrifiers.

    Notes: Abstract The generation of ATP and NADH in total cells of Nitrobacter was measured under aerobic and anaerobic conditions. NADH synthesis was driven by an ATP independent reaction with nitrite or nitric oxide as electron donors. The rate of NADH formation was about 200 times higher, if nitric oxide instead of nitrite served as electron donor. Approximately 2 mol nitric oxide were needed for reduction of 1 mol NAD+. Nitrite caused an end-product inhibition of the nitric oxide induced NADH synthesis. ATP was synthesized by NADH oxidation with oxygen and nitrate as terminal electron acceptors.

    Notes: Abstract Heterologous DNA hybridization using a ribulose- 1,5-biphosphate carboxylase/oxygenase (RuBisCOase) large subunit gene (rbcL) probe from Anacystis nidulans revealed the presence of two rbcL in Nitrobacter hamburgensis. One gene is located on a plasmid, the other on the chromosome. The genes appear to be very similar since both hybridized strongly to the A. nidulans probe. However, restriction endonuclease digestions revealed differences.Two different RuBisCOase enzymes were isolated from N. hamburgensis. The Mr of the native enzymes were 520 000 and 480 000. Sodium dodecyl sulfate-polycrylamide gel electrophoresis (SDS-PAGE) revealed the presence of both LSU and small subunits (SSU) for both enzymes. The Mr were 53 000 and 16 000, and 49 000 and 13 500, respectively. A hexadecameric structure is suggested for both enzymes.

    Notes: Abstract Nitrobacter hamburgensis was shown to synthesize at least two distinct membrane-bound b-type cytochromes. One of these, a minor component detected during nitrite oxidation, was also found in the obligately autotrophic species Nitrobacter winogradskyi. During heterotrophic growth of N. hamburgensis a second (major) cytochrome b was detected, which we assume functions as an alternative terminal oxidase.

    Notes: Summary Sulphuric-acid-producing thiobacilli cause severe corrosion of concrete walls in sewage pipelines. The bacteria excrete sulphuric acid, which degrades carbonaceous binding material. Nitrifying organisms have been found in high cell numbers on historical sandstone buildings as well as on concrete buildings. The nitric acid, which is excreted by these organisms as metabolic end-product, also causes severe corrosion. The microorganisms are able to metabolize inorganic substances like ammonia and NO originating from air pollution. The natural process was remodelled by means of simulation experiments. It became evident that mineral-acid-producing bacteria play an important role in biodeterioration of building materials.

    Notes: Summary The distribution of the neural cell adhesion molecule (NCAM) and Thy-1 in the olfactory mucosa and olfactory bulb, the eye and the inner ear was examined with immunocytochemistry in mouse embryos from embryonic day 12 (E 12) to embryonic day 19 (E 19). In general, neurons are completely outlined with NCAM, whereas Thy-1 outlines only dendrites and axons. A variable cytoplasmic staining for Thy-1 is present in the perikarya. Neurons directly associated with special sense organs express NCAM and Thy-1 already from the earliest stage and throughout the period investigated, apart from the olfactory neurons in which Thy-1 disappears at E 19. The mitral cells in the olfactory bulb show Thy-1 but no NCAM reactivity. In the eye, lens fibers express Thy-1 and the pigmented layer expresses NCAM; neither of the two molecules can be detected at E 19. In the inner ear, hair cells express NCAM at E 19. Based on the distribution during the developmental period studied and on the cellular localisation of reaction products, it is suggested that the NCAM adhesion function could be of a more general nature by keeping appropriate cell membranes in close contact and thereby allowing more specific molecular interactions to take place. Thy-1, which is located on dendrites and axons, could be such a specific factor and function as recognition molecule in the developing nervous system.

    Notes: Summary Calmodulin is a regulator of several calcium-dependent cellular processes. It has been suggested that it plays a role in the mechanism of secretion. Employing an indirect immunoperoxidase technique at the light microscope level, this study demonstrates the presence of calmodulin in several exocytotic cells (mast cells, thyroid follicular cells, neurohypophyseal neurosecretory terminals, pancreaticβ-cells and pancreatic acinus cells) in rat and man. The positive staining reaction for calmodulin was granular and at least in the case of rat mast cells it appeared to be associated with the granule membrane.

    Notes: Abstract Malignant cell lines obtained by ethyl nitrosourea (EtNU)-induced transformation of fetal rat brain cells in culture express protein markers of different types of neural cells. These are the nervous system-characteristic S-100 protein; glial fibrillary acidic protein (GFAP); neuron-specific-enolase (NSE), and the D2-cell adhesion molecule. S-100 protein was absent in fetal brain cells in culture, but gradually appeared in the later stages of malignant transformation and further increased at onset of rapid growth of atypical cells (stage IV). GFAP and D2 were weakly expressed in primary fetal brain cells and did not change throughout malignant transformation. NSE was present in both normal and carcinogen-treated fetal brain cells, and increased at later stages of malignant transformation. From stage III (40–100 days) some cultures were strongly positive and some negative, and the same was seen in the resulting tumourigenic cells about 100 days later. In conclusion the stepwise process of malignant transformation of brain cells in culture ended with a stable phenotype of cells capable of expressing varying types of diffentation markers. The presence of these markers in rat brain cells undergoing malignant transformation may indicate that EtNU given at 18th days of gestation is acting on multipotent neuroectodermal cells.
