    Notes: Abstract. Objective: To investigate whether interleukin-13 (IL-13) can affect arachidonic acid metabolism and phagocytic activity of normal human polymorphonuclear neutrophils (PMN).¶Methods: Normal human PMN (1 × 106 cells/ml) were incubated with different concentrations of IL-13 (0.1–10 ng/ml) for a variety of times (30–120 min). Phagocytosis and intracellular cyclooxygenase-2 (COX-2) were detected by flow cytometry. The expression of COX-1 and COX-2 mRNA was detected by RT-PCR. The concentration of PGE2 in the PMN cultured supernatants was determined by EIA.¶Results: We found that IL-13 at an optimal concentration of 1 ng/ml significantly enhanced COX-2 gene expression and PGE2 production (121.57 ± 22.17 pg/ml in IL-13 stimulation vs. 73.16 ± 11.72 pg/ml in controls) by PMN. In addition, IL-13 stimulated PMN phagocytosis via increased complement receptor type 1 (CR1) and type 3 (CR3), but not IgG Fcγ receptor type 3 (FcγRIII). The cytoplasmic neutral esterase activity of PMN was also enhanced by IL-13 stimulation for 24 h.¶Conclusions: These results suggest that IL-13 can stimulate PMN and modulates the inflammatory reactions via the cyclooxygenase pathway.

    Notes: Abstract  Serine proteases are the most abundant granule constituents of several major hematopoietic cell lineages. Due to their high abundance and their strict tissue specificity they have become important phenotypic cell markers used for studies of various aspects of hematopietic cell development. Using a polymerase chain reaction (PCR)-based strategy for the isolation of trypsin-related serine proteases, we were able to isolate cDNAs for two of the major neutrophil and monocyte serine proteases in the mouse, cathepsin G and mouse protease 3 (myeloblastin). The internal PCR fragments were used as probes to screen a mouse mast cell cDNA library and a cDNA library originating from a mouse monocytic cell line (WEHI-274.1). Full-length cDNAs for mouse cathepsin G and proteinase 3 were isolated and their complete sequences were determined. Northern blot analysis revealed expression of cathepsin G in immature cells of the monocyte macrophage lineage but also in the connective tissue mast cell line MTC. Proteinase 3 was expressed in several cell lines of myelo-monocytic origin and in one B-cell line, but not in any of the other cell lines tested. The isolation of cDNAs for mouse cathepsin G and mouse proteinase 3, together with the previous characterization of the gene for mouse N-elastase, and the entire or partial amino acid sequences for porcine azurocidine, equine N-elastase and proteinase 3, rat, dog, and rabbit cathepsin Gs in evolutionary relatively distantly related mammalian species, indicates that these four members of the serine protease family have been maintained for more than 100 million years of mammalian evolution. This latter finding indicates a strong evolutionary pressure to maintain specific immune functions associated with these neutrophil and monocyte proteases. All amino acid positions of major importance for the cleavage site selection have also been fully conserved between mouse and human proteinase 3 and a few minor changes have occurred between mouse and human cathepsin G.

    Notes: Abstract Simon poplar (Populus simonii) protoplasts were isolated from suspension cells, with protoplast yield of 3.8×107 g−1 F. W. They were cultured in a K8P liquid medium containing 13.57μM 2,4-D, 1.07μM NAA and 0.93 μM KT. Protoplast culture was influenced by the plating density, osmotic pressure, and the sources and amounts of nitrogen and carbon in the culture medium. Multiple shoots were produced from protoplast-derived callus after culture on MS medium containing 4.44 μM BA, 2.32μM KT, 2.28 μM ZT, and 0.54μM NAA. Shoots 2–3 cm in height were isolated from the calli and rooted on 1/2 MS medium. After transplantation into pots, the regenerated plants grew vigorously in greenhouse.

    Notes: Abstract. Respiratory syncytial virus (RSV) is a common cause of lower respiratory infections in infants. Cardiac arrhythmias are an uncommon complication in infants with RSV bronchiolitis. Presumably, the arrhythmias may be secondary to RSV pericarditis, myocarditis, or some other infection of the heart. We report on a patient with RSV bronchiolitis who had clinical myocarditis complicated by ventricular tachycardia.

    Notes: Abstract In this study a restored mandibular molar with different Class II amalgam preparations was examined to analyze the potential for restoration failure attributed to cyclic fatigue crack growth. A finite element analysis was used to determine the stress distribution along the cavo-surface margin which results from occlusal loading of each restoration. The cyclic crack growth rate of sub-surface flaws located along the dentinal cavo-surface margin were determined utilizing the Paris law. Based on similarities in material properties and lack of fatigue property data for dental biomaterials, the cyclic fatigue crack growth parameters for engineering ceramics were used to approximate the crack growth behavior. It was found that flaws located within the dentine along the buccal and lingual margins can significantly reduce the fatigue life of restored teeth. Sub-surface cracks as short as 25 μm were found capable of promoting tooth fracture well within 25 years from the time of restoration. Furthermore, cracks longer than 100 μm reduced the fatigue life to less than 5 years. Consequently, sub-surface cracks introduced during cavity preparation with conventional dental burrs may serve as a principal source for premature restoration failure. ©©1999©Kluwer Academic Publishers

    Notes: Summary This study presents the numerical predictions of the fluid flow and heat transfer characteristics for a backward-facing step by discharging a jet perpendicularly to the main flow. The turbulent governing equations are solved by a control-volume-based finite-difference method with power-law scheme and the well known K−ε model and its associate wall function to describe the turbulent behavior. The interesting parameters include entrance Reynolds number (Re h ), dimensionless jet location (X j/H) and the ratio of jet velocity (U j/U0) with channel expansion ratio ER=1.67. The predicted attachment point is in good agreement with the experiment. It is found that the optimum position is atX j/H=2.1 (X j: location of jet measured from the step position, H: the step height).

    Notes: Summary We hypothesize that the morphology of the neuromuscular junction on different muscle fibre types varies, reflecting differences in activation history. In the rat diaphragm muscle, we used a three-colour fluorescent immunocytochemical technique to simultaneously visualize (1) innervating axons and presynaptic nerve terminals, (2) motor endplates and (3) myosin heavy chain isoform expression (muscle fibre type). Laser-scanning confocal microscopy was then used to optically section the triple-labelled muscle fibres, and create three-dimensional views of the neuromuscular junction. Type I fibres were innervated by the smallest axons, and type IIa, IIx and IIb fibres by progressively larger axons. Absolute planar areas of nerve terminals and endplates progressively increased from type I, IIa, IIx to IIb fibres. When normalized for fibre diameter planar areas of nerve terminals were largest on type I fibres, with no difference among type II fibres. The normalized planar area of endplates were larger for type I and IIb fibres, compared to type IIa and IIx fibres. The three-dimensional surface area of endplates was largest on type I fibres, with no differences across type II fibres. When normalized for fibre diameter, endplate surface areas increase progressively from type I, IIa, IIx to IIb fibres. The branches increased progressively from type I, IIa, IIx to IIb fibres. Conversely, individual branch length was longest on type I fibres, and shortest on type IIb fibres. The extent of overlap of pre- and postsynaptic elements of the neuromuscular junction decreased progressively on type I, IIa, IIx and IIb fibres. We conclude that these morphological differences at the neuromuscular function of different fibre types reflect differences in activation history and may underlie phenotypic differences in neuromuscular transmission.

    Notes: Abstract We have analysed the YJR043c gene of Saccharomyces cerevisiae, previously identified by systematic sequencing. The deletion mutant (yjr043cΔ) shows slow growth at low temperature (15° C), while at 30° C and 37° C the growth rate of mutant cells is only moderately affected. At permissive and nonpermissive temperatures, mutant cells were larger and showed a high proportion of large-budded cells with a single duplicated nucleus at or beyond the bud neck and a short spindle. This phenotype was even more striking at low temperature, the mutant cells becoming dumbbell shaped. All these phenotypes suggest a role for YJR043C in cell cycle progression in G2/M phase. In two-hybrid assays, the YJR043c gene product specifically interacted with Poll, the catalytic subunit of DNA polymerase α. The pol1-1 /yjr043cΔ double mutant showed a more severe growth defect than the pol1-1 single mutant at permissive temperature. Centromeric plasmid loss rate elevated in yjr043cΔ. Analysis of the sequence upstream of the YJR043c ORF revealed the presence of an MluI motif (ACGCGT), a sequence associated with many genes involved in DNA replication in budding yeast. The cell cycle phenotype of the yjr043cΔ mutant, the evidence for genetic interaction with Pol1, the presence of an MluI motif upstream and the elevated rate of CEN plasmid loss in mutants all support a function for YJR043C in DNA replication.

    Notes: Abstract Cholesteric liquid crystalline triheptyl cellulose (THC)/ethyl cellulose binary blend membranes were prepared and the effect of pressure on their oxygen enrichment at elevated temperature was investigated using a constant pressure-variable volume method. The oxygen enrichment increased with the increase of transmembrane pressure difference or with slight increase of the THC content in the blend membranes. The oxygen concentration through the membranes increased linearly with decreasing pressure ratio. Air was directly separated through a 17 μm-thick THC/EC(1.5/98.5) membrane to prepare an oxygen-enriched air containing 39.5% oxygen at the flux of 6.99×10−4 cm3 (STP)/s.cm2 at the pressure difference of 0.43 MPa and 85 °C.
