    Notes: Summary We studied insulin receptor kinase activation in two brothers with congenital muscle fibre type disproportion myopathy and compound heterozygous mutations of the insulin receptor gene, their parents, and their unaffected brother. In the father who has a heterozygote Arg1174→Gln mutation, in situ activation of the receptor kinase in skeletal muscle was reduced about 70 %. Selection of only those receptors that bound to anti-phosphotyrosine antibody showed that these receptors had normal kinase activity and that the reduction in overall kinase activity was due to the inability of about 70 % of the receptors to become insulin-dependently activated. The mother carries a point mutation at the last base pair in exon 17 which, due to abnormal alternative splicing, could lead to normally transcribed receptor or truncated receptor lacking the kinase region. Kinase activation was normal in the mother's skeletal muscle, suggesting that virtually no truncated receptor was expressed. Receptor kinase activity was, however, reduced by 95 and 91 % in the compound heterozygous brothers. This suggests that the mother's mutated allele contributes little to the generation of functional receptor protein and that the receptors in the mother's skeletal muscle are transcribed almost exclusively from the non-mutated allele. The mutation in exon 17 could lead to reduced transcription or rapid degradation of a predominantly transcribed truncated gene product or both. [Diabetologia (1999) 42: 245–249]

    Notes: Abstract  Cordierite powder was synthesized via different sol-gel procedures. The use of 29Si NMR spectra of all stages beginning from tetraethoxysilane (TEOS) up to final ceramic powder material is discussed. In the early stages of the sol-gel process all oligomeric structure units can be identified. Spectra of the higher condensed sols only allow the quantification of Qn-groups. MAS spectra of gels heated up to 750 °C show no fine structure to be evaluated. Spectra of samples heated to 1250 °C indicate a sluggish disorder-order transformation from hexagonal α-cordierite to orthorhombic cordierite.

    Notes: Abstract The culture fluorescence of Alcaligenes eutrophus JMP 134 was determined on-line by an Ingold Fluorosensor™ and correlated to the intracellular concentrations of reduced nicotinamide adenine dinucleotide (phosphate). The data were obtained from aerobic cultures of the strain growing chemostatically on phenol, phenol+sodium formate and fructose, as well as from aerobic/anaerobic transitions and substrate pulse experiments. The total culture fluorescence was corrected to take into account the inner filter effect of cells. Upon analysing the intracellular concentration of the dinucleotides using HPLC, it became evident that both NADH and NADPH contribute significantly to the fluorescence signal. A linear relationship between the sum of NAD(P)H and the net culture fluorescence was obtained from these data with a correlation factor of r=0.82. These investigations indicate that the measurement of culture fluorescence is a practicable tool for monitoring the redox state of a cellular culture, provided the total fluorescence signal is adjusted and the investigations are supported by direct measurements of intracellular levels of reduced dinucleotides.

    Notes: Abstract The extent to which the diagnostic performance of low-dosed storage phosphors can be improved by suitable image postprocessing was investigated. Storage phosphors radiographs and conventional 200- and 400-speed screen-film combinations of an antropomorphic chest phantom with simulated nodules and linear structures were compared. The detection of test bodies was determined in an ROC analysis based on 16 200 individual observations. The two analog screen-film systems did not differ significantly. As far as the storage phosphor radiographs are concerned, a higher diagnostic performance was achieved with a large filter kernel (S 40), whereas small filter kernels and edge-enhanced imaging modes reduced the detection of detail. The image quality of low-dosed storage phosphors is thus similar to high-amplification screen-film combinations if large filter kernels are used in postprocessing of the image.

    Notes: Abstract. The aim of this work was to examine the influence of the filter kernel size on the detectability of differing radiological findings in interstitial lung disease. In 97 patients with confirmed pulmonary fibrosis chest radiographs were obtained with a film-screen system of speed class 200 and with correspondingly exposed storage phosphorous plates. The size of the filter kernel used for the image postprocessing varied between sigma 5 and sigma 70. The detectability of interstitial lung changes was evaluated independently by eight readers on the basis of a defined rating system. The results were analysed using multifactorial analysis of variance with Scheffé test at a significance level of p = 0.05. Small kernel sizes (S 5, S 10) combined with high edge enhancement were only of benefit in the imaging of septal lines, but reduced the detectability of nodular and reticular structures. Good detail detectability of both micronodules and septal lines was obtained with a medium kernel size of sigma 40. Storage phosphor radiography utilizing the appropriate choice of postprocessing parameters provides equivalent image quality for evaluating interstitial lung changes compared with a modern film-screen technique.

    Notes: Abstract Objective: The therapeutic response to PGE1 is highly variable, and a contribution by variable formation of its active tertiary metabolite PGE0 is in question. Hence, the objective of this study was to assess the person-to-person variation of the reduction of the inactive intermediate metabolite 15-KD PGE1 by human liver and human erythrocytes in forming the active metabolite PGE0. Methods: Source of enzyme was lysed erythrocytes from 29 donors, and a bank of 37 donor livers including specimens from 15 children. Tritium-labelled 13,14-dihydro-15-keto-prostaglandin E1 (15-KD PGE1) was used at low nanomolar concentrations and found to be converted almost exclusively to the more polar compound 13,14-dihydro-prostaglandin E1 (PGE0) by an NADPH-dependent carbonyl reductase. The identity of the product PGE0 was established by comparison of its chromatographic and mass spectral characteristics with authentic PGE0. Results: Lysed erythrocytes had readily measurable enzymatic activity; differences between the preparations from 29 subjects were very small with only a twofold range of variation. In contrast to lysed erythrocytes, intact erythrocytes did not catalyse the reaction so that the erythrocyte activity should be medically immaterial. 15-KD PGE1 15-ketoreductase activity of liver cytosol averaged 61.1 fmol · min−1 · mg−1 protein in preparations from 37 human livers. Individual activities varied over an almost tenfold range, with indications of a non-normal distribution. Kinetic studies of selected specimens showed substantially different Vmax values but indistinguishable k M values, suggesting that the individual variation in 15-KD PGE1 15-ketoreduction is the result of differences in enzyme concentration rather than of structural enzyme variations. The activity in 15 livers from children was significantly lower than in those from adults. Inhibition data suggest that both the liver and the erythrocyte enzymes belong to the class of carbonyl reductases. Conclusions: The variations in hepatic enzyme activity may be expected to affect the transformation of 15-KD PGE1 to the active metabolite PGE0 in vivo. The clinical significance remains to be explored.

    Notes: Abstract Severe chemical ozone loss has been detected in the Arctic in the winter and spring of 1995–96 by a variety of methods. Extreme reductions in column ozone due to halogen catalysed chemistry were derived from measurements of the Halogen Occultation Experiment (HALOE) on board the Upper Atmosphere Research Satellite in the Arctic vortex. Here, we discuss further aspects of the HALOE observations in the Arctic over this period. Potential problems, both in the data themselves and in the methodology of the data analysis are considered and the reason for the differences between the Arctic ozone losses deduced from HALOE data version 17 and 18 is analysed. Moreover, it is shown that HALOE measurements in the Arctic in winter and spring 1995–96 compare well with observations by other ground-based and satellite instruments.
