    Notes: Abstract: De novo l-DOPA biosynthesis was studied in stably transfected AtT-20 cells expressing wild-type- or [Leu40]-recombinant tyrosine hydroxylase (rTH). Basal rates of DOPA accumulation were much higher by cells expressing rTH in which Leu was substituted for Ser40 (S40L-rTH) than by those expressing wild-type rTH (WT-rTH). Treatment of WT-rTH cells with forskolin produced an increase in DOPA accumulation and a concomitant increase in WT-rTH phospho-Ser40 content, whereas DOPA production by cells expressing S40L-rTH was entirely unaffected by forskolin. After forskolin treatment of 32Pi-prelabeled cells, WT-rTH was phosphorylated at Ser8, Ser19, Ser31, and Ser40, whereas 32P incorporation into S40L-rTH was restricted to Ser8, Ser19, and Ser31. Relatively prolonged treatment of AtT-20 cells expressing WT-rTH with either a depolarizing agent (elevated potassium) or a phosphatase inhibitor (okadaic acid) increased DOPA production and increased the phosphorylation state of Ser40; but, unlike forskolin, these treatments also increased DOPA production by cells expressing S40L-rTH. Thus, the present studies demonstrate that Ser40 phosphorylation mediates forskolin-induced increases in DOPA biosynthesis directly but that mechanisms other than Ser40 phosphorylation can mediate the increases in DOPA biosynthesis produced either by depolarization or by protein phosphatase inhibition.

    Notes: Abstract: A synthetic peptide corresponding to residues 32–47 of rat tyrosine hydroxylase (TH) was phosphorylated by protein kinase A at Ser40 and used to generate antibodies in rabbits. Reactivity of the anti-pTH32–47 antibodies with phospho- and dephospho-Ser40 forms of TH protein and peptide TH32–47 was compared with reactivity of antibodies to nonphosphorylated peptide and to native TH protein. In antibody-capture ELISAs, anti-pTH32–47 was more reactive with the phospho-TH than with the dephospho-TH forms. Conversely, antibodies against the nonphosphorylated peptide reacted preferentially with the dephospho-TH forms. In western blots, labeling of the ∼60-kDa TH band by anti-pTH32–47 was readily detectable in lanes containing protein kinase A-phosphorylated native TH at 10–100 ng/lane. In blots of supernatants prepared from striatal synaptosomes, addition of a phosphatase inhibitor was necessary to discern labeling of the TH band with anti-pTH32–47. Similarly, anti-pTH32–47 failed to immunoprecipitate TH activity from supernatants prepared from untreated tissues, whereas prior treatment with either 8-bromoadenosine 3′,5′-cyclic monophosphate or forskolin enabled removal of TH activity by anti-pTH32–47. Lastly, in immunohistochemical studies, anti-pTH32–47 selectively labeled catecholaminergic cells in tissue sections from perfusion-fixed rat brain.

    Notes: Fabrication and characterization of YBa2Cu3Oy Josephson junctions with step-edge, bi-epitaxial, and sandwich geometries are performed in this study to investigate the effects of microwave irradiation on the I–V characteristics. I–V curves under microwave irradiation reveal Shapiro steps. The width of the constant voltage steps were modulated by the microwave power. The period of the magnetic induction ΔB detected from the V–Φ curves for superconducting quantum interference devices is temperature dependent. Additionally, Tc and Ic of the proximity sandwich junctions are enhanced by the microwave irradiation. © 1995 American Institute of Physics.

    Notes: The sidegating effect on the Schottky barrier in ion-implanted GaAs was investigated with capacitance-voltage profiling at various negative substrate voltages. It was demonstrated that the negative substrate voltage modulates the Schottky depletion region width as well as the space charge region at the substrate-active channel interface. © 1995 American Institute of Physics.

    Notes: Densely sintered Mn–Zn ferrites with grain size varying between 1 and 6 μm were obtained. Then the effect of grain size was investigated in detail by power loss analysis and scanning electron microscopy examination of microstructure. It is found that the power loss is seen to be lowered from 260 to 120 kW/m3 at 500 kHz, 50 m T and 90 °C with grain size decreased in the order of 3 μm. It is assumed that the residual loss from domain wall resonance is a dominant factor of power loss from 500 kHz to 1 MHz. Therefore we conclude that Mn–Zn ferrite display a grain size dependent power loss with transition at grain size about 3 μm due to the absence of intragranular domain wall and these fine core materials can be prepared by low sintering temperature and the optimum control of atmosphere containing 0.5% O2. © 1996 American Institute of Physics.

    Notes: A new technique for Monte Carlo sampling of the hard-sphere collision force has been applied to study the interaction between a pair of spherical macroions in primitive-model electrolyte solutions with valences 1:2, 2:1, and 2:2. Macroions of the same charge can attract each other in the presence of divalent counterions, in analogy with earlier observations for planar and cylindrical geometries. The attraction is most significant at intermediate counterion concentrations. In contrast to the entropic depletion force between neutral particles, attraction between macroions is of energetic origin. The entropic contribution to the potential of mean force is generally repulsive at conditions corresponding to aqueous colloids with or without salt. For systems with divalent counterions, the potentials of mean force predicted by mean-field approximations like the Derjaguin–Landau–Verwey–Overbeek (DLVO) theory or the Sogami–Ise (SI) theory are qualitatively different from those observed in the simulations. However, for systems with monovalent counterions, predictions of DLVO theory are in fair agreement with simulation results. © 1999 American Institute of Physics.

    Notes: Bent triangular Si crystals with an oblique-cut angle 12.16° are used on the beamline 4W1C at Beijing Synchrotron Radiation Facility, to select monochromatic x-rays and to focus the beam in the horizontal direction. The downstream Pt-coated reflective mirror is bent as a cylindroid focuses the beam vertically. Double-focused monochromatic x rays are available on the diffuse scattering station. Specifications of the beamline with the Si (220) monochromator were measured as follows: size of the focus spot—0.5 mm (horizontal)×0.3 mm (vertical), maximum x-ray density—1.6×109 photons/s mm2, and energy resolution of the monochromatic beam—4.4×10−4. © 1995 American Institute of Physics.

    Notes: Micrometer devices of Co85Cr15 film with Hall bar shape were made using standard electron beam lithography and lift-off process. The film was thermally deposited without bias field and the sample was kept at room temperature during evaporation. The Hall voltage, measured at room temperature, showed square hysteresis loops with an external magnetic field applied in the direction close to the film plane. Based on the variation of the Hall voltage measured under different directions of external magnetic field, the magnetic easy axis was verified to be along the long axis of the device. The formation of the easy axis along the narrow wire due to the physical confinement as an intrinsic mechanism is discussed. © 1999 American Institute of Physics.

    Notes: A series of polycrystalline bulk (La1−xSmx)2/3Sr1/3MnO3 samples over a wide composition range (x=0,0.1,0.33,...,1) was prepared by solid-phase reaction sintering technique. The x-ray diffraction pattern indicates that all the samples have an orthorhombically distorted perovskite structure. A large linear magnetostriction with a value up to −180×10−6 was observed at the temperature of 77 K under the maximum magnetic field of 1.5 T. The linear magnetostriction and magnetoresistance effect of our samples has similar dependence on the Sm content x. At room temperature, the sample with a Curie temperature of 320 K, and a Sm content of x=0.33, shows both maximum linear magnetostriction value of −40×10−6 and a maximum magnetoresistance value of −12%. © 1999 American Institute of Physics.
