    Notes: The Sc+NO→ScO+N reaction has been investigated in a beam-gas arrangement, with characterization of ScO products by cw laser-induced fluorescence: absorption versus laser frequency over the A 2Π(v′=1)–X 2Σ+(v″=0) band and fluorescence over the A 2Π(v′=1)–X 2Σ+(v″=1) one. It leads to the direct determination of the nascent rotational state distribution in the X 2Σ+(v″=0) level of ScO. This distribution is close to a Prior statistical one, with a well-characterized weak "surprisal," indicating that a momentum constraint takes place during the reaction process. In the frame of this statistical distribution, a new accurate value for the dissociation energy of ScO is proposed: D00(ScO)=(6.92±0.01) eV. Spectroscopic data are reported for the A 2Π(v′=1)–X 2Σ+(v=0) band, up to N=98. © 2001 American Institute of Physics.

    Notes: An accurate potential curve for the bound and quasibound region of the B 1Πu state of 7Li2 is produced by analyzing high resolution sub-Doppler B 1Πu←X 1Σg+ excitation spectra in conjunction with lower resolution data of Hessel and Vidal [J. Chem. Phys. 70, 4439 (1979)] and of Russier et al. [J. Mol. Spectrosc. 168, 39 (1994)]. The bound and quasibound part of the curve is generated by the direct fit of molecular energies to a numerical potential; the outermost (repulsive) part of the curve is calculated by an asymptotic method using Coulombic and exchange parameters determined from the 1 1Πg and A 1Σu+ states of the lithium dimer. The full potential energy curve of the B 1Πu state reproduces all measured energies of f parity, and the widths of the predissociated lines, to within the experimental accuracy. © 2001 American Institute of Physics.

    Notes: Abstract Restenosis after initially successful balloon angioplasty of coronary artery stenosis remains a major problem in clinical cardiology. Previous studies have identified pathogenetic factors which trigger cell proliferation and vascular remodeling ultimately leading to restenosis. Since there is evidence that endothelial cells adjacent to the angioplasty wound area synthesize factors which may initiate this process, we investigated the effects of mechanical stimulation on endothelial gene expression in vitro and focussed on the influence of sustained mechanical stress on expression of immediate early genes which have previously been shown to be induced in the vascular wall in vivo. Primary cultured human umbilical vein endothelial cells (HUVEC) and the human endothelial cell line EA.hy 926 were plated on collagen-coated silicone membranes and subjected to constant longitudinal stress of approximately 20% for 10 min to 6 h. Total RNA was isolated and the expression of the immediate early genes c-Fos and Egr-1 was studied by Northern blot analysis. We found a rapid upregulation c-Fos and Egr-1 mRNA which started at 10 min and reached its maxima at 30 min. HUVEC lost most of their stretch response after the third passage whereas immediate early gene expression was constantly in EA.hy 926 cells. Using specific inhibitors we investigated the contribution of several signal transduction pathways to stretch-activated Egr-1 mRNA expression. We found significant suppression of stretch-induced Egr-1 mRNA expression by protein kinase C (PKC) inhibition (p 〈 0.05) and by calcium depletion (EA.hy926, p 〈 0. 05; HUVEC, p = 0.063). No effect on stretch-activated Egr-1 mRNA expression was detected by inhibition of protein kinase A, blockade of stretch-activated cation channels or inhibition of microtubule synthesis. We conclude that sustained mechanical strain induces Egr-1 mRNA expression by PKC- and calcium-dependent mechanisms.

    Notes: Abstract Microstructured silicon wafers were employed as miniaturizedsolid-phase reaction vessels as well as miniaturized microtiter plates. Employing piezoelectric drop-on-demand liquidjets, a combinatorial library of 256 Peptides was synthesizedon single beads. The synthesis protocol was associated to thelocation in the silicon nano-well arrangement. Products werephotolytically cleaved in the same well that was used forsynthesis and subsequently interrogated for thrombin inhibitionin a homogeneous competition assay. The assay procedure wasbased on drop-on-demand liquid delivery and laser inducedfluorescence imaging. The novel format proved useful for theintegration of both synthesis and screening into one platform,a prerequisite for an iterative, evolutionary approach towardsdrug discovery.

    Notes: Abstract To determine regional stress and strain distributions in rabbit ventricular myocardium, an anatomically detailed finite element model was used to solve the equations of stress equilibrium during passive filling of the left ventricle. Computations were conducted on a scalable parallel processing computer and performance was found to scale well with the number of processors used, so that stimulations previously requiring approximately 60 min were completed in just over 5 min. Epicardial strains from the model analysis showed good agreement (RMSE=0.007332) with experimental measurements when material properties were chosen such that cross fiber strain was more heterogeneous than fiber strain, which is also consistent with experimental observations in other species. © 2000 Biomedical Engineering Society.
