Abstract: This study investigated the effects of fiber posts, silanization, and luting agents on the interfacial strength to root dentin and composite cores. Root canals of 120 crownless human teeth were instrumented. Three different posts (opaque and translucent), with and without silane treatment, were bonded using etch‐and‐rinse, self‐etch, and self‐adhesive luting agents. The restored roots were built up with dual‐curing composite. After storage in water for 24 h at 37°C, 2‐mm‐thick slices were cut from each sample: one from the composite core and one from the restored root. Interfacial push‐out bond strengths of the posts were determined in a universal testing machine. Failure modes were analyzed using scanning electron microscopy. The post type and the luting agent had significant effects on both the post‐to‐dentin and post‐to‐core strengths. Silanization did not significantly influence post‐to‐dentin strengths, but enhanced post‐to‐core strengths. With etch‐and‐rinse luting agents, debonding occurred predominantly between the post and the cement, while the self‐etch and self‐adhesive luting agents showed more failures on root dentin. No failures occurred between the composite core and the cement. The combination of translucent posts and etch‐and‐rinse dual‐curing luting agents can positively influence the retention of fiber posts in root canals. Silanization seems to be less relevant for intra‐root canal bonding, but may have beneficial effects on post‐to‐core strengths.

Abstract: We investigate upon the change of an asteroid orbit caused by an impact. We find that, given the assumption of two dimensional motion, the asteroid displacement may be described by an analytic and explicit expression that is the vectorial sum of a radial component and a component along the asteroid velocity. The new formulation bridges the gap between the study of short-term effects, using numerical methods and the analytic study of secular changes of the asteroid orbit. The relation of the method to the established formulations is described and the known results are derived as limiting cases. The application of the new method for the performance evaluation of an asteroid deflection demonstration mission is illustrated. In such a mission the measurement of the change of the asteroid orbit by an impact will be conducted by radio-ranging to a spacecraft orbiting the deflected asteroid. Hence the measurement will primarily be sensitive to the deflection projected onto the Earth-asteroid line of sight. We discuss how the new formulation of the deflection can conveniently be employed for the estimation of the measurement accuracy and the optimal planning of a deflection demonstration mission.

Abstract: Dysfunction of mitochondrial complex I is associated with a wide spectrum of neurodegenerative disorders, including Parkinson's disease (PD). In rodents, inhibition of complex I leads to degeneration of dopaminergic neurons of the substantia nigra pars compacta (SNpc), as seen in PD, through activation of mitochondria-dependent apoptotic molecular pathways. In this scenario, complex I blockade increases the soluble pool of cytochrome c in the mitochondrial intermembrane space through oxidative mechanisms, whereas activation of pro-cell death protein Bax is actually necessary to trigger neuronal death by permeabilizing the outer mitochondrial membrane and releasing cytochrome c into the cytosol. Activation of Bax after complex I inhibition relies on its transcriptional induction and translocation to the mitochondria. How complex I deficiency leads to Bax activation is currently unknown. Using gene-targeted mice, we show that the tumor suppressor p53 mediates Bax transcriptional induction after PD-related complex I blockade in vivo , but it does not participate in Bax mitochondrial translocation in this model, either by a transcription-independent mechanism or through the induction of BH3-only proteins Puma or Noxa. Instead, Bax mitochondrial translocation in this model relies mainly on the JNK-dependent activation of the BH3-only protein Bim. Targeting either Bax transcriptional induction or Bax mitochondrial translocation results in a marked attenuation of SNpc dopaminergic cell death caused by complex I inhibition. These results provide further insight into the pathogenesis of PD neurodegeneration and identify molecular targets of potential therapeutic significance for this disabling neurological illness.
