    Abstract: Morin is a flavonoid isolated from members of the Moraceae family. Morin has been reported to possess antioxidative and anticarcinogenic activities. However, the antiosteoarthritic properties of morin have not been investigated. In this study, we evaluate the antiarthritic properties of morin through in vitro and in vivo studies. We examined the effects of morin on the expression levels of matrix metalloproteinase (MMP)-3, MMP-13 and tissue inhibitors of metalloproteinase (TIMP)-1 in interleukin-1 β (IL-1 β)-induced rat chondrocytes by realtime polymerase chain reaction and Western blotting. The effects of morin on the phosphorylation of mitogen-activated protein kinases were also investigated. The in vivo antiosteoarthritic effects of morin were evaluated in the rat model of anterior cruciate ligament transection (ACLT)-induced osteoarthritis (OA). We found that morin inhibited the expression of MMP-3 and MMP-13 and increased the expression of TIMP-1 in IL-1 β-induced rat chondrocytes. In addition, morin inhibited IL-1 β-induced phosphorylation of extracellular signal-regulated kinase and p38. For the in vivo study in a rat model of OA induced by ACLT, in which morin was orally administered to rat, the results show that morin suppressed cartilage degradation. Our results suggest that morin may be considered as a possible therapeutic agent for the treatment of OA.

    Abstract: We explored the feasibility of reconstructing tracheal wall defects with a mesh patch fashioned from a nickel-titanium shape-memory alloy. Methods: A tracheal wall defect was first constructed surgically by resecting the anterior half of the tracheal wall between the second and sixth tracheal rings. The defect was reconstructed in 8 experimental animals by replacing the resected tracheal mucosa and tracheal cartilage with a pedicle skin flap, which was then enclosed in the mesh patch. In 4 control animals, only a pedicle skin flap with strap muscles was used in the reconstruction procedure. The performance of the animals was observed after surgery. At the end of the experiments, the reconstructed segment was harvested for anatomic evaluation. Results: In the experimental group, 1 animal died 5 days after the operation. Endoscopic and anatomic examination of the 7 animals that survived the observation period showed that the reconstructed trachea was stable, with sufficient airway space for breathing. All 4 control animals died after the operation. After observing successful completion of this operation in animals, we successfully used this method to repair a tracheal wall defect in a human victim of a traffic accident. Conclusions: Tracheal defects can be successfully reconstructed by use of a mesh patch of nickel-titanium shape-memory alloy as an extraluminal stent – a method that avoids complications associated with intraluminal stents.

    Abstract: The purpose of this study was to develop a poly(galacturonic acid) (PGA)-based hydrogel using a short-chain hyaluronate (sHA) cross-linker for medical applications. PGA was grafted with adipic acid dihydrazide (ADH) to yield PGA–ADH, an amine-containing PGA derivative. This PGA–ADH formed a water-insoluble hydrogel by reacting with 1,1′-carbonyldiimidazole (CDI)–grafted sHA (sHA–CDI) in aqueous solution. The sHA–cross-linked PGA hydrogel has a water content of about 94%–97% and compressive modulus of 10.7–26.9 kPa. The in vitro data indicated that the sHA–cross-linked PGA hydrogel is degradable and noncytotoxic, thus suitable for biomedical applications. Animal implant studies showed that the sHA–cross-linked PGA hydrogel membrane exhibited antiadhesion potency, significantly higher than that found in untreated rats and has great potential for future clinical applications.

    Abstract: The protein phosphatase-2A (PP-2A), one of the major phosphatases in eukaryotes, is a heterotrimer, consisting of a scaffold A subunit, a catalytic C subunit and a regulatory B subunit. Previous studies have shown that besides regulating specific PP-2A activity, various B subunits encoded by more than 16 different genes, may have other functions. To explore the possible roles of the regulatory subunits of PP-2A in vertebrate development, we have cloned the PR55/B family regulatory subunits: β and δ, analyzed their tissue specific and developmental expression patterns in Goldfish ( Carassius auratus). Our results revealed that the full-length cDNA for PR55/Bβ consists of 1940 bp with an open reading frame of 1332 nucleotides coding for a deduced protein of 443 amino acids. The full length PR55/Bδ cDNA is 2163 bp containing an open reading frame of 1347 nucleotides encoding a deduced protein of 448 amino acids. The two isoforms of PR55/B display high levels of sequence identity with their counterparts in other species. The PR55/Bβ mRNA and protein are detected in brain and heart. In contrast, the PR55/Bδ is expressed in all 9 tissues examined at both mRNA and protein levels. During development of goldfish, the mRNAs for PR55/Bβ and PR55/Bδ show distinct patterns. At the protein level, PR55/Bδ is expressed at all developmental stages examined, suggesting its important role in regulating goldfish development. Expression of the PR55/Bδ anti-sense RNA leads to significant downregulation of PR55/Bδ proteins and caused severe abnormality in goldfish trunk and eye development. Together, our results suggested that PR55/Bδ plays an important role in governing normal trunk and eye formation during goldfish development.

    Abstract: Thymoquinone (TQ) is the main constituent of Nigella sativa oil, which has been traditionally used against arthritis in the Middle East. In this study, we investigated the effect of TQ against matrix metalloproteinase (MMP) expression in both rabbit chondrocytes and animal mode of osteoarthritis (OA) induced by anterior cruciate ligament transection and tested whether or not nuclear factor kappa B (NF- κB) was involved in this process. TQ down-regulated MMP-1, MMP-3 and MMP-13 expression and up-regulated tissue inhibitors of metalloproteinase-1 expression as assessed by quantitative realtime polymerase chain reaction. In addition, NF- κB p65 protein level as well as its translocation induced by interleukin-1 β were inhibited by TQ. Our findings suggest the potential of TQ in the treatment of OA.

    Abstract: The tumor-suppressor ING3 has been shown to be involved in tumor transcriptional regulation, apoptosis and the cell cycle. Some studies have demonstrated that ING3 is dysregulated in several types of cancers. However, the expression and function of ING3 in human hepatocellular carcinoma (HCC) remains unclear. The aim of this study is to investigate ING3 expression in hepatic tumors and its clinical relevance in hepatic cancer. The expression of ING3 protein was examined in 120 dissected HCC tissues and 47 liver tissues adjacent to the tumor by immunohistochemical assays and confirmed by Western blot analysis in 20 paired frozen tumor and non-tumor liver tissues. The relationship between ING3 staining and clinico-pathological characteristics of HCC was further analyzed. The mRNA expression of ING3 in the dissected tissues was also analyzed by reverse transcriptase polymerase chain reaction (RT-PCR) and realtime PCR. Both mRNA and protein concentrations of ING3 were found to be downregulated in the majority of HCC tumors in comparison with matched non-tumor hepatic tissues. Analysis of the relationship between ING3 staining and clinico-pathological characteristics of HCC showed that the low expression of ING3 protein is correlated with more aggressive behavior of the tumor. Kaplan–Meier curves demonstrated that patients with a low expression of ING3 have a significantly increased risk of shortened survival time. In addition, multivariate analysis suggested that the level of ING3 expression may be an independent prognostic factor. Our findings indicate that ING3 may be an important marker for human hepatocellular carcinoma progression and prognosis, as well as a potential therapeutic target.

    Abstract: The cohesive zone model (CZM), which is based on the elasto-plastic fracture mechanics, is nowadays widely used in the numerical simulation of the composites ductile crack and progressive delamination. For the sake of tracking the angle-ply composites delamination evolution accurately, the exponential CZM is applied. At first the standard double cantilever beam (DCB) specimen is modeled using the exponential interface damage law to verify the feasibility of it. The simulated results are in good agreement with the analytical results and corrected beam theory. Then in order to investigate how the ply angle influences the mode I fracture, five laminates with different ply orientations, [0 12 /0 12 ], [0 12 /30 12 ], [0 12 /45 12 ], [0 12 /60 12 ] and [0 12 /90 12 ], are studied. The main results are obtained as follows. The delamination evolutions corresponding to different ply angles are quite non-uniform. The unidirectional laminate exhibits the stable, continuous crack growth along the specimen, so does the [0 12 /90 12 ] specimen. Whereas the other three specimens show very uneven fracture surfaces. When the plies change from [0 12 /0 12 ] to [0 12 /90 12 ], the stiffness along the specimen is gradually reduced correspondingly, so are the distance of the delamination propagation and the peak load, while the corresponding loading displacement increases slowly. The research provides the numerical simulation method to evaluate the multidirectional laminated composites fracture toughness, and then to optimize the composite performance by the design of ply angles.

    Abstract: The small flowrate and the diaphragm’s short life are two shortcomings of the diaphragm compressor. This paper presents a new generatrix of the cavity profile of a diaphragm compressor to increase cavity volume and decrease diaphragm's radial stress. To verify the design theory, the radial stresses on the oil side of the diaphragm in the cavities with the new and traditional generatrices were tested, and the experimental radial stresses agreed with the theoretical values. As the most important evaluation criteria of the cavity profile, the volumes of the cavities with different generatrices and the radial stress distribution of the diaphragm within were investigated under various design conditions. The results showed that the volume of the cavity with the new generatrix was about 6.5% larger than that with the traditional generatrix under the same design condition. Otherwise, with the same cavity volume and radius, the maximal radial stress of the diaphragm in the cavity with the new generatrix decreased by 10.3% stably, compared to that in the cavity with a traditional generatrix. Likewise, in the diaphragm’s centric region where the additional stress caused by the discharge holes occurred, the maximal radial stress of the diaphragm in the cavity with the new generatrix decreased about 11.5%.

    Abstract: The components and claddings of low-rise buildings are usually destroyed first during typhoon disasters in coastal areas. Roof accessory structures can change the flow pattern on the roof, thus effectively reducing the wind load on the roof surface and the damage to the low-rise buildings. Three types of aerodynamic mitigation plates, that is, (1) 0.3 m high full-length roof-edge plate, (2) 0.3 m high and 0.5 m + 0.5 m long roof-corner plate, and (3) discrete roof-edge plates with different spaces (which can be used as advertisement boards), are studied on the basis of the field measurement results under roof pitches of 10° and 18.4°. By comparing the results of the roof with and without constructed plates, it is implied that the three types of plates can affect the formation of conical vortexes and can significantly reduce the mean and fluctuating pressure coefficients in the windward corner. Compared with the constructed plate, the roof ridge has a larger influence on the wind loads on leeward roof.
