Notes: Abstract— Partially purified myelin from the brains of 17-day-old rats was separated into 4 subfractions on a three-step sucrose gradient by virtue of heterogeneity in density and particle size. Precursor-product relationships between different membrane fractions were investigated by determining the specific radioactivity of individual lipids in each subcellular fraction 15 min after intracranial injection of an appropriate precursor. Rats were injected with [2-3H]glycerol. myelin subfractions prepared, and individual lipids separated by TLC. For choline and ethanolamine phospholipids, specific radioactivity was highest in the densest fraction (D), intermediate in the next densest fraction (C), and lowest in the lighter fractions (B and A). Similar results were observed for cerebroside and sulphatide when [3H]galactose was the precursor. These data are consistent with (but do not prove) a precursor-product relationship for individual lipids from the densest to the lightest subfraction.Another experimental design involving time staggered injections of [3H] and [14C] precursors was developed which enables a more definitive result with regard to precursor-product relationships to be obtained. A precursor-product relationship between a given lipid in a dense myelin membrane fraction, and the same lipid in a lighter subfraction, would be indicated by a change in isotope ratio. If there is no precursor-product relationship. Ihe isotope ratio should be constant. Such experiments were done with [3H] and [14C]glycerol. The data indicated that phosphatidyl ethanolamine and its plasmalogen analog were added first to the densest subfraction and then in turn to the lighter subfractions. In contrast, phosphatidyl choline and its plasmalogen analog were added “simultaneously” (i.e. with delays of much less than 15min) to each of the subfractions. Similar experiments with [3H] and [14C]galactose showed that cerebroside, sulphatide and galactosyl diglyceride also entered the subfractions simultaneously rather than in sequential order. Thus the assembly of the myelin sheath involves an obligate order of addition of certain lipids. while other lipids are probably added in a random order.

Notes: Abstract— This paper specifies methodology for the sequential determination of manganese and catecholamines in selfsame brain samples and shows correlations between them. Small samples were obtained from five regions of brain of cats that had received either saline or levodopa. The doses of levodopa were varied so that although all animals reacted, some developed dyskinesia while others did not. Each sample was first analyzed nondestructively for manganese and then destructively for dopa and dopamine; thus errors inherent in analysing separate samples, due to the structural heterogeneity of the brain, were avoided. Statistically significant correlations were found (1) between levodopa-induced dyskinesia and the concentrations of dopamine and manganese in some of the regions analysed, and (2) between the concentrations of dopamine and of manganese in the caudates of the cats receiving the highest doses of levodopa.

Notes: Summary In mice given3H-thymidine systemically during temporary circulatory occlusion of one hind limb, comparison of the labeling of rapidly-renewing small lymphocytes in the tibial marrows demonstrated that these cells were locally produced. Labeling by3H-thymidine infusion revealed that many marrow large lymphoid cells, presumptive small lymphocyte progenitors, had a marked proliferative activity and rapid turnover which varied according to cell size, was maximal in young mice and declined with increasing age.

Notes: Summary In several vertebrate species it has been clearly demonstrated that neural circuits within the spinal cord are capable of generating the signals underlying locomotory rhythms. In the present paper a halfcentre model is proposed for the spinal locomotor generator which is based entirely on the connections and properties of known spinal neurones. In its simplest form the model involves six groups of neurones: Two sets of alpha motorneurones (α-MN), Renshaw cells (RC) and Ia inhibitory interneurones (Ia-IN), associated, respectively, with flexor and extensor muscles. Mutual inhibition between antagonist Ia-INs provides the basis for a neuronal ‘flip-flop’ in which one group is active and the other depressed. A novel element in the model is the RC which completes a feedback pathway from a-MNs to Ia-INs. This pathways converts the ‘flip-flop’ into a free running oscillator. Reciprocal inhibition betwen groups of RCs is necessary to prevent the oscillator stopping under certain circumstances. The model has been simulated using electronic analogue neurones. Variation in the tonic excitation of α-MNs changes the frequency of the oscillator. Reduction of the tonic excitation of Ia-INs causes the model to lock into a state resembling standing, or even decerebrate rigidity or spasticity. Afferent excitation of Ia-INs produces prolongation of the current step. The oscillator may be entrained by inhibitory inputs to the Ia-INs from another oscillator. The broader implications of the model in relation to the control of locomotion, posture and other types of movement are discussed.

Notes: Abstract The Tamayo transform fault occurs at the north end of the East Pacific Rise where it enters the Gulf of California. The two deep-tow surveys reported here show that the transform fault zone changes significantly as a function of distance from the spreading center intersections. At site 1, near the intersection, one side of the fault is young and the fault zone is narrow and well-defined. Strike slip occurs in a zone approximately 1-km wide suggesting a correspondingly narrow zone of decoupling between the Pacific and North American plates. On the young side of the strike-slip zone, normal faults occur along shear zones which are 45°–50° oblique to the transform strike. They occur parallel to the short axis of the strain ellipse for transform fault strain here, i.e., perpendicular to the least compressive stress. The transform walls are formed by normal faulting as has been pointed out in previous detailed surveys. Here, however, the age contrast of 2.5 m.y. across the transform valley is apparent in the morphology of the normal fault scarps. While the scarps are steep and well-defined on the young side, the scarps on the older side have gradual 10°–30° slopes and appear to be primarily talus ramps. Apparently, the scarps have been tectonically eroded by continued strike slip activity after the initial stages of normal faulting. Thus, transform valleys should be quite asymmetric in cross-section where there is a significant age contrast and one side is less than approximately 0.5 m.y. old. Also, along older sections of the transform valley walls, normal faulting may not be at all obvious due to degradation of the scarps by tectonic erosion. This phenomenon makes the likelihood of transform faults providing ‘windows’ into the oceanic crust most unlikely except in special cases. The picture of transform deformation is more complex at site 2 in the central portion of the fault where both sides of the fault are greater than 1 m.y. old. Here the transform valley is wider (25–30 km as opposed to 2–5 km). There is no clear simple zone of strike slip tectonics. In fact, the only clear evidence for deformation is the intrusion of magmatic or serpentinite diapirs through the sediments of the transform valley floor. The diapirs have deformed the turbidite layers flooring the valley and in one carefully studied case the turbidite sequence has been uplifted, perched atop the diapir. The pattern of deformation on this outcropping diapir shows radial and concentric fractures which can be modeled by a vertical intrusion circular in plan view. Magnetic studies limit the possible composition to basalt or serpentinite. A 60-km-long median ridge is also likely to be the product of intrusion along the transform fault. The survey at site 2 pointed out the importance of vertical tectonics in the transform valley floor and in particular the importance of diapiric intrusions of either basaltic or serpentinite composition. Based on initial boundary conditions and present tectonic elements in the Tamayo fault zone, a possible history of the mouth of the Gulf of California is outlined. The median ridge was emplaced starting approximately 0.8 m.y. ago by regional extension across the transform fault, the result of ‘leaky’ transform faulting. The diapirs occur along a possible ‘relay’ zone of extension midway along the fault which began approximately 0.15 m.y. ago. The extension in this case is parallel to the trend of the transform fault, is still occurring at present, and may evolve into a true spreading center.

Notes: Summary Ethylene production of McIntosh apples was reduced by both a postharvest treatment with CaCl2 and a daminozide treatment applied in the field. The CaCl2 treatment was more effective than daminozide in reducing the rate of softening during storage at 1°C.

Notes: Abstract A serial longitudinal study was undertaken to determine the activities of the alkaline and acid phosphatases in the long bones and mandibles. The optimum pH of the two enzymes was recorded at 10.2 and 5.4 for alkaline and acid phosphatase, respectively. Synchronized and randomized litters of rats were killed, 1 litter daily, starting at day 1 to day 25 post partum. Samples were analyzed for protein concentration and activity of the phosphatases. A pattern of low and high activity was observed in both bony tissues, as well as a pattern of low alkaline phosphatase activity during acid phosphatase activity peaks, and vice versa. The observed peaks suggest a correspondence between phosphatase activity and the other biochemical changes occurring in the growing bone, i.e., organic matrix and mineral formation. A separate study, considering the possibility of rhythmic features of the enzyme activity suggests that there may be a small diurnal rhythm at an early age.
