    Abstract: Dừa cạn (Catharanthus roseus (L.) G. Don) là loài cây dược liệu qu. có khả năng sản xuất alkaloid có giá trị dược liệu cao, trong đó vinblastine và vincristine có tác dụng điều trị ung thư máu, tuy nhiên hàm lượng vinblastine và vincristine trong cây dừa cạn rất thấp. Peroxidase là enzyme chìa khóa trong con đường sinh tổng hợp các terpenoid alkaloid indole (TIA) ở cây dừa cạn, có chức năng xúc tác trong phản ứng tạo tiền chất cho sự tổng hợp vinblastine và vincristine. Nghiên cứu biểu hiện mạnh gen mã hóa peroxidase (CrPrx) để tăng cường hoạt động của peroxidase nhằm cải thiện hàm lượng vinblastine và vincristine trong cây dừa cạn được chúng tôi quan tâm. Trong bài báo này chúng tôi trình bày kết quả thiết kế vector chuyển gen chứa cấu trúc mang gen CrPrx phân lập từ cây dừa cạn. Vector chuyển gen pBI121-CrPrx được thiết kế từ kỹ thuật thu nhận gen CrPrx từ vector tái tổ hợp pBT-CrPrx, tạo vector tái tổ hợp pRTRA7/3-CrPrx, thu nhận cấu trúc CrPrxcmyc- KDEL, ghép nối tạo vector chuyển gen pBI121-CrPrx. Vector chuyển gen pBI121-CrPrx được dòng hóa trong A. tumefaciens và lây nhiễm vào dừa cạn qua nách lá mầm. Trong 666 mẫu biến nạp có 197 mẫu tạo chồi, 65 mẫu có chồi kéo dài trên môi trường chọn lọc bằng kháng sinh kanamycin, thu được 15 cây T0 phát triển trên giá thể và 7 cây dừa cạn chuyển gen ở thế hệ T0 cho kết quả PCR nhân bản đoạn promoter 35S. Kết quả thiết kế cấu trúc mang gen chuyển hoàn chỉnh (cassette) và tăng cường biểu hiện gen mã hóa enzyme chìa khóa sẽ quyết định sự thành công của kỹ thuật tạo cây dừa cạn biến đổi gen có hàm lượng alkaloid cao.

    Abstract: Interleukin là một nhóm các cytokine được tìm thấy đầu tiên trong các tế bào bạch cầu, đóng vai trò điều hòa đáp ứng miễn dịch. Trong đó, interleukin 7 (IL7) là một cytokine có vai trò chính trong sự tăng trưởng của các dòng tế bào B và T, đây là những dòng tế bào có chức năng quan trọng trong hệ miễn dịch của con người. Trong nghiên cứu này, chúng tôi tiến hành tổng hợp nhân tạo gen IL7 với mã di truyền được tối ưu hóa biểu hiện trong dòng tế bào thuốc lá BY-2. Chúng tôi sử dụng vector pK7WG2D.1/cal để tạo cấu trúc pK7WG2D.1/cal/IL7. Cấu trúc này được sử dụng để chuyển gen IL7 vào các dòng tế bào thuốc lá BY-2. Gen đích trong các dòng tế bào BY-2 được đánh giá bằng kỹ thuật PCR và protein tái tổ hợp được xác định bằng lai miễn dịch Western blot. Kết quả cho thấy, đã tạo được các dòng tế bào BY-2 sinh trưởng ổn định trong môi trường lỏng sau 5 lần cấy chuyển với khoảng cách mỗi lần cấy chuyển là 2 tuần. Các dòng BY-2 chuyển gen sinh trưởng ổn định và biểu hiện protein tái tổ hợp IL7 có kích thước khoảng 23 kDa. Đây là kết quả mới ở Việt Nam và trên thế giới, IL7 được biểu hiện trong dòng tế bào thuốc lá BY-2. Kết quả này mang lại tiềm năng lớn cho nghiên cứu và sản xuất protein IL7 tái tổ hợp phục vụ trong y học.

    Abstract: Plant defensins play a role against the seed-feeding insects. Defensin associates with the center of α-amylase activity in the gut of weevils, thus inhibiting the digestion of starch by weevils. In this study, the resistance of eight mungbean cultivars to weevils was evaluated by the method of artificial weevils infection. The Tam TH cultivar had lowest index of susceptibility to weevils (634.63) and DX22 cultivar had highest index (1058.72), and the highest resistance to weevils was found in Tam TH and DX22 was found to have the lowest resistance. VrPDF1 genes isolated from mungbean cultivars are 356 bp in length with two exons interrupted by an intron. The coding region of the VrPDF1 gene is 228 bp in length, encoding 75 amino acids. The comparative results of the nucleotide sequence of cDNA between Tam TH  and DX22 showed that there was a difference in 13 nucleotides and comparison of amino acid sequences of the deduced protein indicated that there was a difference in 9 amino acids. Within the intron region of the VrPDF1 genes there was difference in 5 nucleotides. The genetic distance based on nucleotide sequences of the coding region of VrPDF1 gene of DX22 and seven other mungbean cultivars is 6.2% and based on the amino acid sequence deduced is 7.7%. The coding region of the VrPDF1 gene of DX22 was used to create a transformation vector aimed at creating weevil-resistant transgenic mungbean.

    Abstract: Y-chromosome microsatellites or short tandem repeats (STRs) have been proved to be ideal markers to delineate the differences between individuals in human population. Nowadays, Y-STR testing using the PowerPlex® Y23 amplification kit is considered as an extremely sensitive analysis method and has the potential to be used to perform forensic caseworks, and to explore the complexity in population substructures. However, little is known about the forensic Y-chromosome databases in the Vietnam population. In this study, 23 Y-STR loci (DYS576, DYS389I, DYS389 II, DYS448, DYS19, DYS391, DYS481, DYS549, DYS533, DYS438, DYS437, DYS570, DYS635, DYS390, DYS439, DYS392, DYS393, DYS458 DYS456, DYS643, YGATAH4, and DYS385a/b) were investigated in 120 non-related males of the Kinh population in Northern Vietnam using PowerPlex® Y23 system kit (Promega). Our results showed that allele frequencies of 23 loci in the sample population, with the calculated average gene diversity (GD) for each locus, ranged from 0.24 (DYS438) to 0.92 (DYS385a/b). In addition, a total of 120 different haplotypes were found, all of them were unique. Therefore, we found that the haplotype diversity was 1 with a discrimination capacity of 100%, which serves as an essential prerequisite for using Y-chromosomal STR with PowerPlex® Y23 System kit in forensic application in Vietnam. We also compared genetic distances between Kinh population and 10 other neighboring populations from Y-chromosome haplotype reference database (YHRD). The Kinh population is significantly different from other populations. In conclusion, it was indicated that the 23 Y-STR loci were highly genetically polymorphic in the Kinh population in Vietnam and might be of great value in forensic application.

    Abstract: Hybridization between wild and domestic bovine occurs widely due to the overlapping of the natural habitat of the wild and human farm animals. Due to the loss of habitat, the number of wild gaur (Bos gaurus) in Vietnam was in serious decline. Since 2009, a male wild gaur (Bos gaurus) has appeared and incorporated with female domestic cows (Bos taurus) in buffer zone Phuoc Binh National Park, on the border with Lam Dong and Ninh Thuan provinces. Then, several calves were born that carried some traits of wild gaur including physical characteristics and behaviors. These calves were supposed to be offspring between wild gaur and domestic cows. In previous study, the karyotypes of putative calves were identified with 2n = 58 and non-homologous chromosome 28 and 29 in these hybrids. In this study, we characterized the putative hybrids between track-lacking Bos gaurus and Bos taurus for breeding and preservation using Cytochrome b analysis, microsatellites and a novel marker growth hormone factor 1 (POU1F1). Cytochrome b analysis indicated the maternal lineage of the putative hybrids, with 100% nucleotide sequence identity. Microsatellite BM861 and sequence of ZFY gene region reveal Bos taurus chromosome Y origin was among the male putative hybrids. Importantly, the analysis of POU1F1 gene sequence on 1 chromosome showed efficacy in determining both Bos gaurus and Bos taurus lineage in the putative hybrids. The karyotyping results were confirmed by molecular analysis and our results provide a feasible way for detecting the putative hybrids between wild and domestic cattle in case of lacking the wild trace.

    Abstract: In this paper, the characteristics of wind speed at 20 m height at the Bac Lieu atmospheric physic station (Bac Lieu station) in 2016 were evaluated using the Weibull distribution function. The wind speed data set (every minute) from January 7th  to December 31st, 2016 was used to calculate the two parameters of  Weibull function including Weibull shape factor “k” and Weibull scale factor “c”. The results showed that at the Bac Lieu station in 2016, the values of k and c were 1.69 and 3.91, respectively. Some characteristics of wind speed were also estimated such as wind energy density (Pa/A=57.3 W/m2), wind speed of maximum energy carrier (Vmec=6.2 m/s), the most probable wind speed (Vmp=2.3 m/s), mean wind speed (Vmean­=3.5 m/s)  and standard deviation of wind speeds (s = 2.1 m/s).
