    Notes: SUMMARY. Delayed pressure urticaria in 15 patients has been investigated. It is characterized by the development of deep painful swelling 4–6 hours after pressure has been applied to the skin. It is sometitnes accompanied by systemic manifestations such as a leucocytosis and rigors.Delayed pressure urticaria is unaffected by antihistamines but it is controlled by steroids.The injection into the skin of saline and various pharmacological agents also causes delayed wealing. Prior treatment of the skin with 48/80 prevents the reaction.Immunofluorescent studies showed that the affected tissues contained no excess fibrinogen, immunoglobulins or complement.It is suggested that delayed pressure urticaria is related to delayed dermographism and may be due to a disorder of kinin activity.The timing and depth of the changes in vascular permeability characteristic of urticaria can be correlated. They are influenced by many mediators and by the pattern of the vasculature of the skin. A change in the timing or depth of the urticaria is the most that should be expected from the use of a single pharmacological agent in the treatment of urticaria.

    Description: An unknown member of the family Pasteurellaceae was repeatedly isolated from 20- to 24-week-old pigs with severe pulmonary lesions reared on the same farm in Victoria, Australia. The etiological diagnosis of the disease was inconclusive. The complete genome sequence analysis of one strain, 15-184, revealed some phylogenic proximity to Glaesserella ( Haemophilus ) parasuis , the cause of Glasser's disease. However, the sequences of the 16S rRNA and housekeeping genes, as well as the average nucleotide identity scores, differed from those of all other known species in the family Pasteurellaceae . The protein content of 15-184 was composite, with 60% of coding sequences matching known G. parasuis products, while more than 20% had a closer relative in the genera Actinobacillus , Mannheimia , Pasteurella , and Bibersteinia . Several putative virulence genes absent from G. parasuis but present in other Pasteurellaceae were also found, including the apxIII RTX toxin gene from Actinobacillus pleuropneumoniae , ABC transporters from Actinobacillus minor , and iron transporters from various species. Three prophages and one integrative conjugative element were present in the isolate. Horizontal gene transfers might explain the mosaic genomic structure and atypical metabolic and virulence characteristics of 15-184. This organism has not been assigned a taxonomic position in the family, but this study underlines the need for a large-scale epidemiological and clinical characterization of this novel pathogen in swine populations, as a genomic analysis suggests it could have a severe impact on pig health. IMPORTANCE Several species of Pasteurellaceae cause a range of significant diseases in pigs. A novel member of this family was recently isolated from Australian pigs suffering from severe respiratory infections. Comparative whole-genome analyses suggest that this bacterium represents a new species, which possesses a number of virulence genes horizontally acquired from a diverse range of other Pasteurellaceae . While the possible contribution of other coinfecting noncultivable agents to the disease has not been ruled out in this study, the repertoire of virulence genes found in this organism may nevertheless explain some aspects of the associated pathology observed on the farm. The prevalence of this novel pathogen within pig populations is currently unknown. This finding is of particular importance for the pig industry, as this organism can have a serious impact on the health of these animals.

    Description: Using a large initial condition ensemble of climate model simulations, we examine the impact of volcanic activity on Arctic sea ice cover from 1960 to 2005, a period that includes three very large tropical eruptions. Ensemble averaging across simulations with natural (volcanic and solar) forcings alone reduces noise due to internal variability to show a decade of increased Arctic sea extent (of up to half a million square kilometers) following each of the Mount Agung (1963), Mount El Chichón (1982) and Mount Pinatubo (1991) eruptions. A similar impact is seen when averaging over a large ensemble of simulations with natural and allknown anthropogenic forcings. We show that the volcanic response in sea ice cover is sensitive to pre-eruption temperature, with warmer conditions before an eruption being associated with a larger than average response. Finally, a detection and attribution analysis using CanESM2 did not identify a significant response in the observations, while finding no evidence of inconsistency between observations and CanESM2 since regression coefficients were consistent with unity. A similar detection and attribution analysis using the somewhat stronger volcanic response from the simulations in the average of the CMIP5 models, did identify a detectable natural forcing response in four observational sea ice extent datasets.

    Description: Memory stem T cells (TSCMs) constitute a long-lived, self-renewing lymphocyte population essential for the maintenance of functional immunity. Hallmarks of autoimmune disease pathogenesis are abnormal CD4 + and CD8 + T cell activation. We investigated the TSCM subset in 55, 34, 43, and 5 patients with acquired aplastic anemia (AA), autoimmune uveitis, systemic lupus erythematosus, and sickle cell disease, respectively, as well as in 41 age-matched healthy controls. CD8 + TSCM frequency was significantly increased in AA compared with healthy controls. An increased CD8 + TSCM frequency at diagnosis was associated with responsiveness to immunosuppressive therapy, and an elevated CD8 + TSCM population after immunosuppressive therapy correlated with treatment failure or relapse in AA patients. IFN- and IL-2 production was significantly increased in various CD8 + and CD4 + T cell subsets in AA patients, including CD8 + and CD4 + TSCMs. CD8 + TSCM frequency was also increased in patients with autoimmune uveitis or sickle cell disease. A positive correlation between CD4 + and CD8 + TSCM frequencies was found in AA, autoimmune uveitis, and systemic lupus erythematosus. Evaluation of PD-1, CD160, and CD244 expression revealed that TSCMs were less exhausted compared with other types of memory T cells. Our results suggest that the CD8 + TSCM subset is a novel biomarker and a potential therapeutic target for AA.

    Description: Parasitic worms of the genus Trichinella (phylum Nematoda; class Enoplea) represent a complex of at least twelve taxa that infect a range of different host animals, including humans, around the world. They are foodborne, intracellular nematodes, and their life cycles differ substantially from those of other nematodes. The recent characterization of the genomes and transcriptomes of all twelve recognized taxa of Trichinella now allows, for the first time, detailed studies of their molecular biology. In the present study, we defined, curated, and compared the protein kinase complements (kinomes) of Trichinella spiralis and T. pseudospiralis using an integrated bioinformatic workflow employing transcriptomic and genomic data sets. We examined how variation in the kinome might link to unique aspects of Trichinella morphology, biology, and evolution. Furthermore, we utilized in silico structural modeling to discover and characterize a novel, MOS-like kinase with an unusual, previously undescribed N-terminal domain. Taken together, the present findings provide a basis for comparative investigations of nematode kinomes, and might facilitate the identification of Enoplea-specific intervention and diagnostic targets. Importantly, the in silico modeling approach assessed here provides an exciting prospect of being able to identify and classify currently unknown (orphan) kinases, as a foundation for their subsequent structural and functional investigation.

    Description: Aberrant glycosylation and the overexpression of specific carbohydrate epitopes is a hallmark of many cancers, and tumor-associated oligosaccharides are actively investigated as targets for immunotherapy and diagnostics. Wisteria floribunda agglutinin (WFA) is a legume lectin that recognizes terminal N-acetylgalactosaminides with high affinity. WFA preferentially binds the disaccharide LacdiNAc (β-d-GalNAc-[1→4]-d-GlcNAc), which is associated with tumor malignancy in leukemia, prostate, pancreatic, ovarian, and liver cancers and has shown promise in cancer glycobiomarker detection. The mechanism of specificity for WFA recognition of LacdiNAc is not fully understood. To address this problem, we have determined affinities and structure of WFA in complex with GalNAc and LacdiNAc. Affinities toward Gal, GalNAc, and LacdiNAc were measured via surface plasmon resonance, yielding KD values of 4.67 × 10−4 m, 9.24 × 10−5 m, and 5.45 × 10−6 m, respectively. Structures of WFA in complex with LacdiNAc and GalNAc have been determined to 1.80–2.32 Å resolution. These high resolution structures revealed a hydrophobic groove complementary to the GalNAc and, to a minor extent, to the back-face of the GlcNAc sugar ring. Remarkably, the contribution of this small hydrophobic surface significantly increases the observed affinity for LacdiNAc over GalNAc. Tandem MS sequencing confirmed the presence of two isolectin forms in commercially available WFA differing only in the identities of two amino acids. Finally, the WFA carbohydrate binding site is similar to a homologous lectin isolated from Vatairea macrocarpa in complex with GalNAc, which, unlike WFA, binds not only αGalNAc but also terminal Ser/Thr O-linked αGalNAc (Tn antigen).

    Description: Paroxysmal nocturnal hemoglobinuria (PNH) is a rare acquired disorder originating from hematopoietic stem cells and is a life-threating disease characterized by intravascular hemolysis, bone marrow (BM) failure, and venous thrombosis. The etiology of PNH is a somatic mutation in the phosphatidylinositol glycan class A gene ( PIG-A ) on the X chromosome, which blocks synthesis of the glycolipid moiety and causes deficiency in GPI-anchored proteins. PNH is closely related to aplastic anemia, in which T cells mediate destruction of BM. To identify aberrant molecular mechanisms involved in immune targeting of hematopoietic stem cells in BM, we applied RNA-seq to examine the transcriptome of T cell subsets (CD4 + naive, CD4 + memory, CD8 + naive, and CD8 + memory) from PNH patients and healthy control subjects. Differentially expressed gene analysis in four different T cell subsets from PNH and healthy control subjects showed distinct transcriptional profiles, depending on the T cell subsets. By pathway analysis, we identified novel signaling pathways in T cell subsets from PNH, including increased gene expression involved in TNFR, IGF1, NOTCH, AP-1, and ATF2 pathways. Dysregulation of several candidate genes ( JUN , TNFAIP3 , TOB1 , GIMAP4 , GIMAP6 , TRMT112 , NR4A2 , CD69 , and TNFSF8 ) was validated by quantitative real-time RT-PCR and flow cytometry. We have demonstrated molecular signatures associated with positive and negative regulators in T cells, suggesting novel pathophysiologic mechanisms in PNH. These pathways may be targets for new strategies to modulate T cell immune responses in BM failure.
