    Description: The NE dipping slab of the Hellenic subduction is imaged in unprecedented detail using teleseismic receiver function analysis on a dense 2-D seismic array. Mapping of slab geometry for over 300 km along strike and down to 100 km depth reveals a segmentation into dipping panels by along-dip faults. Resolved intermediate-depth seismicity commonly attributed to dehydration embrittlement is shown to be clustered along these faults. Large earthquakes occurrence within the upper and lower plate and at the interplate megathrust boundary show a striking correlation with the slab faults suggesting high mechanical coupling between the two plates. Our results imply that the general slab rollback occurs here in a differential piecewise manner imposing its specific stress and deformation pattern onto the overriding Aegean plate.

    Description: Imaging the architecture of mountain roots is required to understand the support of topography and for kinematic reconstructions at convergent plate boundaries, but is still challenging with conventional seismic imaging approaches. Here we present a three-dimensional model of both compressional and shear velocities in the lithosphere beneath the western Pyrenees (southwest Europe), obtained by full waveform inversion of teleseismic P waves. This tomographic model reveals the subduction of the Iberian crust beneath the European plate, and the European serpentinized subcontinental mantle emplaced at shallow crustal levels beneath the Mauléon basin. The rift-inherited mantle wedge acted as an indenter during the Pyrenean convergence. These new results provide compelling evidence for the role of rift-inherited structures during mountain building in Alpine-type orogens.

    Description: End-stage renal failure patients exhibit a high incidence of genetic damage and genomic instability. Part of this genetic damage is assumed to be caused by the hemodialysis (HD) procedure. To reduce these effects, different alternative HD procedures have been proposed, such as the use of high efficiency convective therapies to improve the reactive oxygen species/antioxidant ratio. To determine the efficiency of online hemodiafiltration (HDF) technique on the levels of DNA damage, we have measured the frequency of micronucleus in peripheral blood lymphocytes of 33 individuals moving from low-flux HD to post-dilution online HDF. In addition to basal levels of genetic damage, potential changes in radiosensitivity were measured as indicators of genomic instability. Plasma antioxidant capacity was also determined. Second samples were obtained after 6 months on the HDF protocol. Results indicate that moving to online HDF therapy produce a significant reduction of the basal levels of genetic damage, but does not affect the genomic instability status. In addition, a greater increase in plasma antioxidant capacity was observed. In spite of the lack of correlation between these parameters, our results confirm the usefulness of the online HDF technique as a way to reduce DNA damage in HD patients.

    Description: Merkel cell carcinoma is a highly aggressive form of skin cancer. Merkel cell polyomavirus (MCV) infection and DNA integration into the host genome correlate with 80% of all Merkel cell carcinoma cases. Integration of the MCV genome frequently results in mutations in the large tumor antigen (LT), leading to expression of a truncated LT that retains pRB binding but with a deletion of the C-terminal domain. Studies from our laboratory and others have shown that the MCV LT C-terminal helicase domain contains growth-inhibiting properties. Additionally, we have shown that host DNA damage response factors are recruited to viral replication centers. In this study, we identified a novel MCV LT phosphorylation site at Ser-816 in the C-terminal domain. We demonstrate that activation of the ATM pathway stimulated MCV LT phosphorylation at Ser-816, whereas inhibition of ATM kinase activity prevented LT phosphorylation at this site. In vitro phosphorylation experiments confirmed that ATM kinase is responsible for phosphorylating MCV LT at Ser-816. Finally, we show that ATM kinase-mediated MCV LT Ser-816 phosphorylation may contribute to the anti-tumorigenic properties of the MCV LT C-terminal domain.

    Description: Between 2011 and 2013, two dense transects were deployed across the central and western Pyrenees to get better constraints on the deep lithospheric architecture and discriminate the competing models of the structure and formation of the Pyrenees. Each transect recorded the regional and global seismicity during a period of approximately 1 yr. Here, we exploit the records of teleseismic compressional waves and of their conversions to shear waves on internal discontinuities in order to map lithospheric interfaces beneath the two transects. The migrated sections, obtained by performing common conversion point stacks, are in remarkable agreement with the results of the ECORS-Pyrenees and ECORS-Arzacq deep seismic surveys. However, the migrations of converted waves reveal new details of the deep lithospheric architecture that could not be seen with the active source experiments. The new images provide clear and definite evidence for the subduction of a thinned Iberian crust down to at least ~70 km depth, a result that has important implications for the formation of the Pyrenees. The subduction of the Iberian lithosphere leads to reconsider the amount of convergence between Iberia and Europe during the Cenozoic. A recent regional P -wave tomography, relying on the data of the PYROPE and IBERARRAY temporary experiments, revealed the segmentation of lithospheric structures by inherited Hercynian NE–SW transfer faults that were reactivated during the Albian rifting. Our migration images are consistent with this model, and give further support to the idea that the Pyrenees were produced by the tectonic inversion of a segmented hyperextended rift that was buried by subduction beneath the European Plate.

    Description: Although the analysis of genetic interactions and networks is a powerful approach to understanding biology, it has not been applied widely to the pathogenic yeast Candida albicans . Here, we describe the use of both screening and directed genetic interaction studies based on complex haploinsufficiency to probe the function of the R egulation of A ce2 and M orphogenesis (RAM) pathway in C. albicans . A library of 5200 Tn7 -mutagenized derivatives of a parental strain heterozygous at CBK1 , the key kinase in the RAM pathway, was screened for alterations in serum-induced filamentation. Following confirmation of phenotypes and identification of insertion sites by sequencing, a set of 36 unique double heterozygous strains showing complex haploinsufficiency was obtained. In addition to a large set of genes regulated by the RAM transcription factor Ace2, genes related to cell wall biosynthesis, cell cycle, polarity, oxidative stress, and nitrogen utilization were identified. Follow-up analysis led to the first demonstration that the RAM pathway is required for oxidative stress tolerance in a manner related to the two-component-regulated kinase Chk1 and revealed a potential direct connection between the RAM pathway and the essential Mps1 spindle pole-related kinase. In addition, genetic interactions with CDC42 -related genes MSB1 , a putative scaffold protein, and RGD3 , a putative Rho GTPase-activating protein (GAP) were identified. We also provide evidence that Rgd3 is a GAP for Cdc42 and show that its localization and phosphorylation are dependent on Cbk1.

    Description: Galectin-3–binding protein (gal3bp) and its receptor/ligand, galectin-3 (gal3), are secreted proteins that initiate signaling cascades in several diseases, and recent human proteomic data suggest they may play a role in venous thrombosis (VT). We hypothesized that gal3bp and gal3 may promote VT. Using a mouse stasis model of VT, we found that gal3bp and gal3 were localized on vein wall, red blood cells, platelets, and microparticles, whereas leukocytes expressed gal3 only. Gal3 was dramatically increased during early VT and gal3bp:gal3 colocalized in the leukocyte/endothelial cell interface, where leukocytes were partially attached to the vein wall. Thrombus size correlated with elevated gal3 and interleukin-6 (IL-6) vein wall levels. Recombinant gal3 promoted VT and increased vein wall IL-6 mRNA. Although recombinant gal3 restored the VT size in gal3 –/– mice, it had no effect on IL6 –/– mice, suggesting that gal3:gal3bp promotes VT through IL-6. Moreover, significantly fewer activated neutrophils were present in the gal3 –/– vein walls. In a group of human patients, elevated circulating gal3bp correlated with acute VT. In conclusion, gal3bp:gal3 play a critical role in VT, likely via IL-6 and PMN-mediated thrombotic mechanisms, and may be a potential biomarker in human VT.
