    Notes: Abstract: Specific binding sites for cholecystokinin (CCK) have been characterized in a particulate membrane fraction of rat cerebral cortex using a biologically active 125I-labeled derivative of the C-terminal octapeptide of CCK (CCK-8) prepared by reaction with the iodinated form of the imidoester (125IIE), methyl-p-hydroxybenzimidate. The time course of binding to cortical membranes was rapid, temperature dependent, and saturable. Half-maximal binding at 24°C was reached in 30 min and full binding at 120 min. At 37°C there was only a slight increase in 125IIE-CCK-8 bound after 15 min. The addition of a large excess of CCK-8 after 30 min of binding at 24°C caused a prompt and rapid decline in radioligand bound showing that the interaction was reversible. There was a progressive decline in the amount of 125IIE-CCK-8 bound to membranes with increasing concentrations of CCK-8 and other structurally related peptides. CCK-8 displaced 50% of the radioligand at 4 nM, CCK-33 at 10 nM, and gastrin (desulfated CCK-8) at 60 nM. Secretin, a structurally unrelated peptide, was unable to displace the radioligand from cortical membranes at 1.0 μM. Finally, 125IIE-CCK-8 exposed to cortical membranes or to buffers that had previously contained such membranes for 60 min at 24°C bound equally as well to fresh cortical membranes as control radioligand that had not been exposed to the same conditions. Thus the 125I-CCK-8 radioligand used in this study was highly resistant to degradative processes in rat brain tissue.

    Notes: Summary The fine structure of supraependymal cell clusters of the median eminence was studied with TEM. The cluster cells were identified on the basis of ultrastructure and histochemical determination of glial fibrillar acidic protein (GFA). The phagocytic properties were also studied by means of intraventricular injections of HRP. Neurons, neuroglia cells and degenerating ependyma- and glial cells were found. The extrusion of degenerating infundibular elements into the ventricle is a constant phenomenon but its precise localization and intensity are variable. The close proximity of the clusters to capillary loops is stressed. Because of the broken ependyma at the neck of the cluster, the permeability of the infundibular lining for HRP is increased. Clusters may be seen as sites lacking a brain-CSF barrier.

    Notes: Summary We present a family, identified through a girl with “Cri-du-chat syndrome”, in which two different types of recombinants exist [del(5)(qter→p14:) and dup(5)(p13)]. They are due to a 5p paracentric inversion of maternal origin [inv(5) (pter p13)]. We discuss the relationship between the breakpoints and segregation of the inversion carrier, as well as the origin and the identification of the recombinants.

    Description / Table of Contents: Zusammenfassung Die Gallensalze setzen den enterohepatischen Kreislauf fort. Die Cholsäure (Carboxylic14C), nach ihrer intravenösen Injektion verteilt sich im Niveau des Portalkreislaufes, Dünndarmwand, Inhalt des Dünndarms, Leber und Stuhl, welches als gutes Zeichen der Absorbierungskapazität der Epithelzellen des Darms betrachtet werden kann. In Ratten, bei denen eine totale Dünndarmresektion (60–80 cm) durchgeführt wurde, beobachtete man nach 7 Tagen eine bedeutende Verminderung der Absorbierung der Cholsäure. Trotzdem ist 30 Tage nach der Operation das Gewicht der Tiere sowie die radioaktiven Befunde gleich den Tieren der Kontrollgruppe, und zwar im Blut der Portalvene und der V. Cava, sowie im Darminhalt, Leber und Stuhl. In dieser Studie wurde eine Erhöhung der Absorbierungskapazität der Epithelzellen des Dünndarms beobachtet.
    Notes: Summary Bile salts undergo an enterohepatic circulation. The relative distribution of labelled cholic acid in portal blood, enterocytes, intestinal contents and feces, following intravenous injection of (carboxylic-14C) cholic acid, can be considered as an indication of the absorptive capacity of the epithelium cells. Seven days after a massive small intestinal resection (60–80 cm), the animal weight and the bile salt absorption were significantly decreased. Nevertheless, thirty days after the operation, weight and radioactivities reached the values found in the controls: in portal and cava blood, liver, intestinal contents and feces. An increase of the absorptive capacity of the intestinal epithelial cells was also observed.

    Description: The maintenance of the endangered Caspian brown trout Salmo trutta caspius populations in Iran depends on its artificial breeding. There is no information on how current hatchery breeding protocol influences effective population size of breeders (Ne), which is a critical parameter to maintain genetic diversity in future generations. The current hatchery protocol (CHP) was comparatively evaluated with a balanced hatchery protocol (BHP), where mixtures of equal amounts of gametes per male and female breeder were used to balance parental contributions to progeny. To evaluate putative differences in viability between families, individual crosses were performed and fertilized ova of different families were mixed to constitute balanced family pools. 1440 alevins were totally sampled in the crosses performed from the 11 breeders. An exclusion-based parentage approach using three polymorphic microsatellite markers unambiguously assigned more than 93% of progeny to a single pair of parents. Significantly different contributions of breeders to progeny were observed in CHP (p〈0.05). The primary constraint on Ne in BHP was the unbalanced contribution of males, which seemed a consequence of sperm competition in mixed fertilization caused by differences in sperm quality. Sperm motility duration was positively correlated with the number of sired progeny by each male. The results illustrate the limitations of the BHP in minimizing the loss of genetic diversity observed in CHP. A protocol based on mixture of equal number of fertilized ova from individual male × female crosses emerged as the best alternative for conservation of Caspian brown trout.
