    Description: Society’s needs for a network of in situ ocean observing systems cross many areas of earth and marine science. Here we review the science themes that benefit from data supplied from ocean observatories. Understanding from existing studies is fragmented to the extent that it lacks the coherent long-term monitoring needed to address questions at the scales essential to understand climate change and improve geo-hazard early warning. Data sets from the deep sea are particularly rare with long-term data available from only a few locations worldwide. These science areas have impacts on societal health and well-being and our awareness of ocean function in a shifting climate. Substantial efforts are underway to realise a network of open-ocean observatories around European Seas that will operate over multiple decades. Some systems are already collecting high-resolution data from surface, water column, seafloor, and sub-seafloor sensors linked to shore by satellite or cable connection in real or near-real time, along with samples and other data collected in a delayed mode. We expect that such observatories will contribute to answering major ocean science questions including: How can monitoring of factors such as seismic activity, pore fluid chemistry and pressure, and gas hydrate stability improve seismic, slope failure, and tsunami warning? What aspects of physical oceanography, biogeochemical cycling, and ecosystems will be most sensitive to climatic and anthropogenic change? What are natural versus anthropogenic changes? Most fundamentally, how are marine processes that occur at differing scales related? The development of ocean observatories provides a substantial opportunity for ocean science to evolve in Europe. Here we also describe some basic attributes of network design. Observatory networks provide the means to coordinate and integrate the collection of standardised data capable of bridging measurement scales across a dispersed area in European Seas adding needed certainty to estimates of future oceanic conditions. Observatory data can be analysed along with other data such as those from satellites, drifting floats, autonomous underwater vehicles, model analysis, and the known distribution and abundances of marine fauna in order to address some of the questions posed above. Standardised methods for information management are also becoming established to ensure better accessibility and traceability of these data sets and ultimately to increase their use for societal benefit. The connection of ocean observatory effort into larger frameworks including the Global Earth Observation System of Systems (GEOSS) and the Global Monitoring of Environment and Security (GMES) is integral to its success. It is in a greater integrated framework that the full potential of the component systems will be realised.

    Description: Cold-water coral (CWC) reefs are heterogeneous ecosystems comprising numerous microhabitats. A typical European CWC reef provides various biogenic microhabitats (within, on and surrounding colonies of coral species such as Lophelia pertusa, Paragorgia arborea and Primnoa resedaeformis, or formed by their remains after death). These microhabitats may be surrounded and intermixed with non-biogenic microhabitats (soft sediment, hard ground, gravel/pebbles, steep walls). To date, studies of distribution of sessile fauna across CWC reefs have been more numerous than those investigating mobile fauna distribution. In this study we quantified shrimp densities associated with key CWC microhabitat categories at the Røst Reef, Norway, by analysing image data collected by towed video sled in June 2007. We also investigated shrimp distribution patterns on the local scale (〈40 cm) and how these may vary with microhabitat. Shrimp abundances at the Røst Reef were on average an order of magnitude greater in biogenic reef microhabitats than in non-biogenic microhabitats. Greatest shrimp densities were observed in association with live Paragorgia arborea microhabitat (43 shrimp m−2, SD = 35.5), live Primnoa resedaeformis microhabitat (41.6 shrimp m−2, SD = 26.1) and live Lophelia pertusa microhabitat (24.4 shrimp m−2, SD = 18.6). In non-biogenic microhabitat, shrimp densities were 〈2 shrimp m−2. CWC reef microhabitats appear to support greater shrimp densities than the surrounding non-biogenic microhabitats at the Røst Reef, at least at the time of survey.

    Description: Since 2009, unprecedented comprehensive long-term gas hydrate observations have become available from Ocean Networks Canada's NEPTUNE cabled ocean observatory at the northern Cascadia margin. Several experiments demonstrate the scientific importance of permanent power and Internet connectivity to the ocean floor as they have advanced the field of gas hydrate related research. One example is the cabled crawler Wally at Barkley Canyon, enabling live in situ exploration of the hydrate mounds and its associated benthic communities through the crawler's mobility and permanent accessibility throughout the year. Another example is a bubble-imaging sonar at Clayoquot Slope, revealing the strong relationship between ebullition of natural gas and tidal pressure, without apparent correlation to earthquakes, storms, or temperature fluctuations, in year-long continuous recordings. Finally, regular observatory maintenance cruises allow additional science sampling including echo-sounder surveys to extend the observatory footprint. Long-term trends in the data are not yet apparent but can also become evident from continuous measurements, as ocean observatories such as NEPTUNE are built for a 25-year lifetime, and expansion of the observatory networks makes these findings comparable and testable. Plain Language Summary Natural gas near the ocean floor creates a rapidly changing environment where it is important to collect data continuously in order to determine the magnitude, speed, and potential mechanism of change. This long-standing challenge of year-round access to the deep ocean has been tackled by Ocean Networks Canada through cabling the northern Cascadia seafloor, providing power and Internet communication-ideal for power-hungry instruments, large data volumes, and real-time access. The presence of gas influences the shape of the seafloor, animal activity, and potential escape of methane, a potent greenhouse gas. A seafloor crawler Wally was operated around deep canyon mounds of gas hydrate (a solid gas-water composite) since 2009 and helped discover environmental changes influencing sea life. Further along the continental slope, an acoustic sonar monitored rising methane bubbles where the bubbling appears to be controlled neither by earthquakes, winter storms, nor subtle temperature changes but actually strongly by tidal pressure. Regular maintenance of the observatory by ship allows more data to be collected near the cabled seafloor sites, extending the observations to a larger area. Ocean observatories are built to last decades and therefore more data for more research can be collected, potentially detecting relatively slow processes as well.

    Description: Daptomycin is a lipopeptide antibiotic used clinically for the treatment of methicillin-resistant Staphylococcus aureus (MRSA) infections. The emergence of daptomycin-nonsusceptible S. aureus isolates during therapy is often associated with multiple genetic changes; however, the relative contributions of these changes to resistance and other phenotypic changes usually remain unclear. The present study was undertaken to investigate this issue using a genetically characterized series of four isogenic clinical MRSA strains derived from a patient with bacteremia before and during daptomycin treatment. The first strain obtained after daptomycin therapy carried a single-nucleotide polymorphism (SNP) in rpoB (RpoB A 477 D) that decreased susceptibility not only to daptomycin but also to vancomycin, β-lactams, and rifampin. Furthermore, the rpoB mutant exhibited pleiotropic phenotypes, including increased cell wall thickness, reduced expression of virulence traits, induced expression of the stress-associated transcriptional regulator Spx, and slow growth. A subsequently acquired loss-of-function mutation in clpX partly alleviated the growth defect conferred by the rpoB mutation without changing antibiotic susceptibility. The final isolate acquired three additional mutations, including an SNP in mprF (MprF S 295 L) known to confer daptomycin nonsusceptibility, and accordingly, this isolate was the only daptomycin-nonsusceptible strain of this series. Interestingly, in this isolate, the cell wall had regained the same thickness as that of the parental strain, while the level of transcription of the vraSR (cell wall stress regulator) was increased. In conclusion, this study illustrates how serial genetic changes selected in vivo contribute to daptomycin nonsusceptibility, growth fitness, and virulence traits.

    Description: In temperate climates with surplus precipitation and low temperatures during autumn and winter, nitrate catch crops have become crucial in reducing nitrate leaching losses. Preferably, the N retained by the catch crop should remain in the soil and become available to the next main crop. Fodder radish ( Raphanus sativus , L.) has emerged as a promising nitrate catch crop in cereal cropping, although the course of remineralization of residue N following termination of this frost-sensitive crucifer remains obscured. We incubated radish residues of different age (different planting and harvest dates) with a loamy sand soil; mineralization of residue N was determined after 1, 2, 4 and 7 months of incubation at 2 °C and 10 °C. Incubations with soil only and with residues of white mustard ( Sinapis alba, L) and perennial ryegrass ( Lolium perenne , L.) were included as references. Using linear regression, net N release was fitted to plant chemical characteristics (initial concentrations of N, fibre fractions, lignin and C/N ratio). Residue C/N ratio (ranging from 10 to 25) and N concentration (ranging from 17 to 40 mg N/g dry matter) showed superior fits to net N release at both temperatures ( r 2 , 0.64–0.94) while fibre analyses provided inferior fits ( r 2 , 0.12–0.64). This was true across planting date and plant age. Net N release after 7 months of incubation at 2 °C and 10 °C accounted for up to 40% and 50% of residue N, respectively. During most of the incubation period, nitrate dominated the mineral N pool at both temperatures. The N mineralization and nitrification potential at these low soil temperatures suggest that a considerable fraction of the N captured by nitrate catch crops may be remineralized, nitrified and thus available for plant uptake but also for loss by leaching and denitrification.

    Description: Aims Cardiovascular conditions are reported to be the most frequent cause of death in patients with chronic obstructive pulmonary disease (COPD). However, it remains unsettled whether severity of COPD per se is associated with coronary artery disease (CAD) independent of traditional cardiovascular risk factors. The aim of this study was to examine the relationship between the presence and severity of COPD and the amount of coronary artery calcium deposit, an indicator of CAD and cardiac risk, in a large population of current and former long-term smokers. Methods and results In this cross-sectional study, long-term smokers without clinically manifested CAD were recruited from the Danish Lung Cancer Screening Trial and classified according to lung function by the Global Initiative for Chronic Obstructive Lung Disease (GOLD) criteria. Coronary artery calcium deposit as a measure of subclinical CAD and cardiac risk was evaluated with multi detector computed tomography and the Agatston coronary artery calcium score (CACS). Participants were categorized into five CACS risk classification groups according to the CACS. The population ( n = 1535) consisted of 41% participants without COPD, 28% with mild, and 31% with moderate-to-severe COPD ( n = 46 with severe COPD). In addition to age, male gender, hypertension, hypercholesterolaemia, and continued smoking, COPD according to GOLD classification were independent predictors of a higher CACS risk classification group in multivariable analysis [odds ratio (OR): 1.28 (1.01–1.63) and OR: 1.32 (1.05–1.67), for mild and moderate-to-severe COPD, respectively, compared with no COPD]. Conclusion COPD in long-term smokers is independently correlated with the CACS, while COPD severity per se does not show a dose–response relationship.
