    Beschreibung: Viral infectivity factor (Vif) is required for lentivirus fitness and pathogenicity, except in equine infectious anemia virus (EIAV). Vif enhances viral infectivity by a Cullin5-Elongin B/C E3 complex to inactivate the host restriction factor APOBEC3. Core-binding factor subunit beta (CBF-β) is a cell factor that was recently shown to be important for the primate lentiviral Vif function. Non-primate lentiviral Vif also degrades APOBEC3 through the proteasome pathway. However, it is unclear whether CBF-β is required for the non-primate lentiviral Vif function. In this study, we demonstrated that the Vifs of non-primate lentiviruses, including feline immunodeficiency virus (FIV), bovine immunodeficiency virus (BIV), caprine arthritis encephalitis virus (CAEV), and maedi-visna virus (MVV), do not interact with CBF-β. In addition, CBF-β did not promote the stability of FIV, BIV, CAEV, and MVV Vifs. Furthermore, CBF-β silencing or overexpression did not affect non-primate lentiviral Vif-mediated APOBEC3 degradation. Our results suggest that non-primate lentiviral Vif induces APOBEC3 degradation through a different mechanism than primate lentiviral Vif. IMPORTANCE The APOBEC3 protein family members are host restriction factors that block retrovirus replication. Vif, an accessory protein of lentivirus, degrades APOBEC3 to rescue viral infectivity by forming Cullin5-Elongin B/C-based E3 complex. CBF-β was proved to be a novel regulator of primate lentiviral Vif function. In this study, we found that CBF-β knockdown or overexpression did not affect FIV Vif's function, which induced polyubiquitination and degradation of APOBEC3 by recruiting the E3 complex in a manner similar to that of HIV-1 Vif. We also showed that other non-primate lentiviral Vifs did not require CBF-β to degrade APOBEC3. CBF-β did not interact with non-primate lentiviral Vifs or promote their stability. These results suggest that a different mechanism exists for the Vif-APOBEC interaction and that non-primates are not suitable animal models for exploring pharmacological interventions that disrupt Vif–CBF-β interaction.

    Beschreibung: Background Radiological characteristics may reflect the biological features of brain tumors and may be associated with genetic alterations that occur in tumorigenesis. This study aimed to investigate the relationship between radiological features and IDH1 status as well as their predictive value for survival of glioblastoma patients. Methods The clinical information and MR images of 280 patients with histologically confirmed glioblastoma were retrospectively reviewed. The radiological characteristics of tumors were examined on MR images, and the IDH1 status was determined using DNA sequencing for all cases. The Kaplan-Meier method and Cox regression model were used to identify prognostic factors for progression-free and overall survival. Results The IDH1 mutation was associated with longer progression-free survival ( P = .022; hazard ratio, 0.602) and overall survival ( P = .018; hazard ratio, 0.554). In patients with the IDH1 mutation, tumor contrast enhancement and peritumoral edema indicated worse progression-free survival ( P = .015 and P = .024, respectively) and worse overall survival ( P = .024 and P = .032, respectively). For tumors with contrast enhancement , multifocal contrast enhancement of the tumor lesion was associated with poor progression-free survival ( P = .002) and poor overall survival ( P = .010) in patients with wild-type IDH1 tumors. Conclusions Combining the radiological features and IDH1 status of a tumor allows more accurate prediction of survival outcomes in glioblastoma patients. The complementary roles of genetic changes and radiological features of tumors should be considered in future studies.

    Beschreibung: This study aims to investigate the relationship between the development of pharyngeal taste buds (TBs) and first feeding of Hucho taimen. Larvae were fed with live food (water flea and tubifex) for 8 weeks. TBs of larval pharynx were histologically examined using light microscopy during the rearing experiment. The results showed that the first few TBs primordia were visible within the pharynx 27 days after hatch (DAH), which coincides with the onset of feeding, and the first few TBs with open receptor areas appear 45 DAH. TBs of pharynx were well developed 76 DAH. The number and size of TBs were quantified during larval development. The average number of pharynx was 8.63±1.15, 11.29±0.50, 14.50±1.06, and 17.78±0.47 TBs at 27, 36, 45, and 76 DAH, respectively. The number of TBs increased in both the upper and lower pharynx. The ratio of height to width of TBs showed an increase tendency, ranging from 0.81 to 1.11. The height of TBs showed an increase tendency after 29 DAH during the development. However, the width of TBs exhibited a low at 35 DAH. In conclusion, the development of pharyngeal TBs coincides with the first feeding, and this should lead to a better understanding of improvement of larval rearing in H. taimen hatcheries.

    Beschreibung: Power-to-Gas (PtG) is a chemical energy storage technology that converts electrical energy into a high-energy density combustible gas. This technology alleviates the contradiction between power supply and demand due to the intermittent electricity production from environment-friendly renewable energy. Hydrogen (H2), produced by electrolysis, can be used to produce synthetic methane (CH4) by reacting with carbon dioxide (CO2) that is captured from carbon emission sources. Subsurface gas storage is one of the most important processes in a PtG system. However, nearly a half of the total stored gas is used as cushion gas to maintain a suitable reservoir pressure, indicating large amounts of CH4 might be wasted and trapped in geological formations. Based on a PtG system, CO2 can be a promising choice as a cushion gas due to its high effective compressibility near its critical conditions. In this paper, a numerical model is established based on the theory of the fluid flow and molecular diffusion to study the role of CO2 as a cushion gas in increasing the gas storage capacity. The accuracy of this model is verified by comparing with that of Curtis M. Oldenburg. However, because of the declining purity of recovered gas induced by the convection and diffusion of two kinds of gases in the same reservoir. The influences of reservoir thickness on the distribution of mixed region are discussed. The results show that thicker reservoir would be a better choice for the geological storage of CH4 with CO2 as a cushion gas.

    Beschreibung: The Renewable energy power generation capacity has been rapidly increasing in China recently. Meanwhile, the contradiction between power supply and demand is becoming increasingly more prominent due to the intermittence of renewable energies. On the other hand, on the mitigation of carbon dioxide (CO2) emissions in China needs immediate attention. Power-to-Gas (PtG), a chemical energy storage technology, can convert surplus electricity into combustible gases. Subsurface energy storage can meet the requirements of long term storage with its large capacity. This paper provides a discussion of the entire PtG energy storage technology process and the current research progress. Based on the comparative study of different geological storage schemes for synthetic methane, their respective research progress and limitations are noted. In addition, a full investigation of the distribution and implementation of global PtG and CO2 capture and storage (CCS) demonstration projects is performed. Subsequently, the opportunities and challenges of the development of this technology in China are discussed based on techno-economic and ecological effects analysis. While PtG is expected to be a revolutionary technology that will replace traditional power systems, the main issues of site selection, energy efficiency and the economy still need to be adequately addressed. Additionally, based on the comprehensive discussion of the results of the analysis, power-to-gas and subsurface energy storage implementation strategies, as well as outlook in China are presented.
