    Abstract: Cyclic dipeptides are an important class of natural products owing to their structural diversity and biological activities. In fungi, the cyclo-ring system is formed through the condensation of two α-amino acids via non-ribosomal peptide synthetase (NRPS). However, there are few investigations on the functional identification of this enzyme. Additionally, information on how to increase the production of cyclic dipeptide molecules is relatively scarce. Results We isolated the Eurotium cristatum NWAFU-1 fungus from Jing-Wei Fu brick tea, whose fermentation metabolites contain echinulin-related cyclic dipeptide molecules. We cloned the cirC gene, encoding an NRPS, from E. Cristatum NWAFU-1 and transferred it into the heterologous host Aspergillus oryzae . This transformant produced a novel metabolite possessing an l- tryptophan- l- alanine cyclic dipeptide backbone ( Cyclo -TA). Based on the results of heterologous expression and microsomal catalysis, CriC is the first NRPS characterized in fungi that catalyzes the formation of a cyclic dipeptide from l -tryptophan and l -alanine. After substrate feeding, the final yield reached 34 mg/L. In this study, we have characterized a novel NRPS and developed a new method for cyclic dipeptide production. Conclusions In this study we successfully expressed the E. Cristatum NWAFU-1 criC gene in A. oryzae to efficiently produce cyclic dipeptide compounds. Our findings indicate that the A. oryzae heterologous expression system constitutes an efficient method for the biosynthesis of fungal Cyclic dipeptides.

    Abstract: Orsellinic acid (2,4-dihydroxy-6-methylbenzoic acid, OA) and its structural analog o -Orsellinaldehyde, have become widely used intermediates in clinical drugs synthesis. Although the research on the biosynthesis of such compounds has made significant progress, due to the lack of suitable hosts, there is still far from the industrial production of such compounds based on synthetic biology. Results With the help of genome mining, we found a polyketide synthase (PKS, HerA) in the genome of the Hericium erinaceus , which shares 60% amino acid sequence homology with ArmB from Armillaria mellea , an identified PKS capable of synthesizing OA. To characterize the function of HerA, we cloned herA and heterologously expressed it in Aspergillus oryzae , and successfully detected the production of OA. Subsequently, the introduction of an incomplete PKS (Pks5) from Ustilago maydis containing only three domains (AMP-ACP-R), which was into herA -containing A. oryzae , the resulted in the production of o -Orsellinaldehyde. Considering the economic value of OA and o -Orsellinaldehyde, we then optimized the yield of these compounds in A. oryzae . The screening showed that when maltose was used as carbon source, the yields of OA and o -Orsellinaldehyde were 57.68 mg/L and 15.71 mg/L respectively, while the yields were 340.41 mg/Kg and 84.79 mg/Kg respectively in rice medium for 10 days. Conclusions Herein, we successfully expressed the genes of basidiomycetes using A. oryzae heterologous host. As a fungus of ascomycetes, which not only correctly splices genes of basidiomycetes containing multiple introns, but also efficiently produces their metabolites. This study highlights that A. oryzae is an excellent host for the heterologous production of fungal natural products, and has the potential to become an efficient chassis for the production of basidiomycete secondary metabolites in synthetic biology.     Abstract: Circular RNAs (circRNAs) are known to act as key regulators in a variety of malignancies. However, the role of circRNAs in cervical cancer (CCa) remains largely unknown. Herein, we demonstrated that a circRNA derived from the TADA2A gene (hsa_circ_0043280) was significantly downregulated in CCa and that this reduction in expression was correlated with a poor prognosis. Furthermore, our results demonstrated that hsa_circ_0043280 functions as a tumor suppressor to inhibit tumor growth and metastasis in CCa. Mechanistically, hsa_circ_0043280 competitively sponges miR-203a-3p and prevents miR-203a-3p from reducing the levels of PAQR3. Collectively, our results demonstrate that hsa_circ_0043280 plays a pivotal role in the development and metastasis of CCa, thus suggesting that hsa_circ_0043280 has significant potential as a prognostic biomarker and a therapeutic target for CCa.

    Abstract: The identifiable target effect refers to the preference for helping identified victims and punishing identifiable perpetrators compared to equivalent but unidentifiable counterparts. The identifiable target effect is often attributed to the heightened moral emotions evoked by identified targets. However, the specific neurocognitive processes that mediate and/or modulate this effect remain largely unknown. Here, we combined a third-party punishment game with brain imaging and computational modeling to unravel the neurocomputational underpinnings of the identifiable transgressor effect. Human participants (males and females) acted as bystanders and punished identified or anonymous wrongdoers. Participants were more punitive towards identified wrongdoers than anonymous wrongdoers because they took a vicarious perspective of victims and adopted lower reference points of inequity (i.e., more stringent norms) in the identified context than in the unidentified context. Accordingly, there were larger activity of the ventral anterior insula, more distinct multivariate neural patterns in the dorsal anterior insula and dorsal anterior cingulate cortex, and lower strength between ventral anterior insula and dorsolateral prefrontal cortex and between dorsal anterior insula and ventral striatum connectivity in response to identified transgressors than anonymous transgressors. These findings implicate the interplay of expectancy violations, emotions and self-interest in the identifiability effect. Lastly, individual differences in the identifiability effect were associated with empathic concern/social dominance orientation, activity in the precuneus/cuneus and temporo-parietal junction, and intrinsic functional connectivity of the dorsolateral prefrontal cortex. Together, our work is the first to uncover the neurocomputational processes mediating identifiable transgressor effect and to characterize psychophysiological profiles modulating the effect. Significance Statement The identifiable target effect - more help to identified victims or stronger punishment to identifiable perpetrators - is common in daily life. We examined the neurocomputational mechanisms mediating/modulating the identifiability effect on third-party punishment by bridging literature from economics and cognitive neuroscience. Our findings reveal that identifiable transgressor effect is mediated by lower reference points of inequity (i.e., more stringent norms), which might be associated with a stronger involvement of the emotion processes and a weaker engagement of the analytic/deliberate processes. Furthermore, personality traits, altered brain activity and intrinsic functional connectivity contribute to the individual variance in the identifiability effect. Overall, our study advances the understanding of the identifiability effect by shedding light on its component processes and modulating factors.

    Abstract: This study aims to investigate the features of the ionospheric mid-latitude trough over North America by using the MIT total electron content data obtained during three geomagnetic storms that occurred in August 2018, September 2017, and March 2015. The mid-latitude trough position sharply moves equatorward from the quiet-time subauroral latitude to mid-latitude with the decrease in SYM-H during geomagnetic storms. We find that the ionospheric behavior of TEC around the mid-latitude trough position displays three kinds of ionospheric storm effect: negative ionospheric storm effect, unchanged ionospheric behavior, and positive ionospheric storm effect. These ionospheric storm effects around the mid-latitude trough position are not always produced by the mid-latitude trough. The ionospheric storm effects produced by the mid-latitude trough are limited in the narrow mid-latitude trough regions, and are transmitted to other regions with the movement of the mid-latitude trough.

    Abstract: The pathogenesis and developmental mechanism of early-stage (FIGO 2009 IA2-IIA2) cervical cancer (CC) remain unclear. Seeking novel molecular biomarkers based on The Cancer Genome Atlas (TCGA) will facilitate the understanding of CC pathogenesis and help evaluate early-stage CC prognosis. Methods To identify prognosis-related genes in early-stage CC, we analyzed TCGA mRNA-seq data and clinical data by univariate Cox and Kaplan–Meier plotter analyses. Differential expression analysis identified upregulated genes in early-stage CC. Combined with the genes correlated with unfavorable prognosis, we selected desmoglein-2 (DSG2) for further investigation. To detect DSG2 expression in early-stage CC, we used immunohistochemistry (IHC), quantitative real-time PCR (qRT-PCR) and western blotting. The relationship between the expression of DSG2 and clinical features was analyzed by the Chi square test. Cox analysis was applied to assess the relationship between CC overall survival (OS) and risk factors. The correlations between DSG2 expression and CC cell line proliferation and migration were investigated with Cell Counting Kit-8 (CCK-8) and migration assays. Results There were 416 prognosis-related genes in early-stage CC. DSG2, matrix metallopeptidase 1 (MMP1), carbonic anhydrase IX (CA9), homeobox A1 (HOXA1), and serine protease inhibitor B3 (SERPINB3) were upregulated in early-stage CC compared with adjacent noncancerous tissue (ANT) and correlated with unfavorable prognosis. Among them, DSG2 was most significantly correlated with patient survival. Coexpression analysis indicated that DSG2 was probably involved in cell division, positive regulation of transferase activity, positive regulation of cell migration, EGFR upregulation pathway and regulation of lymphangiogenesis. IHC, qRT-PCR and western blotting showed that DSG2 expression was higher in CC than in normal tissue. Significant correlations were identified between DSG2 expression and several aggressive clinical features, including pelvic lymph node metastasis (PLNM). Multivariate Cox analysis showed that DSG2 and PLNM were independent prognostic factors for OS. DSG2 knockdown inhibited CC cell proliferation and migration. Conclusions DSG2 is a biomarker that promotes tumor proliferation and metastasis and is correlated with poor prognosis in early-stage CC.

    Abstract: In this study, we employ three types of satellite data from two different limb sounders: the FORMOSAT-3/COSMIC (F3/C) radio occultation (RO) technique and the Sounding of the Atmosphere using Broadband Emission Radiometry (SABER) instrument to study the vertical coupling of the 16-09-2015 Chile tsunami-induced perturbations from the stratosphere to the ionosphere. All three types of datasets, including temperature profiles from 10 to 55 km and 16 to 107 km, and electron density profiles from 120 to 550 km, recognized perturbations of different scales at different heights after the Chile tsunami. The vertical scales identified by the wavelet analysis are from 1–2 km, 5–9 km, and 25–50 km in the stratosphere, mesosphere, and ionosphere, respectively. Meanwhile, as a comparison and validation of the reliability, we also revisited the 11-03-2011 Tohoku earthquake/tsunami-related perturbations from the stratosphere to the ionosphere using the same data. It is believed that the two tsunamis both disturbed the whole atmosphere space, and the scale of these signals gradually increases with the increase in altitude but decreases with time. In addition, the tsunami-related ionospheric gravity wavefronts are examined by the F3/C observations. Another interesting point is that the temperature perturbations recorded by the SABER from 70–100 km altitude are found to arrive earlier than the 2015 tsunami wavefront. The findings in this study suggest that the limb-sounding technique is a useful instrument for detecting the tsunami-coupling gravity wave and benefits the tsunami warning system.
