    Abstract: MiRNAs‐containing extracellular vesicles (EVs) possess the unique function of mediating intercellular communication and participating in many biological processes such as post‐transcriptional gene regulation of embryo implantation and placental development. In the present study, Illumina small‐RNA sequencing was used to identify differentially expressed (DE) miRNAs in serum EVs of pregnant (P) and non‐pregnant (NP) Kazakh sheep at Day 17 from mating. The specifically and differentially expressed miRNAs at early pregnancy in sheep were verified by using RT‐PCR. The target genes of DE miRNAs were predicted by bioinformatics software, and the functional and pathway enrichment analysis was performed on Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) terms. A total of 562 miRNAs (210 novel miRNAs) were identified by sequencing, of which 57 miRNAs were differentially expressed, 49 were up‐regulated, 8 were down‐regulated and 22 novel miRNAs were specifically expressed in the pregnant sheep. Eight highly expressed known miRNA (miR‐378‐3p, miR‐320‐3p, miR‐22‐3p, let‐7b, miR‐423‐3p, miR‐221, miR‐296‐3p, miR‐147‐3p) in pregnant group were down‐regulated in the control group. miRNAs‐containing pregnancy‐related terms and regulatory pathways regulation were enriched using both GO and KEGG analyses. Moreover, we also envisioned a miRNA‐mRNA interaction network to understand the function of miRNAs involved in the early pregnancy serum regulatory network. The results of RT‐PCR verification confirmed the reliability of small‐RNA sequencing. Among them, miR‐22‐3p and miR‐378‐3p were significantly differentially expressed (DE) between pregnant sheep and non‐pregnant group ( p   〈   0.01). The site at which oar‐miR‐22‐3p binds MAPK3 was determined with a dual‐luciferase system. This is the first integrated analysis of the expression profiles of EV‐miRNAs and their targets during early pregnancy in ewes. These data identify key miRNAs that influence the implantation of sheep in the early stage of pregnancy, and provide theoretical basis for further molecular regulatory mechanisms research.

    Abstract: Background: Parkinson's disease (PD) and irritable bowel syndrome (IBS) are respectively one of the most common neurodegenerative diseases and functional bowel diseases in the world. Recent studies suggest that patients with IBS seem to have a higher risk of PD, which conflicts with the result of previous meta-analysis. Therefore, the purpose of this systematic review is to evaluate all available evidence, in order to clarify the association between PD and IBS. Methods: Two reviewers independently searched the PubMed, Embase, Web of Science, and Cochrane library on April 25, 2021 to identify all records that explore the association between IBS and PD. All reports that clearly define PD and IBS and analyze the relationship between the two were included. The Newcastle-Ottawa scale was used to assess the risk of bias of included studies. Results: Five studies from four articles involving 2,044,110 subjects were included in this analysis. The pooled results demonstrated a significant association between PD and IBS (1.48; 95% CI: 1.35–1.62, P & lt; 0.001), with subtle heterogeneity ( I 2 = 0.0%, p = 0.585). The association was observed across genders and increased with age. However, the available evidence cannot allow a reliable analysis of the causal relationship between IBS and PD. Conclusion: This study demonstrates a higher risk of PD among subjects with IBS. Future studies are required to further clarify the causation and underlying mechanism of the association.

    Abstract: Urbanization promotes the development of the subway system, and the particulate matter (PM) concentrations inside have received increasing attention. This study first measured the dynamic PM2.5 and PM10 concentrations in a metro carriage in Chengdu and explored the dominant influencing factors. The personal inhalation intakes of different routes were evaluated. The results showed that the in-carriage PM2.5 and PM10 concentrations ranged from 11 to 74 μg/m3 (mean: 36.7 μg/m3) and 13 to 89 μg/m3 (mean: 40.1 μg/m3), respectively. When the train passed from the overground to underground, the in-carriage PM2.5 and PM10 concentrations increased by 30.4% and 32.9%, respectively. No specific linear relationship between passenger number and in-carriage PM concentrations was found. In-carriage PM concentrations decreased after the carriage doors were opened on the platforms. PM2.5 inhalation intakes ranged from 1.08 to 9.52, with a mean of 4.24 µg. For the passengers with the same age and sex, the average inhaled PM2.5 intake in the metro carriage on the route with more underground platforms was higher. This study not only revealed the PM characteristics in the Chengdu metro system for the first time, but also provided guidelines for reducing the in-carriage PM concentrations to build a healthier travel environment.

    Abstract: Curcumin (Cur), a traditional Chinese medicine extracted from natural plant rhizomes, has become a candidate drug for the treatment of diseases due to its anti-inflammatory, anticancer, antioxidant, and antibacterial activities. However, the poor water solubility and low bioavailability of Cur limit its therapeutic effects for clinical applications. A variety of nanocarriers have been successfully developed to improve the water solubility, in vivo distribution, and pharmacokinetics of Cur, as well as to enhance the ability of Cur to polarize macrophages and relieve macrophage oxidative stress or anti-apoptosis, thus accelerating the therapeutic effects of Cur on inflammatory diseases. Herein, we review the design and development of diverse Cur nanoformulations in recent years and introduce the biomedical applications and potential therapeutic mechanisms of Cur nanoformulations in common inflammatory diseases, such as arthritis, neurodegenerative diseases, respiratory diseases, and ulcerative colitis, by regulating macrophage behaviors. Finally, the perspectives of the design and preparation of future nanocarriers aimed at efficiently exerting the biological activity of Cur are briefly discussed.

    Abstract: Antivirals that can combat coronaviruses, including SARS‐CoV‐2 and associated mutants, are urgently needed but lacking. Simultaneously targeting the viral physical structure and replication cycle can endow antivirals with sustainable and broad‐spectrum anti‐coronavirus efficacy, which is difficult to achieve using a single small‐molecule antiviral. Thus, a library of nanomaterials on GX_P2V, a SARS‐CoV‐2‐like coronavirus of pangolin origin, is screened and a surface‐functionalized gold nanocluster (TMA‐GNC) is identified as the top hit. TMA‐GNC inhibits transcription‐ and replication‐competent SARS‐CoV‐2 virus‐like particles and all tested pseudoviruses of SARS‐CoV‐2 variants. TMA‐GNC prevents viral dissemination through destroying membrane integrity physically to enable a virucidal effect, interfering with viral replication by inactivating 3CL protease and priming the innate immune system against coronavirus infection. TMA‐GNC exhibits biocompatibility and significantly reduces viral titers, inflammation, and pathological injury in lungs and tracheas of GX_P2V‐infected hamsters. TMA‐GNC may have a role in controlling the COVID‐19 pandemic and inhibiting future emerging coronaviruses or variants.

    Abstract: In the Ordos Basin, Chang 81, a Member of the Yanchang Formation, features the development of braided river thin-bedded tight sandstones. These sandstones constitute one of the main production layers of tight oil and gas in the Yanchang Formation within the basin. This study integrates data from core samples, drilling, and seismic information to identify braided river thin-bedded sandstones in the Chang 81 Member at Daijiaping, Ordos Basin, using a method of constrained correlation between seismic waveform and seismic facies. This approach aids in determining the sedimentary microfacies types and reservoir characteristics of thin-bedded tight sandstones. We establish a quantitative fitting formula for the width-to-thickness ratio of braided channel sand bodies to finely characterize sand body stacking patterns and spatial distribution of thin-bedded tight sandstones in braided channels. Braided delta plain deposits in the Chang 81 Member at Daijiaping mainly comprise four types of sedimentary microfacies: braided channels, crevasse channels, floodplains, and swamps. The thickness of the reservoir sand body of Chang 81 member is mainly concentrated between 5–25 m, with low porosity and permeability, making it a typical thin-bedded tight sandstone reservoir. A method of constrained correlation between seismic waveforms and seismic facies was employed to identify sand bodies of braided river thin-bedded sandstones in the Chang 81 Member, summarizing four sand body stacking patterns: longitudinal incision type, longitudinal separation type, lateral shifting type, and single channel type. Furthermore, a quantitative forecasting formula of width-to-thickness ratio was established for the river channel scale, providing accurate guidance for well deployment. Horizontal wells deployed from the sand body’s side towards its center in a river channel yield a production 1.8 times higher than that of horizontal wells deployed in the opposite direction. Thin-bedded tight sandstones in braided channels, characterized by flat-top and convex-bottom lenticular seismic facies, hold practical significance in guiding the deployment of horizontal well patterns for tight oil and enhancing oil and gas recovery.
