    Abstract: To capture the full spectrum of genetic risk for autism, we performed a two-stage analysis of rare de novo and inherited coding variants in 42,607 autism cases, including 35,130 new cases recruited online by SPARK. We identified 60 genes with exome-wide significance ( P   〈  2.5 × 10 −6 ), including five new risk genes ( NAV3 , ITSN1 , MARK2 , SCAF1 and HNRNPUL2 ). The association of NAV3 with autism risk is primarily driven by rare inherited loss-of-function (LoF) variants, with an estimated relative risk of 4, consistent with moderate effect. Autistic individuals with LoF variants in the four moderate-risk genes ( NAV3 , ITSN1 , SCAF1 and HNRNPUL2 ; n  = 95) have less cognitive impairment than 129 autistic individuals with LoF variants in highly penetrant genes ( CHD8, SCN2A, ADNP, FOXP1 and SHANK3 ) (59% vs 88%, P  = 1.9 × 10 −6 ). Power calculations suggest that much larger numbers of autism cases are needed to identify additional moderate-risk genes.

    Abstract: In patients with severe aortic valve stenosis at intermediate surgical risk, transcatheter aortic valve replacement (TAVR) with a self-expanding supra-annular valve was noninferior to surgery for all-cause mortality or disabling stroke at 2 years. Comparisons of longer-term clinical and hemodynamic outcomes in these patients are limited. Objective To report prespecified secondary 5-year outcomes from the Symptomatic Aortic Stenosis in Intermediate Risk Subjects Who Need Aortic Valve Replacement (SURTAVI) randomized clinical trial. Design, Setting, and Participants SURTAVI is a prospective randomized, unblinded clinical trial. Randomization was stratified by investigational site and need for revascularization determined by the local heart teams. Patients with severe aortic valve stenosis deemed to be at intermediate risk of 30-day surgical mortality were enrolled at 87 centers from June 19, 2012, to June 30, 2016, in Europe and North America. Analysis took place between August and October 2021. Intervention Patients were randomized to TAVR with a self-expanding, supra-annular transcatheter or a surgical bioprosthesis. Main Outcomes and Measures The prespecified secondary end points of death or disabling stroke and other adverse events and hemodynamic findings at 5 years. An independent clinical event committee adjudicated all serious adverse events and an independent echocardiographic core laboratory evaluated all echocardiograms at 5 years. Results A total of 1660 individuals underwent an attempted TAVR (n = 864) or surgical (n = 796) procedure. The mean (SD) age was 79.8 (6.2) years, 724 (43.6%) were female, and the mean (SD) Society of Thoracic Surgery Predicted Risk of Mortality score was 4.5% (1.6%). At 5 years, the rates of death or disabling stroke were similar (TAVR, 31.3% vs surgery, 30.8%; hazard ratio, 1.02 [95% CI, 0.85-1.22]; P  =   .85). Transprosthetic gradients remained lower (mean [SD], 8.6 [5.5] mm Hg vs 11.2 [6.0] mm Hg; P   & amp;lt; .001) and aortic valve areas were higher (mean [SD], 2.2 [0.7] cm 2 vs 1.8 [0.6] cm 2 ; P   & amp;lt; .001) with TAVR vs surgery. More patients had moderate/severe paravalvular leak with TAVR than surgery (11 [3.0%] vs 2 [0.7%] ; risk difference, 2.37% [95% CI, 0.17%- 4.85%]; P  = .05). New pacemaker implantation rates were higher for TAVR than surgery at 5 years (289 [39.1%] vs 94 [15.1%] ; hazard ratio, 3.30 [95% CI, 2.61-4.17]; log-rank P   & amp;lt; .001), as were valve reintervention rates (27 [3.5%] vs 11 [1.9%] ; hazard ratio, 2.21 [95% CI, 1.10-4.45]; log-rank P  = .02), although between 2 and 5 years only 6 patients who underwent TAVR and 7 who underwent surgery required a reintervention. Conclusions and Relevance Among intermediate-risk patients with symptomatic severe aortic stenosis, major clinical outcomes at 5 years were similar for TAVR and surgery. TAVR was associated with superior hemodynamic valve performance but also with more paravalvular leak and valve reinterventions.

    Abstract: Mass occurrences of Periphylla periphylla in Norwegian fjords cause major concerns related to potential regime shifts that could affect ecosystem stability. 15 years of trawl data (2006–2015), complemented with comprehensive sampling in different areas and seasons (2018–2021) allowed new insights on the dynamics, structure and connectivity of P. periphylla populations within and beyond Trondheimsfjorden. Despite assumed population bursts, no clear trend on P. periphylla population size in Trondheimsfjorden were identified. Sampling frequency and population size suggest a local reproduction of P. periphylla, especially in the inner part of the fjord where young-of-the-year (YOY) individuals occur. Size variations occurred in relation to sampling month, thus pointing at seasonal patterns in growth and reproduction. No distinct population structure of P. periphylla populations within Trondheimsfjorden and over larger spatial scales ( & gt; 100 km) along the Norwegian coast was observed. Such poor geographic population structure provides evidence for a strong dispersal of P. periphylla, potentially triggered by frequent deep-water renewals of the fjords’ basins that enable a high gene flow. Data on P. periphylla long-term dynamics, population structure and connectivity provide valuable information for ecosystem state assessments and enable the advancement of ecosystem management approaches, thus accounting for both stakeholder and ecosystem demands.

    Abstract: The role of small prey ( 〈  200 µm) in larval marine fish nutrition is largely understudied. Here, we explore the contribution of protozooplankton (PZP 20–200 µm) to larval fish diets, compared to metazoan microzooplankton (MZP 55–200 µm). More specifically, we tested whether the contribution of PZP increased during the low productivity season and decreased as larvae grow. We used North Sea autumn spawning herring ( Clupea harengus ) as a case study, as it is a key species with high commercial and ecological importance. In autumn and winter, the potential PZP and MZP prey was dominated by cells  〈  50 µm (mainly Gymnodiniales, Pronoctiluca pelagica, Tripos spp. and Strombidium spp.), while copepod nauplii and copepodites where more abundant in autumn than in winter. Based on their trophic enrichment (∆15 N), larvae preferentially grazed on small MZP  〈  50 µm rather than PZP both in autumn and winter. Larvae of different body size (range 8–14 mm standard length) fed at the same trophic level but on different prey (similar δ 15 N but different δ 13 C). Growth rates (based on RNA/DNA estimates) were similar in autumn and winter, suggesting that growth was not affected by station-specific differences in the composition of the prey field. Our results not only underscore the important role of MZP on larval herring diets both in autumn and wintertime, but also emphasize the limitations of bulk stable isotope analysis. Given the current low recruitment in North Sea herring, these results provide significant information for future monitoring approaches relevant to stock assessment of this species.

    Abstract: B cells and tertiary lymphoid structures (TLS) are reported to be important in survival in cancer. Pancreatic Cancer (PDAC) is one of the most lethal cancer types, and currently, it is the seventh leading cause of cancer-related death worldwide. A better understanding of tumor biology is pivotal to improve clinical outcome. The desmoplastic stroma is a complex system in which crosstalk takes place between cancer-associated fibroblasts, immune cells and cancer cells. Indirect and direct cellular interactions within the tumor microenvironment (TME) drive key processes such as tumor progression, metastasis formation and treatment resistance. In order to understand the aggressiveness of PDAC and its resistance to therapeutics, the TME needs to be further unraveled. There are some limited data about the influence of nerve fibers on cancer progression. Here we show that small nerve fibers are located at lymphoid aggregates in PDAC. This unravels future pathways and has potential to improve clinical outcome by a rational development of new therapeutic strategies.

    Abstract: Causes and consequences of jellyfish bloom formation are subject to controversial discussions worldwide. While medusae have been studied to a broader extent, the knowledge on polyp stages of scyphozoans is limited thus hampering reliable prediction of jellyfish bloom formation. This study describes the occurrence, abundance, habitat characteristics and interactions of scyphozoan Aurelia sp. polyp colonies with other fouling organisms in intertidal and subtidal sectors of Trondheimsfjorden (Norway). In total, 982 polyps were found on 70 substrata of varying material types during a field survey in spring-summer 2018 along a longitudinal gradient within and outside Trondheimsfjorden. The polyps were identified as Aurelia sp. based on molecular species identification. Most polyps were found in bays with macroalgae canopy on the down facing side of artificial and natural substrata (rocks, concrete, iron) and inside rock cracks. Polyp microhabitats included the surface of Ascidia mentula (solitary ascidian), increments of Pomatoceros triqueter (polychete) tubes and dead Balanus balanoides (barnacle) shells. Based on the deployment of settling plates, abundance of Aurelia sp. polyps ranged from 1.2 (± 0.7) to 0.12 (± 0.07) polyps cm –2 . Settlement occurred either directly on the PVC settling plates or as epibionts on the ascidian Ascidia mentula and on barnacle shells of Balanus balanoides . This study provides insights into the potential of local Aurelia sp. polyps contributing to the seasonal occurrence and abundance of Aurelia sp. in fjord systems, where intensive blooms occur annually.

    Abstract: Joint Cruise 1-2 with R/V Kronprins Haakon addressed objectives of RF1, RF2 and RF3 on the Nansen Legacy main transect in open water and within the sea ice. The focus was on comparing the state of the physical, chemical and biological conditions in the southern and northern parts of the study area. Given this was the first research cruise on the vessel, the cruise also focused on testing gear and equipment, and establishing routines for gear deployments, collaboration, data management and storing.

    Abstract: Jellyfish are often described as a nuisance species, but as our understanding shifts to more ecosystem-based conceptions, they are also recognized as both important components of marine ecosystems and a resource for humans. Here, we describe global jellyfish fisheries and review production, fishing methods, and applications based on the existing literature. We then focus on future development of a European jellyfish fishery based on current and recent EU research initiatives. Jellyfish have been a staple food in East Asia for eons and now show a potential for non-food applications as well. The main fishing methods are mostly traditional, with set-nets, driftnets, hand-nets, and scoop-nets utilizing small crafts or beach-seines. All require a lot of manual labor, thus providing vital, albeit seasonal, occupation to weaker populations. Larger commercial vessels such as purse seines and trawlers are newly introduced métiers which may enable a larger catch per unit effort and total catch, but pose questions of selectivity, bycatch, vessel stability, and transshipment. Social concerns arising from the seasonality of jellyfish fisheries must be met in SE Asia, Latin America, and in any location where new fisheries are established. In the EU, we recognize at least 15 species showing potential for commercial harvesting, but as of 2021, a commercial fishery has yet to be developed; as in finfish fisheries, we advise caution and recognition of the role of jellyfish in marine ecosystems in doing so. Sustainable harvesting techniques and practices must be developed and implemented for a viable practice to emerge, and social and ecological needs must also be incorporated into the management plan. Once established, the catch, effort, and stock status must be monitored, regulated, and properly reported to FAO by countries seeking a viable jellyfish fishery. In the near future, novel applications for jellyfish will offer added value and new markets for this traditional resource.
