    Kurzfassung: Standard medical treatment for early non‐small cell lung cancer (NSCLC) is surgery; however, stereotactic body radiotherapy (SBRT) can be a viable substitute for elderly patients who can no longer tolerate or refuse surgery. The unsurpassed care for elderly patients with early NSCLC remains inconclusive. We conducted a propensity‐score matching analysis to compare the efficacy and side‐effects of SBRT and video‐assisted thoracic surgery (VATS) in elderly patients with pathologically‐proven early NSCLC. Methods This single‐institution retrospective study included 310 elderly patients who underwent VATS ( n  = 180) or SBRT ( n  = 130) from January 2011 to November 2018 at the Tianjin Medical University Cancer Institute and Hospital. Sex, performance status, pathology type, Charlson Comorbidity Index, and T‐stage according to the eighth tumor node metastasis classification were considered when performing propensity‐score matching. Results After propensity‐score matching, 71 patients were included in each group for analysis. A median follow‐up period of 53.5 months was observed for all patients; 72 and 53 months for patients undergoing VATS and SBRT, respectively. Overall survival ( p  = 0.492), progression‐free survival ( p  = 0.294), locoregional recurrence‐free survival ( p  = 0.866), and distant metastasis‐free survival ( p  = 0.452) did not differ significantly between the VATS and SBRT groups. Multivariate analysis showed a significant correlation between the T‐stage and overall survival. However, treatment‐related toxicity was lower in the SBRT group than in the VATS group. Conclusions SBRT is an option comparable with surgery for elderly patients with pathologically‐proven early‐stage NSCLC.

    Kurzfassung: Higher‐order cycloadditions, particularly [8+2] cycloadditions, are a straightforward and efficient strategy for constructing significant medium‐sized architectures. Typically, configuration‐restrained conjugated systems are utilized as 8π‐components for higher‐order concerted cycloadditions. However, for this reason, 10‐membered monocyclic skeletons have never been constructed via catalytic asymmetric [8+2] cycloaddition with high peri‐ and stereoselectivity. Here, we accomplished an enantioselective [8+2] dipolar cycloaddition via the merger of visible‐l ight activation and asymmetric palladium catalysis. This protocol provides a new route to 10‐membered monocyclic architectures bearing chiral quaternary stereocenters with high chemo‐, peri‐, and enantioselectivity. The success of this strategy relied on the facile in situ generation of Pd‐containing 1,8‐dipoles and their enantioselective trapping by ketene dipolarophiles, which were formed in situ via a photo‐Wolff rearrangement.

    Kurzfassung: Cell pyroptosis is one of the main forms of neuronal injury after cerebral ischemia–reperfusion. It is accompanied by an inflammatory reaction and regulated by the caspase gene family. Electroacupuncture (EA) can reduce neuronal injury caused by cerebral ischemia–reperfusion, and we speculated that EA can prevent neuronal pyroptosis after cerebral ischemia–reperfusion by regulating the nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3)/caspase-1 pathway. The cerebral ischemia–reperfusion injury model of C57 and caspase-1 gene knockout (Cas-1 ko) mice was established by Longa's method. EA was conducted at acupoints Chize (LU5), Hegu (LI4), Sanyinjiao (SP6), and Zusanli (ST36) for 1.5 h after cerebral ischemia–reperfusion injury for 20 min, and observation was carried out after 24 h. Neurological deficit scores evaluated the neurological function, cerebral infarction volume was observed by triphenyl tetrazolium chloride (TTC) staining, hematoxylin and eosin (H & amp;E) staining, TUNEL and caspase-1 double-labeled fluorescence staining, and NLRP3 and caspase-1 double-labeled immunofluorescence staining that were used to observe the morphology of neurons in hippocampus, and the protein expression of NLRP3, pro-caspase-1, cleaved caspase-1 p20, pro-interleukin-1β (IL-1β), cleaved IL-1β, and GSDMD was detected by Western blot assay. Results showed that EA could reduce the score of neurological deficit, reduce the volume of cerebral infarction and improve the degree of nerve cell injury, and inhibit NLRP3, pro-caspase-1, cleaved caspase-1 p20, pro-IL-1β, cleaved IL-1β, and GSDMD protein expression. In summary, EA plays a neuroprotective role by reducing the pyroptotic neurons that were caspase 1-mediated and inflammatory response after cerebral ischemia–reperfusion.

    Kurzfassung: Bagging is one of the most important techniques for producting high-quality fruits. In the actual of cultivating, we found a new kind of browning in peel of apple fruit that occurs before harvest and worsen during storage period. There are many studies on metabonomic analysis of browning about storage fruits, but few studies on the mechanism of browning before harvest. Results In this study, five-year-old trees of ‘Rui Xue’ (CNA20151469.1) were used as materials. Bagging fruits without browning (BFW) and bagging fruits with browning (BFB) were set as the experimental groups, non-bagging fruits (NBF) were set as control. After partial least squares discriminant analysis (PLS-DA), 50 kinds of metabolites were important with predictive VIP  〉  1 and p -value 〈  0.05. The most important differential metabolites include flavonoids and lipids molecules, 11 flavonoids and 6 lipids molecules were significantly decreased in the BFW compared with NBF. After browning, 11 flavonoids and 7 lipids were further decreased in BFB compared with BFW. Meanwhile, the significantly enriched metabolic pathways include galactose metabolism, ABC membrane transporter protein, flavonoid biosynthesis and linoleic acid metabolism pathways et al. Physiological indicators show that, compared with NBF, the content of malondialdehyde (MDA), hydrogen peroxide (H 2 O 2 ) , superoxide anion (O 2 − ) in peel of BFW and BFB were significantly increased, and the difference of BFB was more significant. Meanwhile, the antioxidant enzyme activities of BFW and BFB were inhibited, which accelerated the destruction of cell structure. In addition, the metabolome and physiological data showed that the significantly decrease of flavonoid was positively correlated with peel browning. So, we analyzed the expression of flavonoid related genes and found that, compared with NBF, the flavonoid synthesis genes MdLAR and MdANR were significantly up-regulated in BFW and BFB, but, the downstream flavonoids-related polymeric genes MdLAC7 and MdLAC14 were also significantly expressed. Conclusions Our findings demonstrated that the microenvironment of fruit was changed by bagging, the destruction of cell structure, the decrease of flavonoids and the increase of triterpenoids were the main reasons for the browning of peel.

    Kurzfassung: Strain CY1518 T was isolated from an anaerobic fermentation liquid of food waste treatment plant in Beijing, PR China, and characterized to assess its taxonomy. Cells of CY1518 T were Gram-stain-negative, oxidase-negative, catalase-positive and ellipsoidal. Growth occurred at 20–42 °C (optimum, 37 °C), pH 6.0–10.0 (optimum, pH 8) and with 0–6.0 % (w/v) NaCl (optimum, 1.5%). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain CY1518 T belongs to the genus Alcanivorax , with the highest sequence similarity to Alcanivorax pacificus W11-5 T (95.97 %), followed by Alcanivorax indicus SW127 T (95.08%). The similarity between strain CY1518 T and other strains of Alcanivorax was less than 95 %. The genomic DNA G+C content of strain CY1518 T was 60.88 mol%. The average nucleotide identity, average amino acid identity and digital DNA–DNA hybridization values between strain CY1518 T and the closely related taxa A. pacificus W11-5 T and A. indicus SW127 T were 77.61, 78.03 and 21.2 % and 74.15, 70.02 and 19.3%, respectively. The strain was able to use d -serine, Tween 40 and some organic acid compounds for growth. The polar lipids comprised aminophospholipid, diphosphatidylglycerol, glycolipid, an unknown polar lipid, phosphatidylethanolamine, phosphatidylglycerol and phospholipid. The principal fatty acids ( 〉 5 %) were C 19 : 0 cyclo ω 8 c (36.3%), C 16 : 0 (32.3%), C 12 : 0 3-OH (8.3%) and C 12 : 0 (7.6%). Based on its phenotypic, genotypic and genomic characteristics, strain CY1518 T represents a novel species in the genus Alcanivorax , for which the name Alcanivorax quisquiliarum sp. nov. is proposed. The type strain is CY1518 T (=GDMCC 1.2918 T =JCM 35120 T ).

    Kurzfassung: Government intervention and financial support are two major means to promote the independent innovation performance of enterprises in strategic emerging industries, and government intervention has induced crowding-out effects on financial support, which leads to the uncertainty of the dual incentive effect of government intervention and financial support on enterprises’ independent innovation. The research object of this paper is 657 strategic emerging enterprises listed in Shanghai and Shenzhen. We empirically studied the impact of government intervention and financial support on the comprehensive efficiency of independent innovation of strategic emerging enterprises. The empirical study draws the following conclusions. Firstly, the comprehensive efficiency of independent innovation of enterprises is in the trend of continuous improvement and technical efficiency and scale efficiency are also increasing, but the technical efficiency is lower than the scale efficiency, which shows that the improvement of independent innovation efficiency mainly depends on the expansion of innovation scale. Secondly, both government intervention and financial support promote the comprehensive efficiency of independent innovation of strategic emerging industry enterprises, but the incentive effect of government intervention is more obvious. Thirdly, there is an inverted U-shaped relationship between government intervention and the comprehensive efficiency of independent innovation. Fourthly, the regression coefficient of the interaction between government intervention and financial support and the comprehensive efficiency of enterprise independent innovation is negative, which indicates that government intervention has an inhibitory effect on the effect of financial support on the overall efficiency of enterprise independent innovation. Finally, we put forward countermeasures and suggestions.

    Kurzfassung: The morphology and ciliature of a new freshwater ciliate, Frontonia apoacuminata sp. nov., isolated from an artificial pond in Qingdao, PR China, were investigated using live observation and silver staining methods. The main features separating F. apoacuminata sp. nov. from its congeners are as follows: a broad elliptical body that is slightly pointed at the posterior end, four ophryokineties, one or two spherical micronuclei of a ‘compact’ type, a dorsally positioned contractile vacuole, and peniculi 1–3 each with five kinetosome rows though the left-most two rows in peniculus 3 are extremely shortened (with only two or three kinetosomes each). Additionally, an improved diagnosis of F. acuminata is provided. Phylogenetic analyses based on the small subunit ribosomal RNA (SSU rRNA) gene show that F. apoacuminata sp. nov. clusters with F. atra , F. minuta , F. acuminata and F. terricola . These five species group with Disematostoma , Marituja and Stokesia rather than with other Frontonia species, causing polyphyly of the genus Frontonia .

    Kurzfassung: We evaluated he effects of molecular guided-targeted therapy for intractable cancer. Also, the epidemiology of druggable gene alterations in Chinese population was investigated. Materials and methods The Long March Pathway (ClinicalTrials.gov identifier: NCT03239015) is a non-randomized, open-label, phase II trial consisting of several basket studies examining the molecular profiles of intractable cancers in the Chinese population. The trial aimed to 1) evaluate the efficacy of targeted therapy for intractable cancer and 2) identify the molecular epidemiology of the tier II gene alterations among Chinese pan-cancer patients. Results In the first stage, molecular profiles of 520 intractable pan-cancer patients were identified, and 115 patients were identified to have tier II gene alterations. Then, 27 of these 115 patients received targeted therapy based on molecular profiles. The overall response rate (ORR) was 29.6% (8/27), and the disease control rate (DCR) was 44.4% (12/27). The median duration of response (DOR) was 4.80 months (95% CI, 3.33−27.2), and median progression-free survival (PFS) was 4.67 months (95% CI, 2.33−9.50). In the second stage, molecular epidemiology of 17,841 Chinese pan-cancer patients demonstrated that the frequency of tier II gene alterations across cancer types is 17.7%. Bladder cancer had the most tier-II alterations (26.1%), followed by breast cancer (22.4%), and non-small cell lung cancer (NSCLC; 20.2%). Conclusion The Long March Pathway trial demonstrated a significant clinical benefit for intractable cancer from molecular-guided targeted therapy in the Chinese population. The frequency of tier II gene alterations across cancer types supports the feasibility of molecular-guided targeted therapy under basket trials.
