    Abstract: Sphingosine kinase 1 (SPHK1) regulates cell proliferation and survival by converting sphingosine to the signaling mediator sphingosine 1-phosphate (S1P). SPHK1 is widely overexpressed in most cancers, promoting tumor progression and is associated with clinical prognosis. Numerous studies have explored SPHK1 as a promising target for cancer therapy. However, due to insufficient knowledge of SPHK1 oncogenic mechanisms, its inhibitors’ therapeutic potential in preventing and treating cancer still needs further investigation. In this review, we summarized the metabolic balance regulated by the SPHK1/S1P signaling pathway and highlighted the oncogenic mechanisms of SPHK1 via the upregulation of autophagy, proliferation, and survival, migration, angiogenesis and inflammation, and inhibition of apoptosis. Drug candidates targeting SPHK1 were also discussed at the end. This review provides new insights into the oncogenic effect of SPHK1 and sheds light on the future direction for targeting SPHK1 as cancer therapy.

    Abstract: Lake Donggi Cona is a key area for the exploration of the prehistoric settlement of the inner Tibetan Plateau because of its location in the joint of the higher inner Tibetan Plateau and the upper Yellow River valley. Here, we carried out archeological investigation on the lake basin, and a total of 256 pieces of stone artifacts were collected from the surface of the DJCN 3-2-2 site on the northern shore terraces of the lake. In addition, AMS 14 C and OSL dating were performed on hearths and section (DJCN 3-2-2), respectively; meanwhile environmental proxies, including grain size, charcoal, magnetic susceptibility, fungal spore, and pollen, were analyzed. The results indicate that the AMS 14 C ages of the charcoal range between ~5.4 and 5.0 cal. ka BP, consistent with OSL dating from the charcoal layers in the DJCN 3-2-2 section (~5.5 ka BP). The functional analysis of the stone artifacts and dating results showed that the site was a seasonal, relative long-term, and central camp which was used for processing of stone artifacts, cutting and consuming food and the production of daily necessities. Charcoal, magnetic susceptibility, and fungal spores from the DJCN 3-2-2 section revealed that prehistoric human activities began at ~5.8 ka BP and significantly intensified during the period of ~5.6–5.5 ka BP and then gradually weakened. Pollen assemblages indicated that the vegetation was dominated by alpine steppe during the period of ~5.8–5.0 ka BP, indicative of a relatively warmer and wetter climatic condition during this period. Taken together, we infer that under the impact of the Neolithic culture in lower altitude of upper Yellow River valley and Microlithic culture in the higher altitude inner Plateau, a Neolithic-Zongri culture had emerged in the transitional region between these two cultures during middle Holocene.

    Abstract: A series of symmetrical 1,4-disubstituted buta-1,3-diynes is prepared with excellent yields (up to 95%) through homocoupling of terminal alkynes catalyzed by a copper salt under solvent-free conditions. This method provides an environmentally friendly process to prepare 1,3-diynes in short reaction times under mild conditions. Furthermore, the method is suitable for a wide substrate scope and has excellent functional group compatibility. The reaction can also be scaled up to gram level.

    Abstract: Increased arterial stiffness is highly prevalent in patients with hypertension and is associated with cardiovascular (CV) risk. Increased white blood cell (WBC) counts may also be an independent risk factor for arterial stiffness and CV events. The aim of the study was to investigate the relationship between differential WBC counts and brachial-ankle pulse wave velocity (baPWV) in hypertensive adults. A total of 14 390 participants were included in the final analysis. A multivariate linear regression model was applied for the correlation analysis of WBC count and baPWV. Higher WBC counts were associated with a greater baPWV: adjusted β = 10 (95% CI, 8-13, P 〈 .001). The same significant association was also found when WBC count was assessed as categories or quartiles. In addition, the effect of differential WBC subtypes, including neutrophil count and lymphocyte count on baPWV, showed the similar results. These findings showed that baPWV has positive associations with differential WBC counts in hypertensive adults.

    Abstract: Atypical porcine pestivirus (APPV) had been detected in many countries. However, to date, a commercial detection kit is not available because of a lack of specific monoclonal antibodies (mAbs) to APPV. We generated 7 mAbs targeting the NS3 protein of APPV. Isotyping results indicated that all of these mAbs are IgG1 with a kappa light chain. We analyzed the epitopes recognized by mAbs 2B6, 6G11, 8D1, 8D3, and 8F12, which recognized the same linear epitope (GRIKSAYSDE); the 6H3 and 7E10 mAbs recognized 2 different conformational epitopes. Applications of these antibodies were verified by ELISA, western blot, indirect immunofluorescence assay, and flow cytometry. The antibodies were functionally workable for these immunoassays except for 8F12, which could not be used in flow cytometry.

    Abstract: Polyamides (PAs) based on terephthaloyl chloride and a series of aromatic diamines were synthesized; PA films were prepared by solution casting without mechanical stretching to produce high-performance films with good optical transparency and dimensional stability. The PA films displayed high heat resistance with glass transition temperatures ( Tg) of 342–399°C. Good mechanical properties were obtained with tensile modulus and strength up to 5.38 GPa and 195.4 MPa, respectively. Coefficient of thermal expansion of the as-cast PA films achieved to an ultralow value of 8.3 ppm °C −1 and could be tailored in a wide range as a function of the macromolecular composition. The series of PA films showed a transparent and colorless feature, with the cutoff wavelength in the range of 355–365 nm. The data were discussed from the perspective of structure–property relationship, may give helpful knowledge for the topic of PA chemistry, and this method could provide an alternative for the production of high-performance colorless polymer films.

    Abstract: Au nanodots decorated Cu 2 O nanooctahedrons were fabricated by a facile liquid-phase process combined with a galvanic replacement reaction for nonenzyme glucose detection. A simple rapid test strip based on the nanooctahedrons was proposed to evaluate the possibility of commercial application in nonenzymatic glucose detection. This test strip shows excellent response toward glucose. Linear response was obtained over a concentration ranging from 0.05 mM to 15 mM, and the detection accuracy is 0.05 mM. The good detection performance in selectivity, stability, and feasibility proving the great potential application in human blood glucose monitoring. This study demonstrated the possibility of a high-performance nonenzyme glucose test strip based on metal-oxide nanostructures decorated by catalysts.

    Abstract: To compare the clinical effects between anatomical locking plates and interlocking intramedullary nails in patients with Danis-Weber B lateral malleolus fractures. Methods This retrospective study enrolled patients with Danis-Weber B fractures of the lateral malleolus. All the operations were completed by the same group of surgeons. The reduction effect, operation time, intraoperative blood loss, hospital stay, fracture healing time and ankle functional outcomes (Olerud-Molander Ankle Score [OMAS]) were compared. Postoperative complications, including incision infections, fixation discomfort and internal fixation loosening, were also compared. Results This study enrolled 73 patients that were treated with either an anatomical locking plate (locking plate group; n = 37) or an interlocking intramedullary nail (intramedullary nail group; n = 36). There was no significant difference between the two groups in terms of reduction effect, hospital stay and OMAS. The intramedullary nail group had significantly lower operation time, intraoperative blood loss and fracture healing time compared with the locking plate group. Postoperative complications in the intramedullary nail group were significantly lower compared with the locking plate group. Conclusion Using intramedullary nails resulted in more satisfactory functional outcomes compared with using locking plates in patients with Danis-Weber B fractures of the lateral malleolus.

    Abstract: To conduct a systematic review and meta-analysis evaluating the diagnostic value of folate receptor-positive (FR+) circulating tumour cells (CTCs) as a potential tumour marker for lung cancer diagnosis. Methods The PubMed, Embase, and Web of Science databases were searched for relevant articles published between database inception and November 2022. Eligible studies were selected based on inclusion and exclusion criteria. Sensitivity, specificity, positive and negative likelihood ratios, diagnostic odds ratio, and area under the curve (AUC) were pooled with 95% confidence intervals (CI), using RevMan 5.4 and STATA 17.0 software to assess the diagnostic value of FR+CTC for lung cancer. Results After screening, 11 studies involving 3469 subjects were eligible for inclusion. The pooled sensitivity and specificity were 0.79 (95% CI 0.76, 0.82) and 0.84 (95% CI 0.81, 0.96), respectively, and the pooled positive and negative likelihood ratios were 4.90 (95% CI 4.25, 5.65) and 0.25 (95% CI 0.22, 0.29), respectively. The pooled diagnostic odds ratio was 19.70 (95% CI 16.06, 24.16). The AUC of the pooled summary receiver operating characteristic curve was 0.89 (95% CI 0.85, 0.91). Sensitivity analysis showed that this result was stable after one-by-one study elimination. Conclusion Folate receptor-positive CTCs may have good diagnostic value in lung cancer.

    Abstract: The occupation process and survival strategies of prehistoric humans on the Tibetan Plateau are important scientific questions for understanding human adaptation to extreme high-altitude environments. Here, we report a newly discovered microlithic site at Daiqu (DQ) in the Tongtian River basin of the central-eastern plateau. We collected 239 lithic artifacts from the DQ site for typological analysis. OSL and AMS 14 C dating samples were collected from the human active layer. Lipid residues from hearth sediments were analyzed, and we simulated and assessed environmental extremity and route accessibility for the site. Dating results suggest that the stable sedimentary layers began to form around 10.96 ± 0.56 ka BP at the DQ site. Human activity at DQ as early as 9271 ± 143 cal a BP, making it the earliest reported Holocene site with accurate stratigraphic dating on the plateau hinterland to date. Hearths and lithic artifacts indicate that the DQ site was a frequently used seasonal hunting camp, where quality lithic raw material was obtained and microliths processed. Prehistoric humans occupying the site relied on non-ruminant terrestrial animals as food resources. The DQ site is ideally situated to serve as a transit station for hunter-gatherers as they migrated between high and low elevations. Ameliorating Holocene climate promoted prehistoric human exploration of more environmentally extreme areas.
