    Abstract: Background. The traditional Chinese medicines of Buyang Huanwu decoction (BYHW), Naoxintong capsule (NXT), and Yangyin Tongnao granules (YYTN) have excellent effects in preventing and treating cerebrovascular disease and are widely tolerated by patients. However, their effects on middle cerebral artery occlusion (MCAO) remain unknown. Methods. We evaluated gut microbiota alterations, the brain transcriptome, and nerve cell responses in rats with MCAO. Results. Our results showed that BYHW, NXT, and YYTN not only effectively improved the damaged state of blood vessels in rats and restored nerve function, but also improved survival. Additional experiments showed that treatment with BYHW, NXT, and YYTN regulated the intestinal microflora. Transcriptome analyses showed that BYHW, NXT, and YYTN modulated the transcriptome of rats with MCAO. The common mechanism of the three prescriptions for the treatment of cerebral ischemia may be related to the intestinal flora regulation of 60S ribosomal protein L18 (Rpl18), eukaryotic translation initiation factor 3 subunit, Ras homolog family member C, G protein subunit gamma 13 (Gng13), and Gng10 genes, among which Rpl18 is the most important. In addition, the three prescriptions had great specificity as anticerebral ischemia targets. Moreover, BYHW, NXT, and YYTN mitigated MCAO-induced hyperactivation of microglia and astrocytes. Conclusion. This study provides a foundation for further research on the mechanisms and treatment of IS. The results strongly suggest that key gut microbiota can be used to study functional genomics of brain, leading to novel discoveries about key genes involved in important biological processes.

    Abstract: A series of unconfined compression tests were performed to investigate the influence of wheat straws on the unconfined compression strength for saline-alkaline soils and saline-alkaline soils mixed with cement. In unconfined compression tests, 20 groups of soil specimens were prepared at five different percentages of wheat straws content (i.e., 0.0%, 0.1%, 0.15%, 0.2%, and 0.25% by weight of saline-alkaline soils) and four different percentages of cement content (i.e., 0%, 3%, 6%, and 9% by weight of saline-alkaline soils), and unconfined compression tests were carried out after 3-, 7-, 14-, 28-, and 56-day curing periods. Test results indicated that the inclusion of wheat straws within saline-alkaline soils and saline-alkaline soils mixed with cement leads to an increase in the unconfined compressive strength of specimens and also changed the brittle behavior to a more ductile one for specimens. In addition, based on the results from unconfined compression tests, a formula for predicting the unconfined compression strength of specimens related to cement content, wheat straw content, curing periods, etc., was determined, and comparing with the results from unconfined compression tests, it had higher precision in predicting the unconfined compression strength of specimens.

    Abstract: How moxibustion improves chronic heart failure is extremely complex and still unclear. This study aimed to explore whether moxibustion inhibits autophagy and reduces inflammation by regulating mTOR expression to induce myocardial protective effects and alleviate symptoms associated with chronic heart failure. Echocardiography was used to detect cardiac function and cardiac structure of rats, including heart rate (HR), left atrium diameter (LA), left ventricular diameter (LV), left ventricular posterior wall (LVPW), interventricular septum (IVS), ejection fraction (EF), and fractional shortening (FS). BNP and NT-pro BNP levels were measured by enzyme-linked immunosorbent assay (ELISA). Autophagy-associated protein (ATG) genes and mTOR were detected by PCR. The expression of mTOR and phosphorylated-mTOR was detected through western blotting of proteins from myocardial tissue samples. The left ventricular inflammatory response was detected by immunohistochemistry and included ICAM-1, VCAM-1, MMP-2, and MMP-9 expression. The relationship between autophagy and inflammation was analyzed by correlation analysis. The results from echocardiography and ELISA showed that moxibustion could significantly improve heart function and structure. Western blot and PCR results showed that moxibustion treatment elevated mTOR expression. Further, moxibustion could inhibit autophagy and regulate the expression of key autophagy-related genes, including Vps34, ATG3, ATG5, ATG7, ATG12, and ATG13. By contrast, rapamycin could partially reduce the effects of moxibustion. Immunohistochemistry results indicated that moxibustion could reduce myocardial inflammation. Moreover, there was a positive correlation between autophagy and inflammation. Moxibustion can protect cardiac function in rats with heart failure, possibly inhibiting excessive autophagy of cardiomyocytes and reducing inflammatory reactions through the elevation of mTOR expression.

    Abstract: Background. Diffuse large B-cell lymphoma (DLBCL) is a heterogeneous non-Hodgkin’s lymphoma with great clinical challenge. Machine learning (ML) has attracted substantial attention in diagnosis, prognosis, and treatment of diseases. This study is aimed at exploring the prognostic factors of DLBCL by ML. Methods. In total, 1211 DLBCL patients were retrieved from Huaihai Lymphoma Working Group (HHLWG). The least absolute shrinkage and selection operator (LASSO) and random forest algorithm were used to identify prognostic factors for the overall survival (OS) rate of DLBCL among twenty-five variables. Receiver operating characteristic (ROC) curve and decision curve analysis (DCA) were utilized to compare the predictive performance and clinical effectiveness of the two models, respectively. Results. The median follow-up time was 43.4 months, and the 5-year OS was 58.5%. The LASSO model achieved an Area under the curve (AUC) of 75.8% for the prognosis of DLBCL, which was higher than that of the random forest model (AUC: 71.6%). DCA analysis also revealed that the LASSO model could augment net benefits and exhibited a wider range of threshold probabilities by risk stratification than the random forest model. In addition, multivariable analysis demonstrated that age, white blood cell count, hemoglobin, central nervous system involvement, gender, and Ann Arbor stage were independent prognostic factors for DLBCL. The LASSO model showed better discrimination of outcomes compared with the IPI and NCCN-IPI models and identified three groups of patients: low risk, high-intermediate risk, and high risk. Conclusions. The prognostic model of DLBCL based on the LASSO regression was more accurate than the random forest, IPI, and NCCN-IPI models.

    Abstract: As an emerging data source, feature identification based on GPS trajectory data has become a hot issue in the field of data mining and freight management. A method of trajectory data extraction, classification, and visualization based on stay points was proposed in this paper to analyze the operation characteristics of heavy trucks from the perspectives of intracity transportation, intraprovincial transportation, and interprovincial transportation. The GPS trajectory data of heavy trucks in Sichuan Province in March 2019 were taken as an example to analyze the operation characteristics. The results show that the heavy trucks in Sichuan Province are mainly transported within the province, and the freight efficiency is slightly better than the average level of the national freight trucks in the same period, failing to give full play to the advantages of long transport distance. The manufacturing industry is the main service object of heavy trucks, and the vehicles engaged in transportation within the province are more dependent on logistics enterprises and their ancillary facilities. The north-south longitudinal line and east-west horizontal line are the main interprovincial transport channels, and the provincial and municipal transport is mainly concentrated in some urban trunk lines, ring lines, and express routes. The proposed technical method can describe the operating characteristics of freight trucks from the perspective of microscopic and service market, not only to guide the layout of highway freight yards, logistics parks, and logistics hubs and the determination of service functions but also to provide a reference basis for freight management-related departments and drivers to formulate transportation plans and establish freight information platforms to improve freight efficiency and safety.

    Abstract: To mitigate the environmental impact induced by CO2 emissions and nonrenewable resource consumption, which are typically associated with Portland cement production, ground granulated blast furnace slags (GGBSs) are usually added to the cement. In this study, the stabilisation effect of alkali-activated GGBS on saline soil and the hydration products of alkali-activated GGBS were investigated by unconfined compressive strength tests and scanning electron microscopy, respectively. The results show that Ca(OH)2 and NaOH as alkaline activators for GGBS significantly improve the unconfined compressive strength of saline soils. This strength is also enhanced by Na2SO4; however, the increase is considerably less than that provided by Ca(OH)2 and NaOH. In contrast, Na2CO3 is not a suitable alkaline activator for GGBS and has no significant effect on the unconfined compressive strength of saline soils. The study results further show that the morphology of hydration products varies because of the different alkaline activators involved in the hydration reaction with GGBS in saline soils.

    Abstract: Background . Numerous studies have highlighted that long noncoding RNA (lncRNA) can indirectly regulate the expression of mRNAs by binding to microRNA (miRNA). LncRNA-associated ceRNA networks play a vital role in the initiation and progression of several pathological mechanisms. However, the lncRNA-miRNA-mRNA ceRNA network in endothelial cells under cyclic stretch is seldom studied. Methods . The miRNA, mRNA, and lncRNA expression profiles of 6 human umbilical vein endothelial cells (HUVECs) under circumferential stress were obtained by next-generation sequencing (NGS). We identified the differential expression of miRNAs, mRNAs, and lncRNAs using the R software package GDCRNATools. Cytoscape was adopted to construct a lncRNA-miRNA-mRNA ceRNA network. In addition, through GO and KEGG pathway annotations, we analyzed gene functions and their related pathways. We also adopted ELISA and TUNEL to investigate the effect of si-NEAT1 on endothelial inflammation and apoptosis. Results . We recognized a total of 32978 lncRNAs, 1046 miRNAs, and 31958 mRNAs in 6 samples; among them, 155 different expressed lncRNAs, 74 different expressed miRNAs, and 960 different mRNAs were adopted. Based on the established theory, the ceRNA network was composed of 13 lncRNAs, 44 miRNAs, and 115 mRNAs. We constructed and visualized a lncRNA-miRNA-mRNA network, and the top 20 nodes are identified after calculating their degrees. The nodes with most degrees in three kinds of RNAs are hsa-miR-4739, NEAT1, and MAP3K2. Functional analysis showed that different biological processes enriched in biological regulation, response to stimulus and cell communication. Pathway analysis was mainly enriched in longevity regulating, cell cycle, mTOR, and FoxO signaling pathway. Circumferential stress can significantly downregulate NEAT1, and after transducing si-NEAT1 for 24 h, inflammatory cytokine IL-6 and MCP-1 were significantly increased; furthermore, fewer TUNEL-positive cells were found in the si-NEAT1 treated group. Conclusions . The establishing of a ceRNA network can help further understand the mechanism of vein graft failure. Our data demonstrated that NEAT1 may be a core factor among the mechanical stress factors and that cyclic stress can significantly reduce expression of NEAT1, give rise to inflammation in the early stage of endothelial dysfunction, and promote EC apoptosis, which may play an essential role in vein graft failure.

    Abstract: Liver cirrhosis is a common chronic disease in China. The effect of modified Xianglian Pingwei powder plus Western medicine in the treatment of liver cirrhosis and positive small intestinal bacterial overgrowth is promising. Totally, 100 patients with liver cirrhosis and positive intestinal bacterial overgrowth in Cangzhou Central Hospital from February 2020 to February 2021 were enrolled and randomized via the random number table method at a ratio of 1 : 1 into the study group and control group. The control group received glutathione and levofloxacin hydrochloride, and the study group received Xianglian Pingwei powder plus glutathione and levofloxacin hydrochloride. The traditional Chinese medicine (TCM) syndrome scores, serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and total bilirubin (TBIL) levels of the two groups were decreased after treatment with lower results in the study group. Xianglian Pingwei powder plus glutathione and levofloxacin hydrochloride was associated with a significantly lower positive rate of small intestine bacterial growth, serum endotoxin level, and peripheral blood toll-like receptor 2 (TLR2) and TRL4 levels versus glutathione and levofloxacin hydrochloride. The combined medication achieved a higher efficacy (90.00%) versus glutathione and levofloxacin hydrochloride (66.00%). The two groups experienced similar safety. Xianglian Pingwei powder plus glutathione and levofloxacin hydrochloride achieved significant benefits of clinical efficacy with a high safety profile in patients with liver cirrhosis versus glutathione and levofloxacin hydrochloride.

    Abstract: Deep learning has achieved good results in the crack detection of roads and bridges. However, the timber structures of ancient architecture have strong orthotropic anisotropy and complex microscopic structures, and the law of cracks development is extremely complex. The image data has a large proportion of pixels, which is obviously different from the background gray value, and there is timber grain noise, thus the existing methods cannot accurately extract the complex texture contour feature of cracks. In previous studies, we have verified that YOLO v5s is effective in crack detection in timber structures of ancient architecture. However, there are many different versions of YOLO series models. In order to find a better algorithm, this paper mainly adopts three models including YOLO v3, YOLO v4s-mish, and YOLO v5s to detect cracks in the timber structures of ancient architecture, and compares and analyzes the advantages and disadvantages of the three models. In the comparing process, we mainly have discussed the index performance of the three models in terms of training time, loss function, recall rate, and mAP value. We have summarized and analyzed the advantages and disadvantages of the three models in cracks detection of the timber structures of ancient architecture, and concluded the comparing results of the three models in cracks detection based on experiments. We published the first picture data set of cracks in timber structures of ancient architecture, and applied YOLO model in the intelligent identification field of cracks in timber structures of ancient architecture for the first time, which opened up a new idea for the intelligent operation and maintenance of the timber structures of ancient architecture.

    Abstract: Objective. The objective is to explore the clinical effect of compound lactic acid bacteria capsules on the small intestinal bacterial overgrowth (SIBO) in patients with depression and diabetes. Methods. From January 2020 to January 2021, 60 SIBO patients with depression and diabetes in our hospital were selected and randomized into observation group (compound lactic acid bacteria capsules combined with escitalopram) and control group (Escitalopram) according to the odd and even numbers, 30 cases in each group. The two groups were compared in terms of SAS, SDS, levels of inflammatory factors, immune function, fasting plasma glucose (FPG), treatment effect, and the incidence of adverse reactions. Results. Both self-rating anxiety scale (SAS) and self-rating depression scale (SDS) scores in both groups showed a decline after treatment ( P 〈 0.05 ), and the reduction was more significant in the observation group ( t = 10.047 , 17.862, all P ≤ 0.001 ). Both IL-2 and TNF-α in both groups showed a decline after treatment ( P 〈 0.05 ), and the reduction was more greater in the observation group in relative to the control group ( P 〈 0.05 ). CD3+ and CD+4 in both groups showed an increase after treatment ( P 〈 0.05 ), and the increase was more greater in the observation group as compared to the control group ( P 〈 0.05 ). After treatment, the FPG levels of patients in both groups showed a decline ( P 〈 0.05 ), and the reduction of FPG levels was more significant in the observation group than that in the control group ( t = 3.948 , P ≤ 0.001 ). The control group experienced a remarkably higher incidence of adverse reactions. Conclusion. The compound lactic acid bacteria capsule is a boon for SIBO patients with depression and diabetes. It can mitigate depression symptoms, improve immune function, reduce the level of inflammatory factors, and lower the FPG levels, along with fewer adverse reactions and robust effects.
