    Beschreibung: We have performed global seismic wave propagation calculations using the spectral element method, which is a type of finite element method, with an accuracy of 1.6 seconds by using the Earth Simulator (ES4) system. In this calculation, the theoretical seismic waveform propagating globe was calculated by dividing it into 244.7 billion grid points. The parameters NEX and NPROC indicating the division of the spectral element method in this case are 2656 and 83, respectively, and the total number of cores used in the calculation is 41,334. For this scale of calculation, it took about 30 minutes CPU time to calculate the mesh and 4 hours 40 minutes CPU time to calculate the theoretical seismic waveform for 23 minutes. The size of the mesh is about 41 Tbyte. The Earth's internal structure model used in the calculation is transversly isotropic PREM (Dziewonski and Anderson, 1981) for the radial symmetric structure model, and s362ani (Kustowski et al., 2006) for the mantle three-dimensional structure. We analyse the basal outer core boundary (BOC) with antipodal waveform data in the distance range 179.0°–180° to test the hypothesis whether propagation at the BOC is commensurate with diffraction and/or refraction. We try thin LVZ models. Synthetic modeling of the thin, low velocity structures requires higher resolution (1.6 sec) parametrization to achieve necessary detail. We synthesized the antipodal data at the Qiongzhong (QIZ) station in China due to the April 17, 2009 earthquake in northern Chile (Mw6.1). We discuss the results in our presenation.

    Notizen: Eleven haemophilia treatment centres in the United States collaborated in the Hemophilia Behavioural Intervention Evaluation Projects (HBIEP) to develop theory-based interventions to reduce the risk of HIV transmission from seropositive adolescents and young adults with haemophilia. While the Transtheoretical Model of Behaviour Change and the Theory of Reasoned Action provided the theoretical underpinnings, the exact form in which these theories would be applied depended on developmental research. This paper presents the various phases of the process to develop the theory based interventions: literature review, qualitative interviews, quantitative surveys, a provider survey, a materials review, and the actual planning. All or portions of this process could be applied to the development of interventions for many behaviour-change projects. A description of the HBIEP interventions is also provided.

    Notizen: Summary.  The goal of the project was to develop and evaluate theory-based interventions designed to change sexual behaviour and promote safer sex practices of HIV seropositive young men and adolescents with haemophilia to prevent transmission to sexual partners and offspring. Safer sex was defined as abstinence, consistent condom use, or ‘outercourse’ (intimate, non-intercourse sexual behaviour). This project utilized the Transtheoretical Model developed by Prochaska and DiClemente, which describes behaviour change as an incremental, stage-based process. The 1-year intervention protocol consisted of two individual sessions and two peer-centred activities. One hundred and four adolescents, residing in 22 states, participated. Pre- and post-intervention evaluations were conducted to measure stage progression for participants. The number who were in the action or maintenance stage of change for safer sex was significantly greater at post-test than at pre-test (79 vs. 62%, P 〈 0.0001). Participants also reported an increased use of outercourse. In addition, significant increases in self-efficacy and knowledge regarding safer sex were demonstrated. Following these stage-based interventions, participants were significantly more likely to be engaging in safer sex behaviours than they were previously. These intervention activities can be adapted for use with other adolescent populations and for other behaviour change goals in adolescents with haemophilia.

    Notizen: A dual enzyme disaggregation method using collagenase and then trypsin was developed that allowed the reproducible initiation of primary cultures from Atlantic salmon Salmo salar gills. Cultures had both epithelial and fibroblast morphology and persisted for an average of 20 passages. Growth was dependent upon a minimum concentration of 5% foetal calf serum (FCS) for fibroblasts and 10% FCS for epithelial cells. Growth was mostly independent of substrate, although epithelial cells showed increased growth on type I collagen gels. Matrigel™ cell culture substrate produced reduced growth of fibroblasts and did not benefit epithelial cell growth. Epithelial cells reacted with monoclonal antibodies (MAbs) against mammalian cytokeratins, and fibroblast cells reacted with MAbs against mammalian fibronectin and type I collagen. The method also produced two long-term cultures: one epithelial and one fibroblast that have been designated RGE-2 and RGF respectively.

    Notizen: : A pilot plant steam treatment system was developed to reduce L. monocytogenes contamination on exterior surfaces of King Salmon (Oncorhynchus tshawytscha prior to processing. Numbers of surviving L. monocytogenes cells on the surface were determined using an enrichment-based Most Probable Number (MPN) technique. Inoculated L. monocytogenes cells were not recovered from the surface of salmon after exposure to the steam treatment for 8 sec. A 4-log reduction in L. monocytogenes numbers was obtained. Treated salmon could be processed into a high-yielding, high-quality cold smoked product. An in-plant system was subsequently shown to reduce “naturally” occurring L. monocytogenes numbers and produce a high quality final product.

    Notizen: Neoparamoeba sp., including the putative aetiological agent of amoebic gill disease in cultured fish (N. pemaquidensis), were incubated in vitro with an Atlantic salmon gill epithelium (RGE-2) cell line. Proliferation by the amoeba population was dependent upon culture osmolarity; no growth occurred at 330 mm kg−1 but a sixfold increase was observed at 1000 mm kg−1. At 780 mm kg−1 there was a fourfold increase in the amoeba population but a concurrent decrease in RGE-2 cell density that was significantly greater than that caused by the high culture osmolarity alone. This apparent cytopathic effect (CPE) developed rapidly and resulted in complete cytolysis of the monolayer in 5 days. CPE occurred in multiple foci and presented as cell vacuolation, rounding and clumping, and the rapid clearance of large areas of the cell monolayer. The possibility that CPE is because of the presence of Neoparamoeba sp. derived cytolytic products is discussed in the context of the pathology of the disease in vivo and the occurrence of secreted cytopathogenic compounds in other amoeba species.

    Notizen: Previous studies have indicated that Atlantic salmon, Salmo salar L., affected by amoebic gill disease (AGD) are resistant to re-infection. These observations were based upon a comparison of gross gill lesion abundance between previously infected and naïve control fish. Anecdotal evidence from Atlantic salmon farms in southern Tasmania suggests that previous infection does not protect against AGD as indicated by a lack of temporal change in freshwater bathing intervals. Experiments were conducted to determine if previous infection of Atlantic salmon with Neoparamoeba sp. would provide protection against challenge and elucidate the immunological basis of any protection. Atlantic salmon were infected with Neoparamoeba sp. for 12 days then treated with a 4-h freshwater bath. Fish were separated into two groups and maintained in either sea water or fresh water for 6 weeks. Fish were then transferred to one tank with a naïve control group and challenged with Neoparamoeba sp. Fish kept in sea water had lower mortality rates compared with first time exposed and freshwater maintained fish, however, these data are believed to be biased by ongoing mortalities during the seawater maintenance phase. Phagocyte function decreased over exposure time and freshwater maintained fish demonstrated an increased ability to mount a specific immune response. These results suggest that under the challenge conditions herein described, antigen exposure via infection does not induce protection to subsequent AGD.
