    Notes: Abstract An attempt has been made to examine the properties of the title divalent cation impurity - cation vacancy dipole as well as the adsorptivity and diffusion of atomic hydrogen over the LiH (001) surface using ab initio methods of molecular electronic structure calculations. The LiH crystal surface was simulated and quantum clusters of variable size were surrounded by point charges to examine the I-V dipole orientation, cation vacancy migration and overlap effects. The effects of introducing an I-V dipole on the nature of adsorbate-substrate interactions and diffusion of an H atom over the surface were also examined. The results confirm that the (100) orientation of the I-V dipole is energetically more favorable than the (110) orientation and that the cation vacancy migrates without activation energy barriers. The I-V dipole enhances the adsorptivity of atomic hydrogen by ca. 4.96 eV, changes the nature of adsorption from physisorption to chemisorption and introduces an activation energy barrier to H diffusion over the surface. As the I-V dipole is introduced, the HOMO and LUMO levels of the substrate shift to higher energies, the band gap becomes narrower and the atomic charges on the adsorbed H increases. This change in the electronic structure makes the adsorptivity process, through the charge transfer from the H 1s singly occupied AO to the conduction band and from the valence band to the H 1s singly occupied AO more facile.

    Notes: Abstract.  Oxytetracycline hydrochloride-selective electrodes of both the coated wire and the conventional polymer membrane types based on oxytetracyclinium phosphotungstate and phosphomolybdate have been prepared. A Nernstian response is shown by these electrodes within 1.0×10−6–1.0×10−2 M concentration ranges depending on the type of electrode. The response is unaffected by the change of pH over the range 4–11. The standard electrode potentials, E°, were determined at different temperatures and used to calculate the isothermal temperature coefficients of the electrodes. The electrodes show good selectivity to oxytetracycline hydrochloride with respect to many inorganic cations, sugars and amino-acids. Oxytetracycline hydrochloride is determined successfully in pure solutions and in pharmaceutical preparations using calibration by standard addition and potentiometric titration. A regeneration process for the exhausted electrodes has been developed.

    Notes: Abstract   Object: We present a case of an asymptomatic and neurologically normal 6-year-old girl who was noted to have a gross spinal abnormality. Methods: Neuroimaging demonstrates gross thoraco-lumbar spine duplication, which is at the severe end of the split cord malformation (SCM) group of congenital abnormalities. Despite the position of the neural structures, the clinical condition has so far not suggested that any surgical intervention is indicated, though the girl’s later growth spurts may still unmask features of a tethered spinal cord. Conclusions: We present a brief review of the literature dealing with theories of embryogenesis relating to SCM, the common clinical and radiological features, and finally the surgical options available.

    Notes: Abstract Botulinum neurotoxins, the most potent of all toxins, induce lethal neuromuscular paralysis by inhibiting exocytosis at the neuromuscular junction. The light chains (LC) of these dichain neurotoxins are a new class of zinc-endopeptidases that specifically cleave the synaptosomal proteins, SNAP-25, VAMP, or syntaxin at discrete sites. To facilitate the structural and functional characterization of these unique endopeptidases, we constructed a synthetic gene for the LC of the botulinum neurotoxin serotype A (BoNT/A), overexpressed it in Escherichia coli, and purified the gene product from inclusion bodies. Our procedure can provide 1.1 g of the LC from 1 L of culture. The LC product was stable in solution at 4°C for at least 6 months. This rBoNT/A LC was proteolytically active, specifically cleaving the Glu-Arg bond in a 17-residue synthetic peptide of SNAP-25, the reported cleavage site of BoNT/A. Its calculated catalytic efficiency k cat/K m was higher than that reported for the native BoNT/A dichain. Treating the rBoNT/A LC with mercuric compounds completely abolished its activity, most probably by modifying the cysteine-164 residue located in the vicinity of the active site. About 70% activity of the LC was restored by adding Zn2+ to a Zn2+-free, apo-LC preparation. The LC was nontoxic to mice and failed to elicit neutralizing epitope(s) when the animals were vaccinated with this protein. In addition, injecting rBoNT/A LC into sea urchin eggs inhibited exocytosis-dependent plasma membrane resealing. For the first time, results of our study make available a large amount of the biologically active toxin fragment in a soluble and stable form.
