    Notes: The classical binary nucleation theory is considered. It is shown that as a consequence of the Gibbs–Duhem equation and the Gibbs adsorption isotherm, an equation emerges which can be used together with the interior (bulk) mole fraction and radius of the critical cluster given by the theory to calculate the total numbers of molecules of both species in the cluster. Accordingly the overall (average) composition of the critical cluster is determined. Furthermore, it is noted that it is possible to use this equation to determine the interior (bulk) mole fraction of any sufficiently large spherical cluster with known total numbers of molecules, which allows the construction of a free energy surface within the framework of the classical nucleation theory. However, numerical calculations for the water–ethanol system result in strange predictions at some typical experimental conditions. Possible reasons for the unsuccessful predictions by the classical theory are discussed.

    Notes: Abstract: The effect of the hydrolysis-resistant GTP analogs, guanosine 5′-O-(3-thiotriphosphate) (GTPγS) and guanylyl imidodiphosphate (GMPPNP), on norepinephrine (NE) secretion from digitonin-permeabilized rat pheochromocytoma cells, PC12, was examined. Although secretion in the presence of saturating Ca2+ (10 μM) was not affected by GTP7S or GMPPNP, secretion in the absence of Ca2+ was stimulated by these GTP analogs. Secretion induced by saturating concentrations of GTPγS or GMPPNP was approximately 80% of that induced by 10 μM Ca2+. Half-maximum stimulation was induced by 30 μM GTPγS or GMPPNP. Both Ca2+-stimulated and GTPγS-stimulated secretion were ATP dependent and inhibited by N-ethylmaleimide. The GTPγS-stimulated secretion of NE from permeabilized PC12 cells does not appear to result from either the release of Ca2+ or the activation of protein kinase C. Activation of protein kinase C by pretreatment of intact cells with 12-O-tetradecanoyl-phorbol 13-acetate caused a 50% increase in both Ca2+-stimulated and GTP7S-stimulated secretion. Cholera and pertussis toxins did not affect Ca2+-stimulated or GTPγS-stim-ulated NE secretion. Guanosine 5′-O-(2-thiodiphosphate) (GDPβS) and GTP inhibited GTPγS-stimulated secretion but not Ca2+-stimulated secretion. The inability of GDPβS to inhibit Ca2+-stimulated secretion indicates that the process affected by GTPγS is not an essential step in the Ca2+-stimulated pathway.

    Notes: Abstract— Secretion of catecholamines by rat PC12 cells is strongly stimulated by extracellular ATP via a P2-type pur-inergic receptor. ATP-induced norepinephrine release was inhibited 80% when extracellular Ca2+ was absent. Only four nucleotides, ATP, ATPγS, benzoylbenzoyl ATP (BzATP), and 2-methylthio-ATP, gave substantial stimulation of norepinephrine release from PC12 cells. ATP-induced secretion was inhibited by Mg2+, and this inhibition was overcome by the addition of excess ATP suggesting that ATP4-was the active ligand. ATP-induced secretion of catecholamine release was enhanced by treatment of cells with pertussis toxin or 12-O-tetradecanoylphorbol 13-acetate. The stimulatory effects of 12-O-tetradecanoyl-phorbol 13-acetate and pertussis toxin on norepinephrine release were additive. After brief exposure of intact cells to the photoaffinity analog, [α-32P]BzATP, two major proteins of 44 and 50 kDa and a minor protein of 97 kDa were labeled. An excess of ATP-γS and BzATP but not GTP blocked labeling of the proteins by [32P]BzATP. Labeling of the 50-kDa protein was more sensitive to competition by 2-methylthio-ATP than the other labeled proteins, suggesting that the 50-kDa protein represents the P2 receptor responsible for ATP-stimulated secretion in these cells.

    Description: The latest version of the Soil and Water Assessment Tool (SWAT+) features several improvements compared with previous versions of the model, for example, the definition of landscape units that allow for a better representation of spatio‐temporal dynamics. To evaluate the new model capabilities in lowland catchments characterized by near‐surface groundwater tables and extensive tile drainage, we assess the performance of two SWAT+ model setups in comparison to a setup based on a previous SWAT model version (SWAT3S with a modified three groundwater storage model) in the Kielstau catchment in Northern Germany. The Kielstau catchment has an area of about 50 km2, is dominated by agricultural land use, and has been thoroughly monitored since 2005. In both SWAT+ setups, the catchment is divided into upland areas and floodplains, but in the first SWAT+ model setup, runoff from the hydrologic response units is summed up at landscape unit level and added directly to the stream. In the second SWAT+ model setup, runoff is routed across the landscape before it reaches the streams. Model results are compared with regard to (i) model performance for stream flow at the outlet of the catchment and (ii) aggregated as well as temporally and spatially distributed water balance components. All three model setups show a very good performance at the catchment outlet. In comparison to a previous version of the SWAT model that produced more groundwater flow, the SWAT+ model produced more tile drainage flow and surface runoff. Results from the new SWAT+ model confirm that the representation of routing processes from uplands to floodplains in the model further improved the representation of hydrological processes. Particularly, the stronger spatial heterogeneity that can be related to characteristics of the landscape, is very promising for a better understanding and model representation of hydrological fluxes in lowland areas. The outcomes of this study are expected to further prove the applicability of SWAT+ and provide useful information for future model development.
    Description: The model performance of all three model setups was very good, but the SWAT+ model setup with runoff routing between landscape units performed best. Moreover, the SWAT+ model applications predicted a greater spatial heterogeneity of the water balance components. The representation of hydrological fluxes particularly with regard to groundwater flow, surface runoff, and tile drainage flow differed considerably between the SWAT and SWAT+ model setups.
