    Abstract: Therapeutic responses of non-small cell lung cancer (NSCLC) to epidermal growth factor receptor (EGFR) - tyrosine kinase inhibitors (TKIs) are known to be associated with EGFR mutations. However, a proportion of NSCLCs carrying EGFR mutations still progress on EGFR-TKI underlining the imperfect correlation. Structure-function-based approaches have recently been reported to perform better in retrospectively predicting patient outcomes following EGFR-TKI treatment than exon-based method. Here, we develop a multicolor fluorescence-activated cell sorting (FACS) with an EGFR-TKI-based fluorogenic probe (HX103) to profile active-EGFR in tumors. HX103-based FACS shows an overall agreement with gene mutations of 82.6%, sensitivity of 81.8% and specificity of 83.3% for discriminating EGFR -activating mutations from wild-type in surgical specimens from NSCLC patients. We then translate HX103 to the clinical studies for prediction of EGFR-TKI sensitivity. When integrating computed tomography imaging with HX103-based FACS, we find a high correlation between EGFR-TKI therapy response and probe labeling. These studies demonstrate HX103-based FACS provides a high predictive performance for response to EGFR-TKI, suggesting the potential utility of an EGFR-TKI-based probe in precision medicine trials to stratify NSCLC patients for EGFR-TKI treatment.

    Abstract: Projecting mitigations of carbon neutrality from individual countries in relation to future global warming is of great importance for depicting national climate responsibility but is poorly quantified. Here, we show that China’s carbon neutrality (CNCN) can individually mitigate global warming by 0.48 °C and 0.40 °C, which account for 14% and 9% of the global warming over the long term under the shared socioeconomic pathway (SSP) 3-7.0 and 5-8.5 scenarios, respectively. Further incorporating changes in CH 4 and N 2 O emissions in association with CNCN together will alleviate global warming by 0.21 °C and 0.32 °C for SSP1-2.6 and SSP2-4.5 over the long term, and even by 0.18 °C for SSP2-4.5 over the mid-term, but no significant impacts are shown for all SSPs in the near term. Divergent responses in alleviated warming are seen at regional scales. The results provide a useful reference for the global stocktake, which assesses the collective progress towards the climate goals of the Paris Agreement.

    Abstract: The aim of this study was to explore the current status of the anaesthesia provision, infrastructure and resources in the Heilongjiang Province, China. Design A cross-sectional observational study of hospitals, anaesthesiologists, assistant anaesthesiologists and anaesthetic nurses in the Heilongjiang Province. Setting All hospitals in the Heilongjiang Province. Participants The hospitals, anaesthesiologists (attending physicians, associate chief physicians and chief physicians), assistant anaesthesiologists (licenced assistant physicians, resident physicians and other trainees) and anaesthetic nurses. Main outcome measures Standard descriptive statistics (percentages and numbers) were used to summarise the data. Results The investigation involved 1123 hospitals, 405 of these hospitals had anaesthesiology departments (36.06%). There were 2406 anaesthesiologists, 175 assistant anaesthesiologists and 409 anaesthetic nurses. The proportion of anaesthesiologists was 56.60% in tertiary hospitals, 40.15% in secondary hospitals and 3.25% in primary hospitals and ungraded hospitals, respectively. Anaesthesiologists were present in 91.20% of public hospitals and 8.80% of private hospitals. Anaesthesiologists were present in 83.55% general hospitals and 16.45% of specialised hospitals. The Heilongjiang Province has a total of 2041 operating rooms and 543 beds in recovery rooms. The number of anaesthesia cases per capita per year was 326.86. The percentages of anaesthesiologists’ age ≥46, 36–45, 25–35 and 〈 25 are 24.03%, 41.80%, 33.91% and 0.27%, respectively. The proportions of resident physicians and attending physicians were 60.87%, and the proportions of associate chief physicians and chief physicians were 39.13%. The proportions of anaesthesiologists working 〉 12 hours, 10 hours≤time≤12 hours, 8 hours≤time 〈 10 hours and 〈 8 hours were 0.55%, 22.04%, 64.30% and 13.11%, respectively. Conclusions The present study demonstrated for the first time that the proportion of anaesthesiologists in the Heilongjiang Province, China, is still insufficient. The structure of anaesthesiologists needs to be optimised.

    Abstract: Understanding immune cell phenotypes in the tumor microenvironment (TME) is essential for explaining and predicting progression of non-small cell lung cancer (NSCLC) and its response to immunotherapy. Here we describe the single-cell transcriptomics of CD45 + immune cells from tumors, normal tissues and blood of NSCLC patients. We identified three clusters of immune cells exerting immunosuppressive effects: CD8 + T cells with exhausted phenotype, tumor-associated macrophages (TAMs) with a pro-inflammatory M2 phenotype, and regulatory B cells (B regs) with tumor-promoting characteristics. We identified genes that may be mediating T cell phenotypes, including the transcription factors ONECUT2 and ETV4 in exhausted CD8 + T cells, TIGIT and CTL4 high expression in regulatory T cells. Our results highlight the heterogeneity of CD45 + immune cells in the TME and provide testable hypotheses about the cell types and genes that define the TME.

    Abstract: Residues of neonicotinoid pesticides have potential risks to food, environmental and biological safety. In this study, the hapten toward imidacloprid was adopted to gain antibodies. After molecular modeling, the electrostatic potentials of eight commonly-used neonicotinoid pesticides were individually calculated to analyze the structural similarity. Two representative compounds (imidacloprid and acetamiprid) with moderate similarity were rationally selected for hybridoma screening. Using this strategy, four clones of broad-specific monoclonal antibodies (mAbs) against multiple neonicotinoids were obtained, and the clone 6F11 exhibited the broadest spectrum to six neonicotinoid pesticides and two metabolites, with half-maximal inhibitory concentrations (IC50) ranging from 0.20 to 5.92 ng/mL. Then, the novel antibody gene was sequenced and successfully expressed in full-length IgG form using mammalian cells. Based on the sensitive recombinant antibody, a gold lateral-flow immunosensing strip assay was developed and it was qualified for rapid detection of imidacloprid, clothianidin or imidaclothiz residues in food samples.

    Abstract: Land-atmosphere interactions play an important role in the changes of extreme climates, especially in hot spots of land-atmosphere coupling. One of the linkages in land-atmosphere interactions is the coupling between air temperature and surface energy fluxes associated with soil moisture variability, vegetation change, and human water/land management. However, existing studies on the coupling between hot extreme and surface energy fluxes are mainly based on the parameterized solution of climate model, which might not dynamically reflect all changes in the surface energy partitioning due to the effects of vegetation physiological control and human water/land management. In this study, for the first time, we used daily weather observations to identify hot spots where the daily hot extreme (i.e., the 99th percentile of maximum temperature, Tq99th) rises faster than local mean temperature (Tmean) during 1975–2017. Furthermore, we analyzed the relationship between the trends in temperature hot extreme relative to local average (ΔTq99th/ΔTmean) and the trends in evaporative fraction (ΔEF), i.e., the ratio of latent heat flux to surface available energy, using long-term latent and sensible heat fluxes which are informed by atmospheric boundary layer theory, machine learning, and ground-based observations of flux towers and weather stations. Hot spots of increase in ΔTq99th/ΔTmean are identified to be Europe, southwestern North America, Northeast Asia, and Southern Africa. The detected significant negative correlations between ΔEF and ΔTq99th/ΔTmean suggested that the hotspot regions are typically affected by annual/summer surface dryness. Our observation-driven findings have great implications in providing realistic observational evidences for the extreme climate change accelerated by surface energy partitioning.

    Abstract: Atlantic multidecadal variability (AMV) is a cornerstone for decadal prediction and profoundly influences regional and global climate variability, yet its fundamental drivers remain an issue for debate. Studies suggest that external forcing may have affected AMV during the Little Ice Age (AD 1400–1860). However, the detailed mechanism remains elusive, and the AMV’s centennial to millennial variations over the past 2000 years have not yet been explored. We first show that proxy-data reconstructions and paleo-data assimilations suggest a significant 60-yr AMV during AD 1250–1860 but not during AD 1–1249. We then conducted a suite of experiments with the Community Earth System Model (CESM) to unravel the causes of the changing AMV property. The simulation results under all external forcings match the reconstructions reasonably well. We find that the significant 60-yr AMV during 1250–1860 arises predominantly from the volcano forcing variability. During the period 1–1249, the average volcanic eruption intensity is about half of the 1250–1860 intensity, and a 20–40-yr internal variability dominates the AMV. The volcanic radiative forcing during 1250–1860 amplifies AMV and shifts the internal variability peak from 20–40 years to 60 years. The volcano forcing prolongs AMV periodicity by sustaining Arctic cooling, delaying subpolar sea ice melting and atmospheric feedback to reduce surface evaporation. These slow-response processes over the subpolar North Atlantic results in a persisting reduction of sea surface salinity, weakening the Atlantic overturning circulation, and warm water transport from the subtropical North Atlantic. The results reveal the cause of the nonstationary AMV over the past two millennia and shed light on the AMV’s response to external forcing. Significance Statement AMV plays an important role in the regional and global climate variability. The purpose of this study is to better understand the secular change of AMV during the past 2000 years and its response to the external forcing. Proxy data and model simulation consistently show a significant 60-yr AMV during AD 1250–1860 that is absent during AD 1–1249. Active volcanic eruptions during 1250–1860 amplify the AMV and shift its intrinsic 20–40-yr to a prominent 60-yr variance peak. Volcanoes prolong AMV periodicity by sustaining Arctic cooling, delaying subpolar sea ice melting, reducing evaporation, and increasing surface salinity. These results help us better understand nonstationary AMV and highlight the role of external forcing over the past two millennia.

    Abstract: Background. Complications after colon surgery are a major obstacle to postoperative recovery. The purpose of this study was to investigate the effect of electroacupuncture (EA) at Zusanli (ST36) on gastrointestinal motility in rats after colonic anastomosis and the mechanism of transient receptor potential vanillin 1 (TRPV1) channel in regulating gastrointestinal motility, pain, and inflammation. Methods. The rats were randomly divided into six groups, including the control, model, EA, sham-EA, capsaicin, and capsaicin+EA groups, with preoperative capsaicin pretreatment and EA treatment at ST36 acupoint after surgery. Rats were treated using EA at ST36 or sham acupoints after surgery for 5 days. Capsaicin was intraperitoneally injected into rats 3 hours before surgery. Gastrointestinal motility was assessed by measuring the gastric residue, small intestinal propulsion in vivo, contractile tension, and frequency of isolated muscle strips in vitro. The mechanical withdrawal threshold (MWT) of abdominal incision skin and spontaneous nociceptive scores were observed and recorded in rats after colon anastomosis. The expressions of TRPV1, substance P (SP), neurokinin 1 (NK1) receptor, nuclear factor kappa-B (NF-κB), interleukin- (IL-) 6, L-1β, and tumor necrosis factor- (TNF-) α were determined. Results. Compared with the model group, electroacupuncture at ST36 point could significantly reduce the residual rate of stomach in rats after operation and increase the propulsive force of the small intestine and the contraction tension of the isolated smooth muscle. Electroacupuncture also increased postoperative day 3 MWT values and decreased postoperative spontaneous nociception scores. In addition, electroacupuncture treatment downregulated the expressions of IL-6, IL-1β, TNF-α, TRPV1, NF-κB, SP, and NK1 receptors in the colon tissue of rats after colonic anastomosis. Conclusions. Our study showed that electroacupuncture at ST36 acupoint could improve gastrointestinal motility in rats after colonic anastomosis and relieve intestinal inflammation and pain. The mechanism may be to inhibit the activation of NF-κB and SP/NK1 receptor signaling pathways by inhibiting TRPV1.
