    Geochemical and paleomagnetic variations in basalts from the Wendell Regional Aquifer Systems Analysis (RASA) drill core: Evidence for magma recharge and assimilation-fractional crystallization from the central Snake River Plain, Idaho (2013)

    Description: The temporal and magmatic evolution of central Snake River Plain (SRP; Idaho, USA) olivine tholeiites erupted within the past 4 m.y. is evaluated here. This investigation correlates and merges both geochemical and paleomagnetic measurements to constrain the volcanic history recovered from the 340 m Regional Aquifer Systems Analysis (RASA) test well located near Wendell, Idaho. Only a handful of studies have accomplished this task of shedding light on the chemical stratigraphy of the SRP and the petrogenesis of basalts with depth, and therefore through time. Paleomagnetic relationships suggest that time breaks between individual lava flows represent a few years to decades, time breaks between flow groups represent at least a couple of hundred years or possibly much longer, while significant hiatuses in volcanism, revealed by thick sediment packages or polarity reversals (both are evidenced in this well), are inferred to last thousands to tens of thousands of years. Major element geochemistry from 52 basaltic lava flows demonstrates near primitive compositions (i.e., ~10 wt% MgO) and tholeiitic iron enrichment trends, similar to lavas from the eastern SRP. Trace element concentrations are similar to those of ocean island basalts, with enriched Ba and Pb, and light rare earth element (REE)/heavy REE ratios similar to those of many Neogene volcanics of the western Cordillera. When combined, we identify a total of 11 flow groups, which we also classify as fractionation or recharge on the basis of decreasing or increasing MgO weight percent with depth. Taking into consideration these trends, we review the potential recharge, fractionation, and assimilation processes that characterize much of SRP olivine tholeiite, and conclude that assimilation, in combination with fractional crystallization, is the dominant petrogenesis for the basalts in the central SRP. Although fractionation of Wendell parent magmas was accompanied by assimilation of crustal material, this could not have been assimilation of ancient cratonic crust. The geochemical cycles observed in this well are inferred to represent fractionation and recharge of basaltic magma from a series of sill-like layered mafic intrusions located in the middle crust, similar to what has been proposed for the processes that control the eruptive history of basalts in the eastern SRP.

    Recommended Patient-Reported Core Set of Symptoms to Measure in Prostate Cancer Treatment Trials (2014)

    Description: The National Cancer Institute (NCI) Symptom Management and Health-Related Quality of Life Steering Committee convened four working groups to recommend core sets of patient-reported outcomes to be routinely incorporated in clinical trials. The Prostate Cancer Working Group included physicians, researchers, and a patient advocate. The group’s process included 1) a systematic literature review to determine the prevalence and severity of symptoms, 2) a multistakeholder meeting sponsored by the NCI to review the evidence and build consensus, and 3) a postmeeting expert panel synthesis of findings to finalize recommendations. Five domains were recommended for localized prostate cancer: urinary incontinence, urinary obstruction and irritation, bowel-related symptoms, sexual dysfunction, and hormonal symptoms. Four domains were recommended for advanced prostate cancer: pain, fatigue, mental well-being, and physical well-being. Additional domains for consideration include decisional regret, satisfaction with care, and anxiety related to prostate cancer. These recommendations have been endorsed by the NCI for implementation.

    Staging for distant metastases in operable breast cancer: a suggested expansion of the ESMO guideline recommendation for staging imaging of node-negative, hormonal receptor-negative disease (2013)

    Description: We evaluated the impact of staging procedures to detect asymptomatic distant metastases (DM) in the management of women with operable invasive breast cancer (BC, entire cohort: n = 866). Out of 472 patients with lymph node (LN)-negative disease (pN0), DM were found in four cases (detection rate: 0.8%). All four patients presented with established risk factors: hormone receptor (HR)-negative status, HER2-positive status, n = 3; ‘triple-negative’ disease, n = 1. Considering the subgroup of LN-negative patients whose tumors showed the risk factor ‘negative HR status’ ( n = 66), the detection rate of DM was 6%. The detection rates of DM in higher pN categories were as follows: pN1:1.7%; pN2:9.5%; pN3:13.5%. We generally support the international guidelines, including those published by the European Society for Medical Oncology (ESMO) which emphasize that patients with early-stage BC do not profit from radiological staging for the detection of DM and recommend refraining from this. However, we would expand these guidelines and propose that screening should be carried out in node-negative patients whose tumors show established tumor-related risk factors (e.g. HR-negative and HER2-positive status), since in this particular subcohort, the detection rate of DM is with 6% similarly high as that of patients with four to nine positive LNs.

    Mechanism of Tc toxin action revealed in molecular detail (2014)

    Description: Tripartite Tc toxin complexes of bacterial pathogens perforate the host membrane and translocate toxic enzymes into the host cell, including in humans. The underlying mechanism is complex but poorly understood. Here we report the first, to our knowledge, high-resolution structures of a TcA subunit in its prepore and pore state and of a complete 1.7 megadalton Tc complex. The structures reveal that, in addition to a translocation channel, TcA forms four receptor-binding sites and a neuraminidase-like region, which are important for its host specificity. pH-induced opening of the shell releases an entropic spring that drives the injection of the TcA channel into the membrane. Binding of TcB/TcC to TcA opens a gate formed by a six-bladed beta-propeller and results in a continuous protein translocation channel, whose architecture and properties suggest a novel mode of protein unfolding and translocation.     Regulation of epithelial plasticity by miR-424 and miR-200 in a new prostate cancer metastasis model (2013)

    Description: Using an in vivo cycling strategy, we selected metastatic cancer cells from the lymph nodes (LN) of mice bearing orthotopic DU145 human prostate tumors. Repeated rounds of metastatic selection (LN1–LN4) progressively increased the epithelial phenotype, resulting in a new model of tumor cell mesenchymal-epithelial transition (MET). DU145-LN4 showed increased cell-cell adhesions, higher expression of multiple epithelial markers, such as E-cadherin, EpCAM and cytokeratin 18, and reduced expression of mesenchymal markers such as vimentin. The MET in DU145-LN4 cells was accompanied by increased expression of the miR-200 family, and antimiRs to miR-200c and miR-141 induced an EMT. MET also correlated with the loss of miR-424. Ectopic transient and stable miR-424 expression induced EMT, with reduced epithelial marker expression and increased cell scattering. Our model provides evidence for spontaneous MET in vivo. We show that this cellular plasticity can be mediated through the combined action of miR-424 and the miR-200 family. Scientific Reports 3 doi: 10.1038/srep03151
