    Description: Temora longicornis, a dominant calanoid copepod species in the North Sea, is characterised by low lipid reserves and high biomass turnover rates. To survive and reproduce successfully, this species needs continuous food supply and thus requires a highly flexible digestive system to exploit various food sources. Information on the capacity of digestive enzymes is scarce and therefore the aim of our study was to investigate the enzymatic capability to respond to quickly changing nutritional conditions. We conducted two feeding experiments with female T. longicornis from the southern North Sea off Helgoland. In the first experiment in 2005, we tested how digestive enzyme activities and enzyme patterns as revealed by substrate SDS-PAGE (sodium dodecylsulfate polyacrylamide gel electrophoresis) responded to changes in food composition. Females were incubated for three days fed ad libitum with either the heterotrophic dinoflagellate Oxyrrhis marina or the diatom Thalassiosira weissflogii. At the beginning and at the end of the experiment, copepods were deep-frozen for analyses. The lipolytic enzyme activity did not change over the course of the experiment but the enzyme patterns did, indicating a distinct diet-induced response. In a second experiment in 2008, we therefore focused on the enzyme patterns, testing how fast changes occur and whether feeding on the same algal species leads to similar patterns. In this experiment, we kept the females for 4 days at surplus food while changing the algal food species daily. At day 1, copepods were offered O. marina. On day 2, females received the cryptophycean Rhodomonas baltica followed by T. weissflogii on day 3. On day 4 copepods were again fed with O. marina. Each day, copepods were frozen for analysis by means of substrate SDS-PAGE. This showed that within 24 h new digestive enzymes appeared on the electrophoresis gels while others disappeared with the introduction of a new food species, and that the patterns were similar on day 1 and 4, when females were fed with O. marina. In addition, we monitored the fatty acid compositions of the copepods, and this indicated that specific algal fatty acids were quickly incorporated. With such short time lags between substrate availability and enzyme response, T. longicornis can successfully exploit short-term food sources and is thus well adapted to changes in food availability, as they often occur in its natural environment due seasonal variations in phyto- and microzooplankton distribution.

    Description: Studies of phytoplankton ecology and biogeochemical parameters have been carried out with the ice breaking vessel RV Polarstern since the nineties at various locations in the central Artcic Ocean, the Greenland Sea and the Fram Strait, however, plankton abundance and composition were determined sporadically, and only few biogeochemical components were analysed. Since rapid environmental changes due to increasing temperatures, sea ice loss and ocean acidification in the Arctic Ocean are expected, a more comprehensive impression of the impact of the anticipated changes on pelagic biological processes and the consequences for organic matter cycling is desirable. To get more detailed investigations on the pelagic system the new research group PEBCAO was created. The aim of this group is to complement the measurements of bulk variables and samples on phyto- and protozooplankton abundances by a molecular assessment of the phytoplankton diversity, including the pico- and nanoplankton allowing to better quantifying the intrusion of invading species into the polar habitat. The point measurements during cruises will serve as ground-truthing data to create basin wide satellite images focussing on the quantitative estimation of various phytoplankton functional types, which can serve as an input for modelling approaches. Furthermore, investigations on changes in the composition of organic matter (OM) including molecular analysis of OM are carried out and together with abundance and activity of key species in zooplankton will improve the export estimates under climate change. One local focus of this group is the deep-sea long-term observatory HAUSGARTEN of AWI in the Fram Strait off Svalbard, where investigations on plankton ecology and particle flux have been carried out since the 1990. These observations can be used to identify how current observed changes are related in a historical context. Here we present first results of the multidisciplinary approach form the long term observations and the studies carried out during two Polarstern cruises (ARK 24_1&2 and ARK 25_1&2) in the summer of 2009 & 2010, respectively.

    Description: In calanoid copepods it is poorly understood how enzymatic activities and patterns are affected by abiotic and biotic factors. Such knowledge, however, is crucial to assess metabolic functioning and performance of organisms in different habitats. Therefore, our study focuses on digestive enzyme activities in relation to temperature,pH and food in the Arctic species Calanus glacialis and in Centropages hamatus and Temora longicornis from the North Sea. Enzyme activities were measured over a range from 0 to 70 °C (lipases/esterases, proteinases)and pH 5 to 9 (proteinases). In all species, relative proteinases activity peaked at 40/50 °C and pH 6; relative lipases/esterases activity peaked at 30 °C. Between 0 and 20 °C, lipase activity of C. glacialis was higher (40–70% of maximum) than that of the boreal copepods (25–64%), which suggests thermal adaptation of the lipid metabolismin the polar species. Incubating C. glacialis with the diatom Thalassiosira weissflogii showed (i) that enzyme activities increased especially in the alkaline range and (ii) that enzyme patterns, revealed by gel electrophoresis, differed from that of starving individuals, indicating that feeding induced enzyme expression. Such studies, linking abiotic and biotic conditions to enzyme functioning, can help elucidating the capacity of copepods to respond to environmental changes.
