    Abstract: In this paper, global climatic response to the North Pacific oceanic warming is investigated in a series of coupled ocean–atmosphere modeling experiments. In the model, an idealized heating is imposed over the North Pacific Ocean, while the ocean and atmosphere remain fully coupled both locally and elsewhere. The model explicitly demonstrates that the North Pacific oceanic warming can force a significant change of the atmospheric circulation with a strong seasonal dependence. The seasonal marching of the atmospheric response over the North Pacific is characterized by a quasi-baratropic warm ridge in early winter, a transition to a quasi-baratropic warm trough in late winter, and then to a baroclinic response in summer with a trough and ridge, respectively, in the lower and upper troposphere. The North Pacific warming also forces a significant remote response over the tropical Pacific. In winter, the tropical Pacific response is characterized by a nearly uniform warming coupled with anomalous southerly cross-equatorial winds, while in summer it is dominated by an enhanced zonal SST gradient and anomalous equatorial easterlies. The tropical warming tends to be associated with a reduction of the upper-ocean meridional overturning circulation and equatorial ocean dynamics associated with a reduction of the Hadley circulation and the surface coupled wind–evaporation–SST feedback. The resulting tropical warming can further intensify the seasonal marching of the North Pacific atmospheric response. The global impacts of the North Pacific warming are also discussed.

    Abstract: In this paper, the atmospheric teleconnections of the tropical Atlantic SST variability are investigated in a series of coupled ocean–atmosphere modeling experiments. It is found that the tropical Atlantic climate not only displays an apparent interhemispheric link, but also significantly influences the North Atlantic Oscillation (NAO) and the El Niño–Southern Oscillation (ENSO). In spring, the tropical Atlantic SST exhibits an interhemispheric seesaw controlled by the wind–evaporation–SST (WES) feedback that subsequently decays through the mediation of the seasonal migration of the ITCZ. Over the North Atlantic, the tropical Atlantic SST can force a significant coupled NAO–dipole SST response in spring that changes to a coupled wave train–horseshoe SST response in the following summer and fall, and a recurrence of the NAO in the next winter. The seasonal changes of the atmospheric response as well as the recurrence of the next winter’s NAO are driven predominantly by the tropical Atlantic SST itself, while the resulting extratropical SST can enhance the atmospheric response, but it is not a necessary bridge of the winter-to-winter NAO persistency. Over the Pacific, the model demonstrates that the north tropical Atlantic (NTA) SST can also organize an interhemispheric SST seesaw in spring in the eastern equatorial Pacific that subsequently evolves into an ENSO-like pattern in the tropical Pacific through mediation of the ITCZ and equatorial coupled ocean–atmosphere feedback.

    Abstract: Hepatocellular carcinoma (HCC) is the most invasive and frequently diagnosed malignancy and the second leading cause of cancer death in many regions of Asia. The PI3K/Akt/mTOR signal pathway is involved in multiple cellular functions including proliferation, differentiation, tumorigenesis, and apoptosis. Up-regulation of telomerase activity is thought to be a critical step leading to cell transformation. Methods This study investigated changes in mTOR pathway and telomerase activity in hepatocarcinoma cell line SMMC-7721 treated with chemotherapeutic agent 5-fluorouracil (5-Fu). We detected apoptosis of hepatocarcinoma cells by TUNEL assay. Telomerase activity, hTERT transcription level and p- p70 S6k was demonstrated by the telomeric repeat amplification protocol and silver staining assay, Dual-Luciferase Reporter Assay and Western blot analysis respectively. Results Treating SMMC-7721 cells with 5-Fu leads to apoptosis of the cells, and reduction in telomerase activity, as well as a dramatic reduction in the activated form of p70 S6 kinase, a mTOR substrate. The 5-Fu treatment nearly abolishes transcription of hTERT (the major component of telomerase) mRNA. Treating SMMC-7721 cells with Rapamycin, a specific mTOR inhibitor, significantly reduce hTERT protein level but did not affect hTERT transcription. 5-Fu and rapamycin were synergistic in regards to down-regulation of telomerase activity in hepatocarcinoma cells. Conclusion These results suggest that chemotherapeutic agent 5-Fu may down-regulate telomerase activity at both transcriptional level and PI3K/Akt/mTOR pathway-dependent post-transcriptional level to facilitate hepatocellular carcinoma cell apoptosis.

    Abstract: In its dormant state, the membrane cytoskeletal linker protein ezrin takes on a NH 2 terminal-to-COOH terminal (N-C) binding conformation. In vitro evidence suggests that eliminating the N-C binding conformation by Thr 567 phosphorylation leads to ezrin activation. Here, we found for resting gastric parietal cells that the levels of ezrin phosphorylation on Thr 567 are low and can be increased to a small extent (∼40%) by stimulating secretion via the cAMP pathway. Treatment of cells with protein phosphatase inhibitors led to a rapid, dramatic increase in Thr 567 phosphorylation by 400% over resting levels, prompting the hypothesis that ezrin activity is regulated by turnover of phosphorylation on Thr 567 . In vitro and in vivo fluorescence resonance energy transfer analysis demonstrated that Thr 567 phosphorylation opens the N-C interaction. However, even in the closed conformation, ezrin localizes to membranes by an exposed NH 2 terminal binding site. Importantly, the opened phosphorylated form of ezrin more readily cosediments with F-actin and binds more tightly to membrane than the closed forms. Furthermore, fluorescence recovery after photobleaching analysis in live cells showed that the Thr567Asp mutant had longer recovery times than the wild type or the Thr567Ala mutant, indicating the Thr 567 -phosphorylated form of ezrin is tightly associated with F-actin and the membrane, restricting normal activity. These data demonstrate and emphasize the functional importance of reversible phosphorylation of ezrin on F-actin binding. A novel model is proposed whereby ezrin and closely associated kinase and phosphatase proteins represent a motor complex to maintain a dynamic relationship between the varying membrane surface area and filamentous actin length.
