    Notes: Abstract The biological community that surrounds the hypersaline cold water brine seeps at the base of the Florida Escarpment is dominated by two macrofaunal species: an undescribed bivalve of the family Mytilidac and a vestimentiferan worm, Escarpia laminata. These animals are apparently supported by the chemoautotrophic fixation of carbon via bacterial endosymbionts. Water column and sediment data indicate that high levels of both sulfide and methane are present in surface sediments around the animals but absent from overlying waters. Stable isotopic analyses of pore water indicate that there are two sources of sulfide: the first is geothermal sulfide carried in groundwater leaching from the base of the escarpment, and the second is microbial sulfide produced in situ. The vestimentiferan E. laminata, and the mytilid bivalve (seep mussel) live contiguously but rely on different substrates for chemoautotrophy. Enzyme assays, patterns of elemental sulfur storage and stable isotopic analyses indicate that E. laminata relies on sulfide oxidation and the seep mussel on methane oxidation for growth.

    Notes: Abstract Premethylation of purified porcine cardiac sarcolemma (SL) in the presence of 0.15, 10 and 150 µM S-adenosyl-L-methionine (AdoMet) did not change the phosphorylation of SL proteins catalyzed either by intrinsic cyclic AMP-dependent protein kinase (cAK) or by added catalytic (C) subunit of this enzyme. On the other hand, membrane exhibited increased lipid methyltransferase activity after preincubation with MgATP and C subunit. Prephosphorylation of membranes stimulated the total [3H]-methyl incorporation into SL lipids assayed at 0.15 µM [3H]AdoMet due to an enhancement of Vmax and without changes in the Km value for AdoMet. Analysis of the methylated lipid products revealed an increased methyl group incorporation into a nonpolar lipid fraction whereas phosphatidylethanolamine-N-methylation was not affected by phosphorylation. The results suggest that the cyclic AMP-mediated signal transduction at the level of cardiac SL is not affected by methylation-induced modifications of the membrane lipid microdomains. On the other hand, an intrinsic SL lipid methyltransferase activity is apparently not related to the N-methylation of phospholipids, is modulated by cyclic AMP-dependent protein phosphorylation.

    Notes: Abstract Cu 3d and O 2p electronic states of Cu2O, CuO, and the highT c compound YBa2Cu3O7−δ have been probed by means of high resolution x-ray emission spectroscopy (XES). The CuLα and OKα XES bands are compared in detail with recently reported x-ray photoelectron and ultraviolet photoelectron spectroscopy (XPS and UPS) measurements and densities of states obtained by local density functional (LDF) theory. The XES data show that the hybridization between Cu 3d and O 2p states is completely modified in CuO and YBa2Cu3O7−δ, whered-d correlation energy is large, as compared to LDF predictions. Such is not the case for Cu2O where agreement between theory and experiment is good. The Cu 3d states are found to be highly localized in YBa2Cu3O7−δ (though less so than in CuO). The O 2p states lie at lower binding energies than in the simpler oxides and are mainly situated above the Cu 3d states. The respective positions of the centre of gravity of the OK α emission bands on an x-ray energy scale indicate that the oxygen sites are less well screened by the O 2p states in the highT c compound. This provides indirect evidence for the presence ofd-like states at the oxygen sites.

    Notes: Abstract Several noninvasive measurement methods are used for evaluation of metabolic disease. Single-photon (125I) scans of the peripheral skeleton are useful in some diseases but are ineffective in osteoporosis (even on the distal radius or os calcis) because they cannot predict spinal or femoral density. Also, peripheral measurements show high percentages of false negatives, that is many patients with fractures have normal peripheral density. Dual-photon (153Gd) scans of the spine, femur, and total skeleton are precise and accurate (2% error) and provide direct measurements of bone strength at fracture sites. This gives the best discrimination of abnormality and the most sensitive monitoring. Quantitative computed computed tomography (QCT) allows measurement of the spine but not the critical proximal femur area. QCT has a large accuracy error because (a) the limited are measured (under 5 cm3) fails to represent the total vertebral body, (b) technical errors and (c) variable fat and osteoid influence the results.

    Notes: Accuracy of dosage. Confusion sometimes surrounds the concept of dosage accuracy. For this reason, standard definitions and testing and calculation procedures are proposed for dosage accuracy, dosage error, and dosage constancy. Representation of the relationship between dosage error and dosage constancy within the area of tolerance of the dosage accuracy is new. Since the measurement of all error quantities is always bound to relate terms such as short-term, long-term, and momentary dosage error to a stated time base. This also facilitates a conceptually smooth transition to operating dosage errors observed in long-term operation. It would be convenient if some standardization of the concepts, and possibly a set of rules, could be developed on this basis. Evaluation of measured results on comparing guarantees would then be conducted in relation to the accuracy of measurement.
