    Notes: Summary The anticancer alkaloid vindesine (VDS) was conjugated to four mouse monoclonal antibodies recognizing human tumor-associated antigens. The antibodies were 96.5 (antimelanoma, IgG2a); 791T/36 (antiosteogenic sarcoma, IgG2b); 11.285.14, and 14.95.55 (anticarcinoembryonic antigen, IgG1 and IgG2a respectively). Conjugates VDS-96.5 and VDS-791T/36 were tested in vitro and shown to be specifically cytotoxic for target cells expressing the appropriate antigen. The in vivo effects of the antibodies and conjugates were tested against human tumor xenografts in athymic or immunodeprived mice using multiple treatments. Conjugate VDS-96.5 retarded the initial growth of a melanoma xenograft, whereas free antibody was without effect. Similarly, VDS-791T/36 but not free antibody retarded the growth of osteogenic sarcoma 791T. The most marked antitumor effects observed were those obtained with VDS conjugates of the anti-CEA antibodies against a colorectal tumor xenograft. Antibody 14.95.55 suppressed tumor growth both alone and as a VDS conjugate, whereas 11.285.14 produced only a slight effect alone but an almost complete and lasting suppression of tumor growth as a VDS conjugate. Free VDS had little effect at nontoxic levels. Acute studies showed that VDS-11.285.14 conjugate was considerably less toxic than free VDS in Balb/c mice.

    Notes: Summary The distribution of cefuroxime (250 mg) was studied in patients with renal failure undergoing continuous ambulatory peritoneal dialysis (CAPD). 10 uninfected patients received the drug intravenously and intraperitoneally, while 9 patients with peritonitis were randomly allocated to intravenous or intraperitoneal administration. Samples were taken over the first 6 hour dialysis period. In the infected patients, more drug (p〈0.01) crossed into the peritoneal cavity following intravenous injection and reached the systemic circulation following intraperitoneal administration than in the uninfected group. This increased permeability of the peritoneal membrane during infection may result in unexpected systemic toxicity in patients treated with intraperitoneal antibiotics.

    Notes: Abstract (−)-Epicatechin has previously been suggested to rapidly reverse alloxan diabetes in rats. We have assessed the therapeutic value of the compound in two further animal models of insulin-dependent diabetes mellitus, namely streptozotocin - diabetic rats and the spontaneously diabetic BB/E rat . There was no indication of a reversal of established diabetes in either the streptozotocin-diabetic or the spontaneously diabetic BB/E rats. Moreover, epicatechin also failed to halt the progression of the disease in prediabetic BB/E rats. Earlier claims of the potential use of epicatechin as an antidiabetic agent must therefore be treated with some caution.

    Notes: Summary Crosses between Fragaria moschata (2n=42) and Potentilla fruticosa (2n=14) produced many seeds; about half of these germinated, but only nine plants were obtained from 554 seedlings. Five plants died without flowering, and four were vigorous but sterile. Four plants had the expected chromosome number (2n=28), one plant had 21 chromosomes, and four were aneuploid with 23, 24, 25 and 27 chromosomes respectively. Response to growth substances giving elongated stem internodes and swollen but not succulent receptacles provided evidence that two of the plant were hybrids. It is suggested that all nine plants originated from a normal fertilisation which was followed in some cases by chromosome elimination at an early stage of embryo development.

    Notes: Summary An aneuploid hybrid (2n=23) of Fragaria moschata (2n=42) and Potentilla fruticosa (2n=14) was backcrossed with pollen of both parents, separately and combined in a pollen mixture. Seven vigorous progeny were obtained. The origin of the exeptional chromosome numbers, 2n=44, 49, 63, 63, 65, 67, 67, is discussed, and it is shown that each of the numbers could be produced by the fertilisation of unreduced and double unreduced gametes of the hybrid by normally reduced gametes of one of the parental species.

    Notes: Abstract The influence of low surface roughness of deserts on the July circulation is examined by employing numerical simulations with a GLAS GCM. Two identical sets of simulations were made with the model starting from the initial state of the atmosphere based on the NMC analysis of observations for June 15, at OOZ for the years 1979 and 1980. The first set, called the Control, had land surface roughness set to 45 cm, everywhere. The second set called the Experiment, had surface roughness set to 0.02 cm for deserts, but 45 cm everywhere else on land. All other prescribed boundary conditions were the same in both runs. A comparative analysis of these simulations showed that the rainfall in the Sahara desert was reduced significantly in both Experiments as compared to the corresponding Controls; the ITCZ (inter-tropical convergence zone) moved southward, to about 14° N, which is close to its observed location at about 10° N. This was primarily caused by the relative moisture divergence from the smoother Sahara. In other deserts, which anyway had little rainfall in the July simulation of the Control run, there was virtually no change. The differences in regional heat and moisture budgets, particularly for the Sahara desert, are significant as compared to the sample standard deviation for a set of three July simulations (i.e., Control runs for three different initial conditions). In a third simulation, in which the surface roughness was changed over all land, similar results were obtained in the Sahara desert region. The study reveals the influence of low surface-roughness of deserts on the July rainfall. For the Sahara desert, this influence is comparable to that of an increase in surface albedo. In nature, formation of deserts leads to reduction of surface roughness as the vegetation perishes and soil erosion ensues. It is inferred that the smoothness of land then causes reduction in rainfall and further promotes desertification.

    Notes: Abstract A search for inclusive muon-neutrino (v µ) and muon-antineutrino $$(\bar v_\mu )$$ oscillations has been performed in the Fermilab narrow band neutrino beam using two detectors running simultanously at two distances from the neutrino source. The data show no evidence for either neutrino or antineutrino oscillations and rule out oscillations ofv µ or $$\bar v_\mu $$ into any other single type of neutrino for 15〈Δm 2〈1,000 eV2/c4 and sin2(2θ)〉0.02–0.40.

    Notes: The precise anatomical relation by which autonomic nerve endings contact gastric epithelial cells to enhance the rate of gastric secretions is not fully understood. The aim of the present study was to clarify this issue by using the technique of serial section reconstruction of areas of the gastric mucosa. The work also explored the possibility of a functional role for a system of smooth muscle strands in the gastric mucosa that emanate from the muscularis mucosa, run in the lamina propria, and are associated in a unique manner with the gastric glands. Electron microscopic serial sections of the gastric mucosa were performed to visualize the entire limiting membrane of gastric epithelial cells to determine any nerve associations (especially varicose endings) with these cells. Evaluation of serial sections of five separate parietal cells showed that their basal membrane did not come in close contact (nearest distance 500 nm) with any nerve axon or varicosity. Moreover, the axons passing in the area of these cells ultimately showed varicose endings associated with smooth muscle cells in the adjacent connective tissue (often separated by only 20 nm), with mast cells or with vascular elements. Additionally, the lateral membrane of these five parietal cells did not contact any endocrine cell in the epithelium, although other parietal cells in the area were adjacent to endocrine cells. Chief cells in the immediate area also did not form any close associations with nerve varicosities. Random analysis of 5,000 additional epithelial cells in these sections showed no close associations to nerve elements with significant accumulations of neurosecretory vesicles (varicosities). Because of the observed existence of innervation to the smooth muscle strands in the area of the gastric glands, serial 1-μm epoxy sections of the gastric mucosa were prepared, and profiles of smooth muscle and gastric glands were entered into a computer-assisted reconstruction system. Three-dimensional reconstruction techniques were employed to reveal the existence of a unique association between the mucosal smooth muscle strands and the gastric glands. The muscle strands arose from the muscularis mucosa at regular intervals and became branched to form an intricate wrap around a series of gastric glands that empty into one gastric pit. Branching of the muscle strands initially occurred at the point where they approached the base of the glands and then emanated into the connective tissue around the glands in a crossing pattern, ending at the base of the gastric pit. Although muscarinic agents have been shown to directly stimulate parietal cells to secrete acid, these findings have led us to postulate that autonomic nerve stimulation may also aid gastric secretion both by stimulation of mast cells and by glandular excretion mediated via mucosal muscular contractions.

    Notes: Compacted discs prepared from a mixture of finely divided particles of a compound diluted in a large excess of metal powder facilitate the measurement of Raman spectra for some compounds which give poor scattering by conventional techniques. The method is most useful with coloured compounds. The advantages include simplicity, increased signal-to-noise ratio by factors of up to 1000 and decreased or altered fluorescence. The effect appears to be caused mainly by small crevices in the compacted metal surface which act as internal mirrors to produce many small multiple-reflection cells, but there is also evidence of an electronic interaction with the metal surface. Silver, aluminium and copper matrices were studied, silver being found to be the most advantageous, although aluminium under certain conditions may also be effective.
