    Notes: The evolution of the latex particle diameter distribution during batch emulsion polymerization is investigated, with emphasis on changes in the breadth of the size distribution. A model utilizing a surface area-dependent volumetric growth rate of a single particle results in a time-invariant standard deviation of the size distribution during periods of particle growth only. This behavior is reconciled with some experimental observations by considering the occurrence of particle nucleation during some part of the growth interval. Conclusions based on the results of the model suggest that higher inhibitor and low emulsifier concentrations favor narrow particle size distributions.

    Notes: Two monoclonal antibodies reactive for α-tubulin but not for β-tubulin have been prepared, characterized in terms of their relative binding to tubulins from differnt sources by a solid-phase binding assay, immunoautoradiography, and indirect immunofluorescence, and utilized to study flagellar motility. Our results demonstrate that α-tubulins from different species, and even from different tissues of the same species, are nonidentical. Especially interesting was the observation that one of the antibodies, Ab2, immunofluorescently stained microtubules of chick embryo fibroblast cells, but was completely unreactive for microtubules of rat kangaroo (PtK2) fibroblasts; a different antibody, Ab1, stained both cell types. Results of these and additional experiments clearly show that Ab1 and Ab2 recognize discrete and different epitopes on α-tubulin.Monoclonal antitubulins Ab1 and Ab2 each inhibited the bend amplitude of reactivated sea urchin spermatozoa without affecting beat frequencies or the ability of the outer doublet microtubules to slide past each other in elastase-digested models. These results, together with those obtained previously using rabbit polyclonal antitubulin antibodies [Asai and Brokaw, 1980], demonstrate that inhibition of bend amplitude is a common property of antitubulin antibodies and is not due to the binding of antibodies to one specific site on the axoneme. Our results suggest that tubulin subunit conformational changes may occur on the outer doublet lattice and may be integrally involved in the mechanism and control of flagellar bending.

    Notes: Treatment of CEM-C7 cells with glucocorticoids produces a 2.5-fold increase in the activity of the enzyme glutamine synthetase (GS). This increase is specific for steroids with glucocorticoid activity and occurs over a range of steroid concentrations consistent with a receptor-mediated mechanism. Half-maximal and maximal inductions by dexamethasone (dex) occur at 2 × 10-8M and 2 × 10-7 M dex, respectively, concentrations approximately equal to those necessary to produce half and full occupancy of glucocorticoid receptors. GS activity began to increase 1 hour after dex treatment and was complete by 12 hours. This is well before any of the growth inhibitory or cytolytic effects of dex on this cell line occur. This increase was dependent on the presence of glucocorticoid receptors and required both RNA and protein synthesis. Removal of dex following stimulation to maximal levels resulted in a decrease of GS activity to preinduced levels with a half-time of 5 hours. Glutamine deprivation of cells resulted in increased GS activity. However, even in the total absence of glutamine, dex treatment elicited a 2.0-2.5-fold increase in GS activity, ruling out inhibition of glutamine uptake as a mechanism for the dex-induced increase. Experiments with 5′-bromodeoxyuridine (BrdU) demonstrated that GS elevation was sensitive to BrdU substitution of DNA, while dex-induced growth inhibition was not. Therefore GS elevation and growth inhibition in this cell line appear to be independently expressed steroid responses.

    Notes: We have studied the uptakes of the glucose analogs, 2-deoxyglucose (2DG), and 3-O-methylglucose (3OMG) in serum-deprived BHK-21 fibroblasts. Incubation for 4 hours with 0.25 mM 1-methyl-3-isobutylxanthine (MIX) increased cellular cyclic AMP 3 to 15 fold, without affecting the initial rates of uptake of these sugars. Incubation with 100 nM insulin, doubled hexose uptake without affecting cyclic AMP. Insulin did not modify the cyclic AMP response to MIX; nor did MIX alter the hexose uptake response to insulin. There was no effect of 0.1 mM L-isoproterenol (ISO) or 0.1 mM prostaglandin E1 (PGE1) on cyclic AMP. PGE1 slightly stimulated uptake of 2DG but not 3OMG; ISO did not affect hexose uptake. We conclude that cyclic AMP does not directly regulate hexose uptake in these cells.

    Notes: A technique for the preparation of self-supporting windows of anodic barrier films on aluminium, which are essential for the precise interpretation of Rutherford backscattering data, is described. The generation of the window comprises several steps which include: (1) electropolishing of the superpure aluminium specimen in a perchloric acid/ethanol mixture; (2) masking a region of surface of the electropolished specimen with ‘Lacomit’ paint; (3) anodizing to develop a barrier-type anodic film over the exposed aluminium surface; (4) removal of the paint to reveal the non-anodized region on one face of the aluminium specimen which is then removed by electropolishing to leave the film window. Following this procedure, self-supporting windows of anodic films, free from the aluminium substrate, having effective diameters up to about 10 mm, are produced. It is shown that the use of the self-supporting window enables the Rutherford backscattering technique to be used to its full potential for the analysis of the barrier-type anodic films, enabling Al and O profiles through the film to be resolved clearly.

    Notes: Improvements of isoelectric focusing (IEF) in agarose are described that result in consistently obtaining broad, linear pH gradients in the range pH 4 to 9.2 at equilibrium. Cathodic drift has been reduced by the use of a blend of commercial carrier ampholytes and by incorporating lysine and arginine in the catholyte as spacers. Resolution has also been enhanced by the use of thin gels, supplemented with sorbitol and glycerol. In addition, a comparison of two agarose preparations for isoelectric focusing was performed using two commercial carrier ampholytes. To minimise background staining, Triton complexes were removed by modification of the fixing and clearing procedures. These modifications to IEF in agarose are being applied to direct tissue isoelectric focusing (DTIF) of normal and diseased human skeletal muscle.

    Notes: This study was designed to measure the degree of correlation between vein formation and the specification of campaniform sensillae positions in the wing of Drosophila melanogaster. The campaniform sensillae are sensory organs placed at various locations on the wing. Those on the third longitudinal vein (L3) were the focus of this analysis. The system of polygenic modifiers of vein length is comparatively simple, as shown in whole chromosome assays of selected lines. This variability provides a sensitive method of altering vein-forming ability and of assessing correlated changes in other parts of the vein pattern. In selection lines of veinlet, sensillae were displaced toward the base of the wing as vein length decreased by distal loss of vein material. Changes in the amount of vein were, however, not directly proportional to changes in sensillae positions. The more distal sensillae were shifted the largest amount. In the mutant tilt, in which reduced L3 vein-forming competence results in subterminal gaps, distal campaniform sensillae were almost completely eliminated. The remaining sensillae were shifted toward the base of the wing where vein formation is normal. The placement of sensillae therefore appears to be sensitive to the same underlying determinants involved in vein-forming competence.
