    Notizen: Abstract A cell suspension culture was established from nodal callus of Cymbopogon martinii (Roxb.) Wats in a liquid medium containing Murashige and Skoog (1962) basal salts, vitamins, 100 mg l–1 myo-inositol and 20 g l–1 of sucrose (MS) that was supplemented with 13.6 µM 2,4-dichlorophenoxyacetic acid and 1.15 µM kinetin. An initial inoculum density of 2 × 104 cells ml–1 exhibited optimum cell growth. Calli were obtained 12–15 days after the suspension was plated onto semisolid medium of a similar composition. When calli were transferred to semisolid regeneration medium containing MS + 6.7 µM N 6-benzyl-adenine + 1.15 µM kinetin, somatic embryogenesis and plantlet regeneration occurred after 10–25 days. There was no significant decrease in the regeneration potential of the calli even when the cultures were initiated from 47-week-old cell suspensions. Chromosome counts of cells in suspensions, calli and somatic embryos derived from cultures of different ages revealed the presence of diploids, tetraploids and octaploids. However, the 33 regenerated plants tested were all diploid, indicating that only diploid cells were capable of regeneration in vitro.

    Notizen: Abstract An efficient and reproducible procedure is described for the large-scale propagation of an epiphytic orchid,Acampe praemorsa (Roxb.) B latter and McCann using foliar explants. Shoot buds were induced in basal parts of foliar explants on Murashige and Skoog medium supplemented with N6-benzyladenine (BA), kinetin (Kn) or thidiazuron (TDZ), the latter being most effective at 1.0 mg/1. Shoots formed to a TDZ-containing medium elongated following transfer to a substrate supplemented with 2.0 mg/l 1-naphthaleneacetic acid (NAA) and 0.5 mg/1 BA. NAA at lower concentrations had no beneficial effects on shoot regeneration, whether added to the medium along with BA, Kn or TDZ. However, it promoted shoot elongation and leaf expansion. Higher concentrations of NAA suppressed shoot regeneration. The frequency of shoot regeneration was greatly influenced by the developmental stage and orientation of the leaf. Shoots regenerated from the foliar explants were rooted successfully on MS medium containing 1.0 mg/l indole-3-butyric acid. The plantlets were acclimated and eventually transferred to a garden.

    Notizen: Abstract The kinetics of formation, acid-catalysed aquation, ligand isomerisation (CoIIIOSO2+→CoIIISO3+), intramolecular electron-transfer, and base-catalysed hydrolysis and isomerisation of O-bonded sulphito complexes, cis-[Co(en)2(B)(OSO2–O)]+[B=benzimidazole (bzimH), N-methylimidazole (N-meim)] and the anation of cis-[Co(en)2(B)OH)]2+ [B=bzimH, N-meim and imH (imidazole)] by oxSO2−3 are reported. Steric acceleration is observed in the formation and acid-catalysed aquation of the O-sulphito complexes. The ligand isomerisation leads to loss of the monodentate amine with the formation of trans-[Co(en)2(SO3–S)2]− (in an excess of sulphite). Steric acceleration is more pronounced in the isomerisation and base hydrolysis than in the redox process. The results indicate cis labilisation of the coordinated O-sulphite. The [(tetraethylenepentamine)Co(OSO2–O)]+ cation undergoes base hydrolysis 103 times faster than the corresponding (en)2(B) complexes; base-catalysed ligand isomerisation for the former is not observed unlike in the latter. The anation of cis-[Co(en)2(B)OH]2+ (B=imH, bzimH, N-meim) by SO2−3 in a mild alkali pH range (pH=7.9–9.6) and in an excess of SO2−3, yields exclusively trans-[Co(en)2(SO3–S)2]− with no evidence for the formation of the cis-[Co(en)2(B)(SO3–S)]+ or its O-sulphito analogue. The intramolecularly generated amido conjugate base of the sulphite ion-pair, {cis-[Co(en)2(B)OH]2+,SO32−}.41cm{cis-[Co(en)(en-H) (B)- OH2]2+,SO32−}, is believed to generate a five-coordinate intermediate (TBP) that captures the S-end of SO2−3 selectively from a site trans-to the amine B so that the amine is labilised by the trans effect of the sulphite. The NH-deprotonated coordinated imidazolate or benzimidazolate species, cis-[Co(en)2(bzm/im)OH]+, do not undergo anation by SO2−3.

    Notizen: Abstract The fermentative production of β-galactosidase from Escherichia coli CSH50 containing the plasmid pOU140 is mechanistically complex and difficult to model in a nonideal bioreactor. A spectral analysis has been done for a fed-batch fermentation with Gaussian disturbances in the feed stream. Inflow noise converts a smooth operation into aperiodic motion, as observed in some chemical reactions also. The disturbances also cause significant differences in the frequency responses of intra-cellular (plasmid DNA and β-galactosidase) and extra-cellular (concentration and mass fraction of recombinant cells) variables, whose implications are discussed.

    Notizen: The design optimization of air-breathing propulsion engine concepts has been accomplished by soft-coupling the NASA Engine Performance Program (NEPP) analyser with the NASA Lewis multidisciplinary optimization tool COMETBOARDS. Engine problems, with their associated design variables and constraints, were cast as non-linear optimization problems with thrust as the merit function. Because of the large number of mission points in the flight envelope, the diversity of constraint types, and the overall distortion of the design space, the most reliable optimization algorithm available in COMETBOARDS, when used by itself, could not produce satisfactory, feasible, optimum solutions. However, COMETBOARDS' unique features-which include a cascade strategy, variable and constraint formulations, and scaling devised especially for difficult multidisciplinary applications-successfully optimized the performance of subsonic and supersonic engine concepts. Even when started from different design points, the combined COMETBOARDS and NEPP results converged to the same global optimum solution. This reliable and robust design tool eliminates manual intervention in the design of air-breathing propulsion engines and eases the cycle analysis procedures. It is also much easier to use than other codes, which is an added benefit. This paper describes COMETBOARDS and its cascade strategy and illustrates the capabilities of the combined design tool through the optimization of a high-bypass-turbofan wave-rotor-topped subsonic engine and a mixed-flow-turbofan supersonic engine. ©1997 John Wiley & Sons, Ltd.

    Notizen: Small-angle x-ray scattering methods were applied to calculate macromolecular parameters like specific inner surface (O/V), length of coherence (lc), and the percentage of air present in agave hybrid, treating it as a densely packed colloidal system belonging to a general micelle system. The parameters were equal to O/V = 8.50 × 10-6 Å-1, lc = 150 Å, and the percentage of air = 0.03. The Kratky camera with latest design was used for the experimental measurement. Because the sample agave ybrid is a natural fiber and a highly oriented system that is celluosic in character, the theories of Kratky and Porod have been used to evaluate these parameters.
