    Notes: Summary Pretreatment of rat vascular smooth muscle cells with cyclosporin A caused concentration- and time-dependent enhancement of both angiotensin II- and platelet-derived growth factor-stimulated cellular functions, which may be related to a rise in vascular tone. In particular, cyclosporin A increased cytosolic free calcium, and augmented the agonist-induced formation of inositol polyphosphate. In addition, it markedly increased the sensitivity of the cells to platelet-derived growth factor-mediated stimulation of thymidine incorporation. The potentiation of vascular smooth muscle cell activity by cyclosporin A may be a novel mechanism by which it exerts an adverse vasoconstrictor effect.

    Notes: Summary Possible circadian changes in the pharmacokinetics and effect on serum angiotensin-converting enzyme (ACE) activity of the ACE inhibitor enalapril have been studied in 8 healthy subjects after oral ingestion of 10 mg enalapril maleate either at 08.00 h or 20.00 h. The time to peak serum concentration (tmax) of enalapril was increased after administration at 20.00 h compared to 08.00 h (2.4 h versus 1.3 h), where as other kinetic parameters were not significantly altered. The 24 h-kinetics of the active metabolite enalaprilat did not differ significantly between the two treatments, but the area under the curve (AUC (0–24)) and the peak serum concentration (Cmax) were slightly higher after intake at 20.00 h. The relationship between the measured serum enalaprilat level and the degree of inhibition of serum ACE was the same after both treatments. Overall, the evening and morning administration of enalapril did not differ markedly in the pharmacokinetics and the time course of ACE inhibition.

    Description / Table of Contents: Summary Plasma aldosterone, plasma renin activity (PRA), plasma cortisol as parameter of ACTH activity and the serum concentrations of sodium and potassium were determined at short time intervals in 10 healthy students after an overnight bedrest and during three hours of ambulation. While PRA rose significantly within 15 minutes of orthostasis in all students, plasma aldosterone showed a similar rapid increase in some of the subjects only. These persons demonstrated also a simultaneous increase of serum potassium or of plasma cortisol. Plasma aldosterone rose not before 30 to 60 minutes after change to the upright position in subjects who showed neigher plasma cortisol nor serum potassium increases. It is concluded that the immediate rise of plasma aldosterone during orthostasis seems to depend on a stimulation by ACTH of by potassium. The main stimulus of plasma aldosterone during orthostasis appears to be the renin angiotensin system. If the aldosterone response to posture is mediated only through this system a delay of 30 to 60 minutes is observed.
    Notes: Zusammenfassung Plasma-Aldosteron, Plasma-Renin-Aktivität (PRA), Plasmacortisol (als Parameter der ACTH-Aktivität) und die Serumkonzentrationen von Natrium und Kalium wurden bei 10 gesunden Studenten nach Bettruhe und in kurzen Zeitabständen während drei Stunden nach dem Aufstehen gemessen. Während die PRA innerhalb von 15 min nach dem Aufstehen bei allen Studenten signifikant anstieg, wurde ein ähnlich schneller Anstieg des Plasmaaldosterons nur bei einigen der Probanden beobachtet. Nur bei diesen wurde ein gleichzeitiger Anstieg des Serumkaliums oder des Plasmacortisols beobachtet. Bei den Studenten, bei denen weder das Plasmacortisol noch das Serumkalium anstiegen, stieg das Plasmaaldosteron frühestens nach 30–60 min nach dem Aufstehen an. Aus den Ergebnissen wird geschlossen, daß der sofortige Anstieg des Plasmaaldosterons nach dem Aufstehen von einer Stimulation durch ACTH oder Kalium abzuhängen scheint. Der Hauptstimulus für das Aldosteron scheint während der Orthostase jedoch das Renin-Angiotensin-System zu sein. Wird Aldosteron nur über dieses System stimuliert, erfolgt der Aldosteronanstieg mit einer Verzögerung von 30–60 min.

    Description / Table of Contents: Zusammenfassung Plasmaaldosteron, Plasmacortisol und die Serumkonzentrationen von Kalium und Natrium wurden bei 5 nierenlosen Patienten vor und in kurzen Zeitabständen bis zu 180 min nach Beendigung der Hämodialyse gemessen. Unter Hämodialyse stieg das Plasmaaldosteron bei 4 der 5 Patienten an, während bei allen Patienten Cortisol, Kalium und Natrium abfielen. Nur bei einem Patienten ließ sich unter Hämodialyse ein Abfall des Aldosterons nachweisen. Nach Beendigung der Hämodialyse kam es über den untersuchten Zeitraum bei 3 der 5 Patienten zu cinem fast kontinuierlichen Abfall des Plasmaaldosterons, während die beiden anderen Patienten Sekretionsepisoden des Aldosterons mit spontanen Anstiegen und anschließendem Abfall der Hormonkonzentration zeigten. Bei allen Patienten stieg nach Hämodialyse das Serumkalium rasch wieder an, während nur geringgradige Veränderungen des Serumnatriums nachweisbar waren. Plasmacortisol zeigte während der Beobachtungsperiode typische tageszeitliche Schwankungen. Wir nehmen an, daß ein noch unbekannter Faktor für den Anstieg des Plasmaaldosterons unter Hämodialyse verantwortlich ist. Einiges spricht dafür, daß über den nach Hämodialyse beobachteten Zeitraum die intracelluläre Kaliumkonzentration und nicht das Serumkalium die Aldosteronsekretion beeinflußt. Dies würde den bei 3 der 5 Patienten scheinbar paradoxen Abfall des Aldosterons bei steigendem Serumkalium erklären.
    Notes: Summary Plasma aldosterone, plasma cortisol and the serum concentrations of sodium and potassium were determined in 5 anephric patients before and at short time intervals up to 180 minutes after hemodialysis. Plasma aldosterone increased in 4 of 5 patients during hemodialysis while in all patients plasma cortisol, sodium and potassium decreased. Only one patient showed a fall in aldosterone during hemodialysis. After hemodialysis plasma aldosterone gradually decreased over a period of 3 hours in 3 of 5 patients, whereas the remaining two patients showed typical secretory episodes of aldosterone. In each patient serum potassium rapidly increased while serum sodium showed only minor variations. Plasma cortisol followed the normal circadian rhythm. We suggest that a still unknown factor had caused the observed increases in plasma aldosterone during hemodialysis. There are reasons to believe that over the period observed after hemodialysis the intracellular potassium concentration and not serum potassium levels has influenced adrenal aldosterone release. This would explain the paradoxical decrease in plasma aldosterone in 3 of the 5 patients while serum potassium increased.

    Description / Table of Contents: Zusammenfassung Arterieller Blutdruck, Plasma-Reninaktivität (PRA) und die Konzentration von Aldosteron (PA) und Cortisol (PC) im Plasma wurden bei 8 männlichen Patienten mit essentieller Hypertension vor und nach β-Receptorenblockade mit Propranolol und Prindolol bestimmt. Die Blutentnahme zur Bestimmung von PRA, PA und PC erfolgten halbstündlich zwischen 22.00 und 6.00 Uhr. Vor der β-Receptorenblockade fand sich ein typisches rhythmisches Verhalten von PRA, PA und PC mit relativ niedrigen Werten zwischen 22.00 und 24.00 Uhr und höheren Werten zwischen 1.00 und 4.00 Uhr. Unter einer Dosis von 3×50 mg Propranolol täglich über 4 Wochen kam es bei allen Patienten zu einer signifikanten Senkung des arteriellen Blutdruckes sowie der Pulsfrequenz. PRA und PA fielen signifikant ab. Der typische Rhythmus von PRA war nicht mehr nachweisbar. Dagegen wiesen PA und PC eine qualitativ unveränderte Rhythmik auf. Prindolol führte in einer Dosis von 3×10 mg zu einer vergleichbaren Blutdrucksenkung. Die mittlere PRA und PA blieben unverändert. Die typische Rhythmik der PRA wurde durch Prindolol ebenfalls aufgehoben, während der PA- und PC-Rhythmus unbeeinflußt blieb. Die hypotensive Wirkung von Prindolol kann nicht über eine Hemmung der PRA erklärt werden. Die Dissoziation des PRA- und des PA-Rhythmus weist darauf hin, daß unter β-Receptorenblockade die zirkadiane Rhythmik der Aldosteronsekretion nicht durch das Renin-angiotensin-System reguliert wird. Die normale Cortisolrhythmik ist mit einem unveränderten ACTH-Rhythmus vereinbar, der für die qualitativ normale zirkadiane Aldosteronsekretion unter β-Receptorenblokkade verantwortlich zu sein scheint.
    Notes: Summary Blood pressure (BP), plasma renin activity (PRA) and plasma concentration of aldosterone (PA) and cortisol (PC) were determined in essential hypertensive patients before and after β-adrenergic blockade with propranolol and prindolol. Serial measurements of PRA, PA and PC at 30 min intervals (8 p.m. to 6 a.m.) were performed in 8 patients. Administration of propranolol (50 mg three times daily) over a period of 4 weeks was followed by a significant reduction in BP, PRA and PA. PC remained unaltered. The pattern of rhythmic secretion of renin was abolished whereas that of aldosterone persisted at a lower level. Prindolol (10 mg three times daily) had a similar effect on BP as propranolol but failed to lower PRA and PA. Rhythmic secretion of renin was also markedly altered with prindolol in the absence of any change in rhythmicity of PA and PC. The hypotensive action of prindolol was not mediated via inhibition of renin release. The dissociation between PRA and PA rhythmicity indicates that during β-adrenergic blokkade diurnal rhythm of aldosterone secretion is not regulated by the renin-angiotensin-system. Rhythmicity of PC was normal, indicating that also ACTH-secretion was unaltered. Thus, under β-blockade unchanged diurnal rhythm of aldosterone may be due to normal ACTH-secretion.

    Notes: Abstract The effect of reagents that modify sulfur-containing amino acid residues in the phosphatidylethanolamine N-methyltransferase was studied in the isolated rat cardiac sarcolemma by employing S-adenosyl-L-[methyl-3H]methionine as a methyl donor. Dithiothreitol protected the sulfhydryl groups in the membrane and caused a concentration- and time-dependent increase of phospholipid N-methylation at three different catalytic sites. This stimulation was highest (9-fold) in the presence of 1 MM MgCl2 and 0.1 µM S-adenosyl-L-[methyl-3H]methionine at pH 8.0 (catalytic site 1), and was associated with an enhancement of Vmax without changes in Km for the methyl donor. Thiol glutathione was less stimulatory than dithiothreitol; glutathione disulfide inhibited the phosphatidylethanolamine N-methylation by 50%. The alkylating reagents, N-ethylmaleimide and methylmethanethiosulfonate, inhibited the N-methylation with IC5O of 6.9 and 14.1 µM, respectively; this inhibition was prevented by 1 mM dithiothreitol. These results indicate a critical role of sulfhydryl groups for the activity of the cardiac sarcolemmal phosphatidylethanolamine N-methyltransferase and suggest that this enzyme system in cardiac sarcolemma may be controlled by the glutathione/glutathione disulfide redox state in the cell.

    Notes: Abstract Three sex pheromone components of the carob moth were isolated and identified from the extract of female pheromone glands, using a variety of techniques including coupled gas chromatographic-electroantennographic recordings, coupled gas chromatographic-mass spectrometric analysis, microozonolysis, electroantennographic assays of monounsaturated standards, wind-tunnel bioassays, and field trials. The major component was identified as (Z,E)-9,11,13-tetradecatrienal, a novel lepidopterous pheromone component structure. Two minor components, either one of which improves the upwind flight response of males when blended with the major component, were identified as (Z,E)-9,11-tetradecadienal, and (Z)-9-tetra-decenal.
